It was suggested that the cryodamage to oocytes' DNA has been responsible for the compromised developmental competence of cryopreserved oocytes. Vitrification of bovine oocytes affected not only cellular components, but also nuclear material. A significant rate of DNA fragmentation was found in bovine frozen or vitrified oocytes analysed by Comet assay regardless of cryopreservation method. Our method of vitrification using droplet system after gentle pre-equilibration treatment is one of the most effective cryopreservation methods employed for bovine oocytes so far, making it possible to develop 30% blastocyst stage embryos. In this study, the extent of DNA damage in bovine oocytes vitrified using three vitrification methods (droplet system, Open Pulled Straw and traditional vitrification in 0.25 ml insemination straws) was compared using Comet assay. Vitrification in droplet system and Open Pull Straws vitrification did not result in detectable cryoinjuries of DNA of bovine oocytes. On the contrary, DNA fragmentation was found in four of 26 oocytes vitrified in 0.25 ml straws (15.4%, p   ≤ 0.05 in comparison with the other vitrification methods).  相似文献

Toxigenic strains of Pasteurella multocida were readily differentiated from non-toxigenic strains by an agarose overlay method using bovine turbinate cells or bovine lung cells. Cells which were young and densely confluent were best suited to this assay. The incubation period required to distinguish toxigenic strains was dependent on the confluence of the monolayers, which was affected by the seeding rate, cell passage level and growth time prior to overlay. The agarose overlay method correctly identified 11 of 11 reference strains of Pasteurella multocida, and visible cytotoxic changes were present in the monolayers after 48 to 65 h. Outbreaks of the enzootic form of atrophic rhinitis in 2 New South Wales piggeries were associated with the isolation of toxigenic type D strains of P. multocida.  相似文献

Objectives of this study were to evaluate maternal and fetal influences on uterine and umbilical blood flows and nutrient fluxes of gravid uterine tissues of cows. Brahman cows with Brahman or Charolais fetuses and Charolais cows with Brahman or Charolais fetuses were used. Indwelling catheters were placed into a uterine artery and vein, an umbilical vein, and a fetal femoral artery and vein at 220 +/- .4 d after mating. Uterine and umbilical blood flows (liters/min) and net uptakes of oxygen, glucose, lactate, alpha-amino N, urea N, ammonia N, and estrone sulfate by the gravid uterus, fetus, and uteroplacenta were determined on d 227 +/- .4. Uterine blood flows in Brahman cows with Brahman (5.01) or Charolais (4.66) fetuses were similar but less (P less than .001) than in Charolais cows with Brahman (7.14) or Charolais fetuses (9.24), which differed (P less than .01). Umbilical blood flows of Charolais (3.78) were greater (P less than .01) than those of Brahman (2.29) fetuses. Rate of placental D2O clearance as well as net fetal uptake of oxygen, glucose, and alpha-amino N, gravid uterine uptake of alpha-amino N, and uteroplacental uptake of glucose and release of estrone sulfate were greater with Charolais than with Brahman fetuses. Gravid uterine oxygen uptake and estrone sulfate release and gravid uterine and uteroplacental lactate output were influenced by the interaction between cow and fetal breed. It is suggested that fetal growth may be limited by uterine blood flow and by function of the uteroplacenta, particularly in late gestation.  相似文献

Our objective was to determine the impact of supplemental energy, N, and protein on feed intake and N metabolism in sheep fed low-quality forage. Six Texel x Dorset wethers (16 mo, 63+/-3.1 kg) fitted with mesenteric, portal, and hepatic venous catheters were used in a Latin square design with five sampling periods. Lambs were fed chopped bromegrass hay (4.3% CP) to appetite, and a mineral mixture was given. Treatments were 1) control (no supplement), 2) energy (cornstarch, molasses, and soybean oil), 3) energy plus urea, 4) energy plus soybean meal (SBM), and 5) energy plus ruminally undegraded protein (RUP; 50:50 mixture of blood and feather meals). Supplements were fed once daily (.3% BW). Forage DMI did not differ (P = .13), but intake of total DM, N, and energy differed (P<.01) among treatments. Apparent digestibilities of DM, OM, and energy were less (P<.01) for control than for other treatments. Apparent N digestibility was least for control and energy and greatest for urea treatments (P<.05). As a result, digested DM, OM, and energy ranked from least to greatest were control, energy, urea, SBM, and RUP, respectively. Apparently digested N was 2.44, 2.24, 11.39, 9.80, and 11.25 g/d for control, energy, urea, SBM, and RUP (P<.01; SE = .10). Hour of sampling x treatment was a significant source of variation for blood concentrations of ammonia N and urea N, net ammonia N release from portal-drained viscera (PDV) and liver, and urea N release from splanchnic tissues. These results were primarily because patterns through time for the urea treatment differed from the other treatments. Net PDV release of alpha-amino N did not differ (P>.05) between control and energy treatments. Values for those treatments were about one-half of values for urea, SBM, and RUP treatments, which did not differ (P>.05). Hepatic net uptake (negative release) of alpha-amino N for control was 53% of values for the other treatments, which did not differ (P>.05). Net release of alpha-amino N from splanchnic tissues did not differ among treatments (P = .34) and did not differ from zero. The data indicate that arterial alpha-amino N concentration, hepatic alpha-amino N uptake, PDV release and hepatic uptake of ammonia N, and hepatic release of urea N were greater in energy than in control treatments. We also found that hepatic uptake of alpha-amino N was 187% of PDV release in energy-supplemented lambs. These results suggest that energy supplementation of a protein-limiting diet stimulated mobilization of body protein.  相似文献

