With the move away from use of mouse bioassay (MBA) to test bivalve mollusc shellfish for paralytic shellfish poisoning (PSP) toxins, countries around the world are having to adopt non-animal-based alternatives that fulfil ethical and legal requirements. Various assays have been developed which have been subjected to single-laboratory and multi-laboratory validation studies, gaining acceptance as official methods of analysis and approval for use in some countries as official control testing methods. The majority of validation studies conducted to date do not, however, incorporate shellfish species sourced from Latin America. Consequently, this study sought to investigate the performance of five alternative PSP testing methods together with the MBA, comparing the PSP toxin data generated both qualitatively and quantitatively. The methods included a receptor binding assay (RBA), two liquid chromatography with fluorescence detection (LC-FLD) methods including both pre-column and post-column oxidation, liquid chromatography with tandem mass spectrometry (LC-MS/MS) and a commercial lateral flow assay (LFA) from Scotia. A total of three hundred and forty-nine shellfish samples from Argentina, Mexico, Chile and Uruguay were assessed. For the majority of samples, qualitative results compared well between methods. Good statistical correlations were demonstrated between the majority of quantitative results, with a notably excellent correlation between the current EU reference method using pre-column oxidation LC-FLD and LC-MS/MS. The LFA showed great potential for qualitative determination of PSP toxins, although the findings of high numbers of false-positive results and two false negatives highlighted that some caution is still needed when interpreting results. This study demonstrated that effective replacement methods are available for countries that no longer wish to use the MBA, but highlighted the importance of comparing toxin data from the replacement method using local shellfish species of concern before implementing new methods in official control testing programs.  相似文献

Natural products containing a N‐heterocycle motif are widespread in nature and medicinal plants, in particular, have proved to be a source of almost unlimited N‐derived structures with high molecular diversity. Because of their intrinsic potential for use in both biomedical and agricultural applications, there is a general need for new compounds and for the synthesis of ‘natural‐inspired’ analogues. Importantly, transition of a natural product from discovery to a ‘market lead’ is associated with an increasingly challenging demand for more of the compound, which cannot be met by isolation from natural plant sources, often due to low extraction yields and uneven availability of the plant source itself. Synthesis remains the most reliable approach to provide valuable products for the market. In this review, a comprehensive overview of our contribution to synthetic access to N‐derived natural products is given. Longer life expectancy has led to an increase in efforts directed to the discovery of new healing agents for disorders related to aging, such as bone diseases. Harboring an incredible variety of bioactive metabolites, marine organisms are standing out as fruitful sources also in this therapeutic field. On the other hand, the in vivo zebrafish model has proven to be an excellent low-cost screening platform for the fast identification of molecules able to regulate bone development. By using zebrafish larvae as a mineralization model, we have thus evaluated the effects of the crude acetonic extract from the marine sponge Aplysina aerophoba and its bromotyrosine components on bone development. Obtained results led to the selection of aerophobin-1 (1) as a promising candidate for applications in regenerative medicine, paving the way for the development of a novel therapeutic option in osteoporosis treatment.  相似文献

1997年在墨西哥坦马乌利帕斯南部国家试验农场,对墨美中三国的26个棉花品种进行了比较研究。籽棉产量分析表明,品种间具有显著的差异。参试品种可以分为三种类型:第一类12个美国品种为高产类型,籽棉产量为2300~2800kg/hm2,包含3个佩马斯特品种,四个岱字棉品种和5个苏热格罗品种;第二种类型7个品种,属中产类型,籽棉产量为2000~2300kg/hm2,其中包括2个中国品种(湘棉10号和泗棉2号)和1个墨西哥品种CESTAM871。第三类7个品种,包括6个墨西哥品种和1个中国品种,籽棉产量为1800~2000kg/hm2。  相似文献

In Mexico most of the agricultural production originates from large irrigation districts in the northern part of the country. This region is characterized by its semiarid desert climate with a winter rainy season dominated by frontal storms, and a summer monsoon season dominated by highly localized convective storms, yielding most of the annual precipitation. Essentially all irrigation needs must be met by surface water stored in various reservoirs. Precipitation is, therefore, the most important limiting factor in Mexico's agricultural production. Traditionally, long-time averages of statistical climate data from few and widely-spaced weather stations were used to determine frequency and amount of water applied, and the algorithms employed usually did not consider the effects of great spatial climate variability and plant physiology. In the past five years, great parts of Mexico, especially in the North, have been affected by severe water shortages resulting from insufficient precipitation (perhaps related to the ‘El Niño' phenomenon), combined with inefficient water resources management. Irrigation districts increasingly have to deal with the considerable uncertainty in water resources availability as a limiting factor in the decision making process. In order to address these irrigation water shortages, the Mexican National Water Commission and the Mexican Water Resources Institute are introducing new technologies using agrometeorological networks for more efficient, real-time irrigation scheduling in the main irrigation districts of Mexico. Validation plots established in one particular irrigation district (Carrizo Valley, Sinaloa), demonstrate water savings in the order of at least 20% without any appreciable decrease in crop yields.  相似文献

