Pectin methylesterase (PME) from carrots (Daucus carrota L.) was extracted and purified by affinity chromatography on a CNBr-Sepharose 4B-PME inhibitor column. A single protein and PME activity peak was obtained. A biochemical characterization in terms of molar mass (MM), isoelectric points (pI), and kinetic parameters of carrot PME was performed. In a second step, the thermal and high-pressure stability of the enzyme was studied. Isothermal and combined isothermal-isobaric inactivation of purified carrot PME could be described by a fractional-conversion model.  相似文献

Supplemental Feeding of Tilapia in Fertilized Ponds   总被引:1,自引:0,他引:1  
The addition of feed to fertilized fish ponds was evaluated by adding feed alone, feed plus fertilizer, or fertilizer alone to nine ponds stocked with Nile tilapia Oreochromis niloticus . Two experiments were conducted. The first had 500 fish per 250 m2 pond in 3 treatments: ad-libitum feeding; fertilizer only; or fertilizer and ad-libitum feeding. The second experiment had 5 treatments with 750 fish per pond ad-libitum feed only; fertilizer only; or 0.25, 0.50, and 0.75 satiation ration plus fertilizer. Ponds in Thailand were maintained for 155–162 d, during which chemical and physical properties were monitored. In experiment 1 tilapia growth was highest in feed only ponds, and lowest in fertilizer only ponds. Net yield did not differ significantly among treatments, due to variation in survival. In experiment 2, tilapia growth was lowest in fertilizer only ponds, intermediate in 0.25 ration ponds, and highest in 0.50, 0.75, and ad-libitum ponds. The latter treatments were not significantly different. Multiple regressions for each experiment indicated only 47–87% of the variance in growth was explained by feed and fertilizer input, while 52–89% of the variance in yield was explained by those factors. For both experiments combined, 90.3% of the variance in growth was explained by feed input, fertilizer input, alkalinity, and total inorganic nitrogen concentration. For yield, R 2 was 0.888 and the regression included feed input, pH, and number of low dissolved oxygen events. Experiment 1 appeared to approach carrying capacity near the end, while no reduction in growth occurred in experiment 2 at higher fish density and biomass. Reductions in growth in experiment 1 were not correlated with declining water quality late in the grow out. Combinations of feed and fertilizer were most efficient in growing tilapia to large size (500 g) compared to complete feeding or fertilizing alone.  相似文献

The German Salmonella Monitoring Programme started by the QS-System in 2002 (Blaha, 2004) is mandatory due to the so-called "Salmonella Regulation for Pigs" since 2007 (Anonym, 2007). The Regulation does not clearly prescribe the specific muscle which is to be taken as source of the meat juice. Thus, at different slaughter plants meat samples are also taken from different muscles and several scientific papers describe various muscles as source of the meat juice, too. The objective of this study was to compare the serological results of meat juices from three different locations (diaphragm pillar, neck, belly muscle) to each other and to those of the blood serum from exactly the same animals. All samples were simultaneously tested for Salmonella antibodies by two serological tests (Salmo-type Pig Screen, LDL, Germany; HerdChek Swine Salmonella, IDEXX, Germany). Comparisons were carried out between the various sample kinds per animal and between the two test systems. The analysis of all results of the meat juices revealed in both test systems a clear decline of the OD% values from the diaphragm pillar to the neck to the belly muscle. The average OD% values of all samples were higher when measured by the HerdChek ELISA (IDEXX, Germany) than by the Salmotype ELISA (LDL, Germany), especially in blood serum. Since the results of the meat juice samples gained from the diaphragm pillar were in both test systems by far the closest to the results of the corresponding serum blood samples, it is recommended to amend the "Salmonella Regulation for Pigs" by prescribing meat from the diaphragm pillar as the only muscle for gaining meat juice.  相似文献

Our previous studies showed that the subcutaneous pretreatment of rats with heat killed cells of Cryptococcus neoformans (HKC) emulsified in complete Freund adjuvant (CFA) promotes protection against an intraperitoneal challenge with viable C. neoformans. In this model, an appropriate activation of adherent peritoneal cells after antigenic treatment is very important for the control of the infection. Here, we investigated the immune response developed in spleen and lymphatic nodes as a result of treatment with HKC-CFA, which might also contribute in the protective phenomenon of this treatment against cryptococcal infection. The results show that, compared with adjuvant alone, rats which received treatment with HKC-CFA presented a greater activation of adherent splenic cells, with up-regulation of major histocompatibility complex class II (MHC II) and CD86 expression and secretion of anticryptococcal metabolites. Furthermore, this treatment also induced an increase in the blastogenic response and the secretion of Th1 and Th2 cytokines by spleen cells in comparison with cells from CFA-phosphate-buffered saline (PBS) treated rats. On the other hand, lymph node cells from animals treated with HKC-CFA presented a rise in the expression of MHCII but not of CD86 with respect to control cells from rats treated with CFA-PBS. These cells also showed a high proliferative response and secretion of Th1-related cytokines, interleukin (IL)-12 and tumor necrosis factor (TNF). These results show that treatment of rats with HKC-CFA is able to induce an early immune response in secondary lymphoid organs, which may contribute to the protective effect induced by this treatment.  相似文献

A carcinoid tumour in the liver of a red deer hind (Cervus elaphus) is described. Macroscopically, the liver was considerably enlarged with multifocal, firm, yellow and red nodular neoplastic masses, which were histopathologically diagnosed as hepatic carcinoids. The diagnosis was confirmed by modified Grimelius staining, which demonstrated numerous small argyrophilic granules in the cytoplasm of neoplastic cells, and by immunohistochemistry. The neoplastic cells gave a strong positive reaction for neuron-specific enolase (NSE) and synaptophysin and a weak positive reaction for chromogranin A. To our knowledge, this is the first report of a hepatic carcinoid in red deer.  相似文献
