The purpose of this study was to estimate the expression and relative amounts of estrogen (ER) and progesterone receptors (PR) and their isoforms as well as heat shock protein 70 (HSP70) in ovaries of rats with induced cystic ovarian disease (COD). Primary, secondary, tertiary, atretic and cystic follicles were evaluated by immunohistochemistry and total ovarian proteins were analyzed by Western blot. In the granulosa layer, growing and cystic follicles in the treated group have a higher expression of ERα than growing follicles of control individuals. In the theca interna layer, tertiary follicles presented a significantly higher expression of ERα in the treated group. An increase in total ERα protein was detected in the treated group. Granulosa cells of all growing, atretic and cystic follicles show a lower expression of ERβ in animals with COD, and the total protein expression of ERβ was lower in this group. The expression of PR was lower in the granulosa cell layer of tertiary and cystic follicles in treated animals, and theca interna layer had less intense immunostaining in this group. Although there were no differences in the expression of PR-B by Western blotting, the expression of PR-A was higher and the expression of PR-C was smaller in the treated group. An intense HSP70 immunostaining was observed in the cells of cystic follicles. By Western blotting, higher protein expression of HSP70 was detected in the ovarian samples of the control group than those of the treated ones. Ovaries of animals with COD exhibited an altered steroid receptor expression and subtype balance as compared with control animals, and an increase in HSP70 immunoexpression. 相似文献

Objective Part 1: compare the use of a Metricheck™ device (a stainless steel probe with a semi-spherical rubber cup attached at one end) to sample the contents of the anterior vagina with a vaginal speculum examination for the diagnosis of pus in the vagina of postpartum dairy cows and to investigate the association of that pus with reproductive performance. Part 2: assess the effect of a single intrauterine infusion of 500 mg cephapirin in cows diagnosed with vaginal purulent or mucopurulent discharge 7 to 28 days after calving on reproductive performance.
Procedure Six herds were visited fortnightly to examine cows that had calved between 7 and 28 days (n = 423) with both the Metricheck device and a vaginal speculum to score, by each method, the vaginal discharge from 0 (clear or absent) to 3 (purulent) for each animal included in the study. Half of the cows that had a positive discharge score (1 to 3 by either examination method) were then treated with an intrauterine infusion of 500 mg of cephapirin. The relationship between Metricheck score, vaginoscopy score, treatment and reproductive performance was assessed.
Results There was a substantial measure of agreement between each method when scores were analysed by status. Cows that were positive with either method had inferior reproductive performance compared with cows with a score of zero. Treatment of cows diagnosed with a purulent or mucopurulent discharge with intrauterine cephapirin improved reproductive performance in both the vaginoscopy and Metricheck groups. 相似文献

Typical controlled drainage structures in drainage ditches provide drainage management strategies for isolated temporal periods. Innovative, low-grade weirs are anticipated to provide hydraulic control on an annual basis, as well as be installed at multiple sites within the drainage ditch for improved spatial biogeochemical transformations. This study provides evidence toward the capacity of low-grade weirs for nutrient reductions, when compared to the typical controlled drainage structure of a slotted riser treatment. Three ditches with weirs were compared against three ditches with slotted risers, and two control ditches for hydraulic residence time (HRT) and nutrient reductions. There were no differences in water volume or HRT between weired and riser systems. Nutrient concentrations significantly decreased from inflow to outflow in both controlled drainage strategies, but there were few statistical differences in N and P concentration reductions between controlled drainage treatments. Similarly, there were significant declines in N and P loads, but no statistical differences in median N and P outflow loads between weir (W) and riser (R) ditches for dissolved inorganic phosphate (W, 92%; R, 94%), total inorganic phosphate (W, 86%; R, 88%), nitrate-N (W, 98%; R, 96%), and ammonium (W, 67%; R, 85%) when nutrients were introduced as runoff events. These results indicate the importance of HRT in improving nutrient reductions. Low-grade weirs should operate as important drainage control structures in reducing nutrient loads to downstream receiving systems if the hydraulic residence time of the system is significantly increased with multiple weirs, as a result of ditch length and slope. 相似文献

