AIM: To determine the aetiolog y of a recurring and severe form of infectious keratoconjunctivitis (IKC) in sheep.

METHODS: Five sheep flocks that had experienced a severe form of IKC were examined. Clinical history, conjunctival swabs and blood samples were collected from affected animals. Culture for bacteria, and also specifically for Mycoplasma and Chlamydophila spp, and detection of Mycoplasma conjunctivae DNA by polymerase chain reaction (PCR) were attempted. Serum samples were tested for antibodies to M. agalactiae, M. capricolum, M. conjunctivae and Chlamydophila spp.

RESULTS: Mycoplasma conjunctivae DNA was detected using PCR in 3/5 flocks, and in all flocks antibodies to M. conjunctivae were detected in sera. A pure growth of Branhamella ovis was cultured from conjunctival swabs from a small proportion of sheep in two flocks. No other pathogens were detected.

CONCLUSIONS: This investigation demonstrated that M. conjunctivae was a primary pathogen causing severe IKC in sheep, and is the first report of detection of this organism in sheep in New Zealand. Introduction of clinically normal carrier sheep appeared to have caused the outbreaks.  相似文献

Chitosan is a natural polycationic linear polysaccharide derived from chitin. The low solubility of chitosan in neutral and alkaline solution limits its application. Nevertheless, chemical modification into composites or hydrogels brings to it new functional properties for different applications. Chitosans are recognized as versatile biomaterials because of their non-toxicity, low allergenicity, biocompatibility and biodegradability. This review presents the recent research, trends and prospects in chitosan. Some special pharmaceutical and biomedical applications are also highlighted.  相似文献

Lipoxygenase (LOX)-inhibiting compounds from the leaves of Chisocheton polyandrus Merr. were isolated in this study using a bioactivity-guided fractionation technique. Two dammarane triterpenoids, dammara-20,24-dien-3-one (1) (IC(50)=0.69±0.07 μM) and 24-hydroxydammara-20,25-dien-3-one (2) (IC(50)=1.11±0.38 μM), were isolated and identified based on the soybean LOX assay. Dammara-20,24-dien-3-one (1) exhibited dual inhibition of both human 5-LOX (IC(50)=24.27±2.92 μM) and cyclooxygenase-2 (COX-2) (IC(50)=3.17±0.90 μM), whereas 24-hydroxydammara-20,25-dien-3-one (2) did not exhibit any significant inhibitory effects. This report is the first to detail the inhibition of LOX and COX by both C. polyandrus and its isolated compounds.  相似文献

This study characterizes the effects of water–soil flooding volume ratio and flooding time on copper (Cu) desorption and toxicity following multiple floodings of field-collected soils from agricultural sites acquired under the Comprehensive Everglades Restoration Plan (CERP) in south Florida. Soils from four field sites were flooded with three water–soil ratios (2, 4, and 6 [water] to 1 [soil]) and held for 14 days to characterize the effects of volume ratio and flooding duration on Cu desorption (volume ratio and flooding duration study). Desorption of Cu was also characterized by flooding soils four times from seven field sites with a volume ratio of 2 (water) to 1 (soil) (multiple flooding study). Acute toxicity tests were also conducted using overlying waters from the first flooding event to characterize the effects of Cu on the survival of fathead minnows (Pimephales promelas), cladocerans (Daphnia magna), amphipods (Hyalella azteca), midges (Chironomus tentans), duckweed (Lemna minor), and Florida apple snails (Pomacea paludosa). Acute tests were also conducted with D. magna exposed to overlying water from the second and third flooding events. Results indicate that dissolved Cu concentrations in overlying water increased with flooding duration and decreased with volume ratio. In the multiple flooding study, initial Cu concentrations in soils ranged from 5 to 223 mg/kg (dw) and were similar to Cu concentration after four flooding events, indicating retention of Cu in soils. Copper desorption was dependent on soil Cu content and soil characteristics. Total Cu concentration in overlying water (Cuw) was a function of dissolved organic carbon (DOC), alkalinity, and soil Cu concentration (Cus): log(Cuw)?=?1.2909?+?0.0279 (DOC)?+?0.0026 (Cus)???0.0038 (alkalinity). The model was validated and highly predictive. Most of the desorbed Cu in the water column complexed with organic matter in the soils and accounted for 99% of the total dissolved Cu. Although total dissolved Cu concentrations in overlying water did not significantly decrease with number of flooding events, concentrations of free Cu2+ increased with the number of flooding events, due to a decrease in DOC concentrations. The fraction of bioavailable Cu species (Cu2+, CuOH+, CuCO3) was also less than 1% of the total Cu. Overlying water from the first flooding event was only acutely toxic to the Florida apple snail from one site. However, overlying water from the third flooding of six out of seven soils was acutely toxic to D. magna. The decrease in DOC concentrations and increase in bioavailable Cu2+ species may explain the changes in acute toxicity to D. magna. Results of this study reveal potential for high Cu bioavailability (Cu2+) and toxicity to aquatic biota overtime in inundated agricultural lands acquired under the CERP.  相似文献