Two major quantitative trait loci (QTLs) associated with resistance to powdery mildew (Blumeria graminis f. sp. hordei) were previously identified on chromosome 7H of the Spanish barley line SBCC097. The two QTLs seemed to share the same chromosomal position as the major genes mlt and Mlf, which were formerly described in Hordeum vulgare ssp. spontaneum-derived lines. In the present work, different lines that carry mlt (RS42-6*O), Mlf (RS137-28*E), or a combination of both (SI-4 and SI-6) were compared with SBCC097 to evaluate their relatedness at the phenotypic, cellular, and genetic levels. The resistance of the lines was characterised by inoculating them with a set of 27 isolates of B. graminis, which displayed a wide range of virulence. It was revealed that SBCC097 possessed a distinctive resistance spectrum. Microscopic assessment of the cytological development of the resistance response showed that SBCC097 clearly formed fewer well-established colonies and secondary hyphae than the other lines. This was confirmed by the infection type recorded after visual inspection. Genetic analyses of all five lines, based on markers flanking the QTLs derived from SBCC097, supported the macroscopic and microscopic data and pointed to the presence of a combination of novel genes or alleles in SBCC097, which may be included in the category of “intermediate-acting” genes, governing resistance mainly at the post-penetration stage.  相似文献

A two years lysimeter experiment was carried out using wheat plants (Triticum aestivum L. cv. Lotti) on two texturally contrasting soils. The main purpose of this study was to evaluate the influence of increasing applications (5,10, 15,20, and 25 t.ha‐1) of solid phase (SP) from pig slurry on soil nitrogen (N), phosphorus (P), potassium (K), calcium (Ca), magnesium (Mg), and sodium (Na) content, nitrate‐N (NO3‐N) leaching as well as on wheat composition and yield. As the control, a basic dressing of NPK fertilizer was applied. Results showed that plant growth was stimulated by increasing amounts of SP, yet the additions of 15 to 20 t SP ha‐1 led to similar effects on yield as that for the control. An accumulation of P on both soils was observed as well as a significant increase on NO3‐N leaching due to increasing rates of SP added to the soils. The N and P content in wheat plants (straw and grain) increased with increasing rates of applied SP.  相似文献

The characteristic of proteins, starch and pectic substances in cotyledons of two bean cultivars varying in cooking time were determined to investigate their possible contribution to bean cooking quality. Both cultivars showed the same enthalpies of starch gelatinization but different protein denaturation enthalpies. The proportion of hot water soluble pectins was higher in Michigan, the cultivar with the lower cooking time, than in Ojo de Cabra, the cultivar with the higher cooking time. These results were not due to differences in pectin methylation or in the ratio of monovalent to divalent cations in the tissue, suggesting that in fresh beans the β-elimination reaction is not the sole or predominant route of thermal pectin degradation. Overall, this study indicates that varietal differences in bean cooking quality may be reflections of the rate of pectin loss during soaking/heating and that the thermal properties of starch and protein fractions seem to have a minor contribution. Researchers involved in this study propose that in fresh beans, the thermal pectin loss results from a two step mechanism: pectin enzymic breakdown during the bean soaking followed by thermal solubilization rather than β-elimination during the bean heating.  相似文献

This article presents the results of a research study that analyzed the effect that secondary biofilm treatment systems had on particle size distribution (PSD) in the effluent of wastewater treatment plants. This study focused on three biofilm technologies (i.e. a submerged biofilter system, a trickling filter system, and a rotating biological contactor system) in three working urban wastewater treatment plants. For this purpose, the variation of the fit parameters was analyzed after modeling the PSD with the power law. The greatest reduction in particle number was obtained with the submerged biofilter system, followed by the trickling filter system. In contrast, the rotating biological contactor showed the smallest reduction in particle number under the conditions of our study. It was also found that the variation of the fit parameters of the PSD, caused by the wastewater treatments was related to other wastewater parameters, such as the chemical oxygen demand, suspended solids, and the mean particle size. This showed a direct relation between these parameters and the particles in wastewater.  相似文献

A trial to assess the effects of applying several Fe-containing formulations on Fe-deficient (chlorotic) peach leaves was carried out under field conditions. Solutions consisting of an Fe-containing compound (FeSO4·7H2O, Fe(III)–citrate, Fe(III)–EDTA, Fe(III)–DTPA or Fe(III)–IDHA) and one of five different surfactant treatments (no surfactant, an organo-silicon, an ethoxylated oil, a non-ionic alkyl polyglucoside and a household detergent) were applied to one half of the leaf via dipping, first at the beginning of the trial and then after 4 weeks. The re-greening of treated and untreated leaf areas was estimated with a SPAD apparatus, on a weekly basis, during 8 weeks. At the end of the experimental period, leaves were detached, and tissue Fe, N, P, K, Ca, Mg, Mn, Zn and Cu concentrations were determined in Fe-treated and untreated leaf areas. Treatment with Fe-containing solutions always resulted in leaf chlorophyll (Chl) increases, which however significantly depended on the Fe-source, the surfactant-type and the combination between both formulation components. Untreated leaf zones experienced a Chl increase only in some cases, and this depended on the type of surfactant used. Iron application significantly increased the Fe concentration of treated and untreated leaf areas, especially with some formulations. Foliar treatment with Fe-containing solutions induced significant changes in the concentration of several nutrients as compared to those found in Fe-deficient peach leaves, with changes being similar in treated and untreated leaf areas, although in some elements the extent of the changes was of a different magnitude in both materials. This indicates that some leaf mineral composition changes typical of chlorotic leaves are dependent on leaf Fe concentration rather than on leaf Chl levels. Results obtained are relevant to help understand the factors involved in the penetration and bioavailability of leaf-applied Fe, and to assess the potential of foliar Fe fertilization to control Fe deficiency in fruit trees.  相似文献
