The aims of this study were to evaluate the immunomodulatory role of TGF-β1, IL-10, and INF-γ in spleen and liver extracts and supernatant cultures of white spleen cells from male symptomatic and asymptomatic dogs, naturally infected by Leishmania (Leishmania) chagasi. Thirty dogs from Araçatuba, São Paulo, Brazil, an endemic leishmaniosis area, were selected by positive ELISA serological reaction for Leishmania sp. and divided into two groups: asymptomatic (n = 15) and symptomatic (n = 15) consisting of animals with at least three characteristic signs (fever, dermatitis, lymphoadenopathy, onychogryphosis, weight loss, cachexia, locomotion problems, conjunctivitis, epistaxis, hepatosplenomegaly, edema, and apathy). After euthanasia, spleen and liver fragments were collected for ex vivo quantification of TGF-β1, IL-10, and INF-γ. Naturally active in vitro produced TGF-β1 was also evaluated in spleen cell culture supernatant. Spleen and liver extract of asymptomatic dogs had higher mean TGF-β1 levels than symptomatic dogs. High concentrations of IL-10 were found in spleen, and mainly in liver extract of both groups. Higher INF-γ concentrations were found in spleen extracts of symptomatic dogs, and in liver extracts of asymptomatic dogs. Extract of this cytokine was lower in spleen extract. Although INF-γ is being produced in canine infection, mean levels of TGF-β1 and IL-10 from spleen and liver extracts were quantitatively much higher; suggesting that immune response in both asymptomatic and symptomatic dogs was predominantly type Th2. 相似文献

Blood and bone marrow samples were taken from 112 Didelphis spp., collected between March 2005 and February 2006, from urban and peri-urban areas of Bauru, São Paulo State, Brazil, to evaluate the hypothesis that these animals might constitute a reservoir of Leishmania spp. Anti-Leishmania ssp. antibodies were screened in the serum samples using an enzyme-linked immuno-sorbent assay (ELISA) and the polymerase chain reaction (PCR). PCR was performed on fragments of DNA samples from Leishmania spp. using primers 13A and 13B, and showed a positive outcome in 91.6% of the 112 samples tested. Of the 107 samples analyzed by ELISA, 71% were positive. Evidence of epidemiological risk factors such as a circulating parasite and freely moving vectors suggests that Didelphis spp. may participate in the transmission cycle of Leishmania spp. in Bauru. 相似文献

In a WTO battle and the press the argument is often made that eliminating US cotton subsidies would have a large effect on the incomes and competitive position of farmers in developing countries. In Francophone West Africa cotton productivity has stagnated after rapid gains in the first two decades following independence (1960-1980). A farm model was constructed based on farmers’ definition of their decision-making framework which they use to respond to income and weather risks. With this model the effects on farmers of eliminating US subsidies are compared with various productivity increasing measures for cotton and sorghum in Dioila, Mali. Dioila is located in a representative cotton region producing 16% of the cotton in Mali. We include sorghum due to its importance for consumption and the observation of Malian farmers substituting cereals (sorghum and maize) for cotton as the returns to cotton have fallen in the 21st Century. In the farm model, the elasticity of transmission of a change in the world cotton price to the farm gate price is taken into account. The gains from eliminating US subsides are small. In contrast, the various technological alternatives including Bt cotton introduction, the use of higher fertilization levels for cotton, and the introduction of improved sorghum cultivars and moderate fertilization along with a marketing package all have substantially higher returns Even with substantial improvement in the mechanisms enabling farmers to benefit from the higher prices resulting from elimination of US subsidies, there are still much higher returns resulting from the various types of productivity increases. 相似文献

When tropical shrimps are kept in recirculating aquaculture systems (RAS), one of the limiting factors is the maintenance of a sufficient water quality, and therefore, often disinfectants like peracetic acid (PAA) are added to the water either as prophylactic or treatment measure. In this study, PAA in concentrations of 0.1 mg/L, 1 mg/L and 10 mg/L was applied continuously for 56 days to small-scale seawater RAS stocked with Litopenaeus vannamei. Treatment with 0.1 mg/L did not result in a reduction in the total bacterial amount and therefore was not effective. A concentration of 10 mg/L led to significant changes in the chemical water parameters already after 2 days and was therefore not recommendable. A concentration of 1 mg/L led to increased levels of ammonia and nitrite within 2 days and to a significant increase in the bacterial amount in the water, most probably due to an enhanced growth of heterotrophic bacteria. The microflora showed significant fluctuations, and there were indications that the welfare of the shrimps was affected. Using 1 mg PAA/L for prophylactic use is therefore also not recommendable but might be an alternative option for short-term treatment in cases of disease outbreaks. 相似文献