Dry bacterial cellulose nanofiber (BC) sheet coated with poly(lactic acid) (PLA) was developed and characterized towards acute wound healing applications. This new approach of PLA coating on BC revealed enhanced physical and antibacterial properties. Commercial BC sheets originated from the manufacturing of nata de coco jelly were dried and coated with the PLA at various concentrations of 2, 4, 6, 8, 10 and 12 % w/v for the purpose of improving the mechanical properties and followed by loading of antiseptic such as benzalkonium chloride (BAC). PLA has been proposed for the use of coating materials at a concentration of 8 %, the biocomposite sheet started exhibiting a low moisture uptake, prolonged swelling in simulated wound fluid solution and high tear (9.17 Nm2/kg) and burst indices (32.5 kPa·m2/g). The 8 % PLA coating revealed porous fiber-like morphology as observed under scanning electron microscope. Therapeutic loading capacity of the BC/8 PLA was substantially higher than the pristine BC. Furthermore strong antimicrobial activities against Staphylococcus aureaus and Escherichia coli were observed for the BC/8PLA biocomposite film. These reports were clearly suggestive of the fact that synthetic biodegradable polymers, such as PLA, may be exploited for the synergistic combination with BC for antimicrobial and acute wound management. This new and modified fiber source material could reduce the dependency on plant based cellulose for more demanding biomedical applications such as wound healing materials, vascular graft, cartilage replacement, drug delivery and tissue engineering.  相似文献

ABSTRACT The soilborne fungus Cylindrocarpon destructans (teleomorph: Neonectria radicicola) causes root rot in a wide range of plant hosts; the disease is of particular concern in ginseng production, and in conifer and fruit tree nurseries. beta-Tubulin gene and rRNA gene internal transcribed spacer (ITS) sequence data and pathogenicity assays were used to characterize isolates of C. destructans from ginseng and other hosts. The results of these studies demonstrated a high amount of sequence divergence among strains identified as C. destructans or N. radicicola, suggesting the existence of several phylogenetic species in this complex. Accordingly, we propose that the two varieties of N. radicicola be raised to species status. Certain highly aggressive ginseng isolates from Ontario, Korea, and Japan have identical ITS and beta-tubulin sequences, and form a monophyletic clade (designated "clade a"); these strains are identified as C. destructans f. sp. panacis. Other ginseng strains clustered in monophyletic groups with strains from angiosperm and conifers. A subtractive hybridization method was used to isolate genomic DNA sequences with diagnostic potential from the aggressive C. destructans Ontario ginseng isolate 1640. One of these sequences was similar to the rRNA gene intergenic spacer from a Fusarium oxysporum isolate from Pinus ponderosa, and hybridized to DNA from F. oxysporum and all C. destructans isolates tested. Primers were designed that could be used to amplify this sequence specifically from the highly aggressive, ginsengadapted C. destructans isolates from Ontario and Korea and other members of clade a.  相似文献