A high molecular weight polyhydroxyamide (PHA) solution in N, N-dimethyl acetamide (DMAc) was prepared from 3,3′-dihydroxybenzidine
and isophthalic chloride (IPC), which was used for spinning PHA fiber. Before spinning, the diffusion property of DMAc into
various coagulants was examined. The fiber was well formed in coagulants such as water/ethanol with a composition of 5/5,
ethanol, and ethanol/isopropanol with a composition of 7/3 and 5/5. However, the PHA fiber spun in the water/ethanol mixture
contained voids. After the fiber spun in ethanol was annealed at over 350°C, the ultimate stress and initial modulus of the
fiber increased from 75.5 MPa and 3.22 GPa to 369 MPa and 29.5 GPa, respectively. These properties of the PHA fiber spun by

Enhanced accumulation of follicular PGF2 alpha with respect to PGE2 during the later phase of the preovulatory period is an apparent prerequisite for ovulation in sheep. Prostaglandin (PG) E2-9-ketoreductase is the enzyme that converts PGE2 into PGF2 alpha. Expression of activity of this enzyme by tissue homogenates of preovulatory ovine follicles was assessed. Homogenates were incubated in the presence of tritiated PGE2. Prostaglandin F2 alpha (i.e., product) was separated from PGE2 by Sephadex chromatography and quantitated by liquid scintillation counting. Progesterone in follicular fluid was measured by RIA. Follicular activity of PGE2-9-ketoreductase and content of progesterone increased approximately sixfold as the time of ovulation approached. Formation of PGF2 alpha from PGE2 was not influenced by inhibition of follicular synthesis of prostaglandins by indomethacin, nor did such treatment affect follicular production of progesterone. Inhibition of follicular synthesis of progesterone by isoxazol suppressed enzymatic conversion of PGE2 into PGF2 alpha; this effect was reversed by progesterone. It appears that progesterone plays an intrafollicular role in induction of activity of PGE2-9-ketoreductase in sheep. 相似文献

The objective of this study was to examine the effects of canthaxanthin (Cx) treatment during in vitro maturation (IVM) of porcine oocytes on embryonic development after parthenogenetic activation (PA) and somatic cell nuclear transfer (SCNT), on intracellular glutathione (GSH) and reactive oxygen species (ROS) levels in mature oocytes, and on gene expression in both PA‐ and SCNT‐derived blastocysts. To determine the optimal effective concentration of Cx, porcine oocytes were cultured in IVM medium supplemented with various concentrations (0, 20, 40 and 80 μM) of Cx for 22 hr. Compared to other groups, supplementation with 40 μM Cx significantly improved blastocyst formation rates after PA (p < .05), but no significant differences were observed among groups in total blastocyst cell numbers. Subsequently, oocytes were cultured in IVM medium supplemented with or without 40 μM Cx. Oocytes treated with 40 μM Cx showed significantly increased cleavage and blastocyst formation rates after SCNT compared to the control group (p < .05). Moreover, significantly increased intracellular GSH and reduced ROS levels were observed in the Cx‐treated group (p < .05). In addition, both PA‐ and SCNT‐derived blastocysts from the 40 μM Cx‐treated group showed significantly increased mRNA expression of Bcl2 and Oct4 and decreased Caspase3 expression level (p < .05), when compared with the control group. PA‐derived blastocysts from the 40 μM Cx‐treated group also exhibited significantly decreased expression of Bax (p < .05). Our results demonstrated that treatment with 40 μM Cx during IVM improves the developmental competence of PA and SCNT embryos. Improvement of embryo development by Cx is most likely due to increased intracellular GSH synthesis, which reduces ROS levels in oocytes, and it may also positively regulate apoptosis‐ and development‐related genes. 相似文献