A mono-proline analogue of the potent anthelmintic cyclooctadepsipeptide PF1022A has been synthesized. NMR studies in combination with molecular dynamics simulations demonstrate that the confirmation of this analogue is both rigidified and distorted compared with the asymmetric conformation of PF1022A. On some landscapes periodic fire may be necessary to develop and maintain oak-dominated savannas. We studied the effects of two annual prescribed burns to determine their effect on microbial activity and soil and litter nutrients 1 year after the last burn. Surface litter and soil from the upper 0–5 cm soil layer in three developing savannas (oak-hickory, Quercus-Carya), oak-hickory-pine (Quercus-Carya-Pinus), and pine (Pinus) were collected one year after the second of two annual prescribed burns. Surface litter was analyzed for nutrients and soil was analyzed for phospholipid fatty acids (PLFAs) and nutrients. Surface litter chemistry differed across the three savannas for potassium (K) and boron (B), being significantly (P < 0.05) higher for unburned forest than for burned forest. Among savannas, only sulfur (S) was higher for the pine savanna and B for the oak-hickory savanna, both were higher for unburned forest than for burned forest. For soil, calcium (Ca) and B differed across savannas, being higher for burned forest than for unburned forest. Among savannas, soil pH, Ca, and B concentrations were higher in soil from burned forest than from unburned forest. Total PLFA differed among savannas, but was not affected by burning treatments. However, the amounts of biomarkers for Gram-positive and Gram-negative bacteria were higher while the amount of biomarker for fungal PLFA was lower for burned forest than for unburned forest. Our results indicate that the two annual prescribed burns moderately affected PLFA microbial community structure and litter and soil nutrient concentrations. However, the long-term effects of fire on these study sites are not known and merit further study. 相似文献

Drosophila suzukii (Diptera: Drosophilidae) is a highly polyphagous invasive pest threatening fruit production in the Americas and Europe. The current knowledge of its host plants is mainly based on oviposition and larval development in fruits, while little is known on the diet of the adult flies. This information is important for developing effective control strategies. Here, we examine DNA-based techniques to determine food plants of D. suzukii. Adult flies were fed with raspberries (Rubus idaeus) and allowed to digest up to 72 h after feeding. Raspberry DNA was detected by diagnostic PCR for up to 48 h post-feeding with a significant negative effect of time on DNA detection success but no significant differences between male and female flies in detection probabilities. As D. suzukii walks on plants, its body surface can get contaminated with DNA. With a bleaching experiment, we succeeded to remove contaminating external plant DNA, while the DNA in the gut content stayed unaffected. Finally, field-collected flies were subjected to a next-generation sequencing approach, demonstrating that plant DNA from different host plants can be efficiently detected in both bleached and non-bleached specimens. In order to safeguard against erroneous host plant detections, we recommend bleaching flies before they are subjected to DNA extraction. The current findings encourage the use of DNA-based gut content analysis in D. suzukii to obtain a better understanding of its feeding ecology which is a prerequisite for developing successful control strategies. 相似文献

Biological control is an important ecosystem service delivered by natural enemies. Together with breeding for plant defence, it constitutes one of the most promising alternatives to pesticides for controlling herbivores in sustainable crop production. Especially induced plant defences may be promising targets in plant breeding for resistance against arthropod pests. Because they are activated upon herbivore damage, costs are only incurred when defence is needed. Moreover, they can be more specific than constitutive defences. Nevertheless, inducible defence traits that are harming plant pest organisms may interfere with biological control agents, such as predators and parasitoids. Despite the vast fundamental knowledge on plant defence mechanisms and their effects on natural enemies, our understanding of the feasibility of combining biological control with induced plant defence in practice is relatively poor. In this review, we focus on arthropod pest control and present the most important features of biological control with natural enemies and of induced plant defence. Furthermore, we show potential synergies and conflicts among them and, finally, identify gaps and list opportunities for their combined use in crop protection. We suggest that breeders should focus on inducible resistance traits that are compatible with the natural enemies of arthropod pests, specifically traits that help communities of natural enemies to build up. © 2017 The Authors.