Twenty-five wether lambs (34 +/- 0.9 kg) fitted with ruminal and abomasal infusion catheters were used in a completely randomized design to determine the effects of differing proportions of ruminal and abomasal casein infusion on N balance in lambs fed a high-concentrate diet (85% corn grain, 1.6% N; DM basis) for ad libitum intake. Wethers were infused with 0 (control) or 10.4 g/d of N from casein with ruminal:abomasal infusion ratios of 100:0, 67:33, 33:67, or 0:100% over a 14-d period. Feed, orts, feces, and urine were collected over the last 5 d. Total N intake and excretion were greater (P < 0.01) in lambs infused with casein than in controls; however, N retention did not differ in lambs infused with casein compared with controls, suggesting that N requirements were met without casein supplementation. Total N intake and total N excretion did not differ among casein infusion treatments. Urinary N excretion decreased linearly (P = 0.07) with decreasing ruminal infusion of casein. Site of casein infusion quadratically (P = 0.06) influenced N retained (g/d), with the greatest retention observed in the 33:67 ruminal:abomasal infusion treatment. Dry matter intake from feed decreased from 1,183 to 945 g/d (P = 0.02) in lambs infused with casein compared with controls, but apparently digested DM did not differ among treatments. These data indicate that decreasing the ruminal degradability of supplemental protein above that required to maximize N retention results in decreased urinary excretion of N without greatly affecting apparent diet digestion.  相似文献

Simulations of carbon storage suggest that conversion of old-growth forests to young fast-growing forests will not decrease atmospheric carbon dioxide (CO(2)) in general, as has been suggested recently. During simulated timber harvest, on-site carbon storage is reduced considerably and does not approach old-growth storage capacity for at least 200 years. Even when sequestration of carbon in wooden buildings is included in the models, timber harvest results in a net flux of CO(2) to the atmosphere. To offset this effect, the production of lumber and other long-term wood products, as well as the life-span of buildings, would have to increase markedly. Mass balance calculations indicate that the conversion of 5 x 10(9) to 1.8 x 10(9) megagrams of carbon to the atmosphere.  相似文献

[目的]定位监测高产膜下滴灌制种玉米田间土壤水分动态变化,为膜下滴灌制种玉米拔节后水分管理提供理论依据.[方法]采用时域反射仪(TDR)对膜下滴灌制种玉米田间土壤水分动态变化连续监测并分析干物质的积累情况,运用水分平衡法计算膜下滴灌制种玉米各生育时期耗水量和水分生产率.[结果]拔节期至灌浆期45d左右是玉米耗水高峰期,日平均耗水强度为6.4 mm;灌浆期日耗水强度为3.2 mm.[结论]制种玉米拔节后根区的土壤体积含水量总是维持在23.5;~31.4;(田间持水量的60;~80;).  相似文献

Global Oscillation Network Group data reveal that the internal structure of the sun can be well represented by a calibrated standard model. However, immediately beneath the convection zone and at the edge of the energy-generating core, the sound-speed variation is somewhat smoother in the sun than it is in the model. This could be a consequence of chemical inhomogeneity that is too severe in the model, perhaps owing to inaccurate modeling of gravitational settling or to neglected macroscopic motion that may be present in the sun. Accurate knowledge of the sun's structure enables inferences to be made about the physics that controls the sun; for example, through the opacity, the equation of state, or wave motion. Those inferences can then be used elsewhere in astrophysics.  相似文献
