The caprine arthrite encephalite (CAE) is a disease that affects especially dairy goat. The virus shows compartmentalization features, that allows it to hide at certain times during the course of the disease, making it difficult to control. The present study was conducted to identify the major seminal plasma protein profile of goats infected by CAE and its associations with seroconversion using Western blotting. Two groups containing five males each, were used in this experiment. The first group was composed by seropositive animals and the control by seronegative confirmed by Western blotting and PCR. The semen was collected through artificial vagina and after that, two-dimensional electrophoresis and MALDI-TOF MS were used. Seventy-five spots were identified in the goat seminal plasma gels, equivalent to 13 different proteins with more expression. The similar proteins found in both groups and related to reproduction were spermadhesin Z13-like, bodhesin and bodhesin-2, Lipocalin, protein PDC-109-like, and albumin. In infected goats, proteases such as arisulfatase A have been identified, whose function probably is related to metabolism control of sulfatides, involved to virus control. The other ones were bifunctional ATP-dependent dihydroxyacetone kinase/FAD-AMP lyase, cathepsin F isoform X1, disintegrin and metalloproteinase domain-containing protein 2-like isoform X1, clusterin, carbonic anhydrase 2, electron transfer flavoprotein subunit beta, and epididymal secretory glutathione peroxidase. We carried out an inter-laboratory trial to compare the serological tests commonly used for the detection of specific Neospora caninum antibodies in cattle in Ibero-American countries. A total of eight laboratories participated from the following countries: Argentina (n = 4), Brazil (n = 1), Peru (n = 1), Mexico (n = 1), and Spain (n = 1). A blind panel of well-characterized cattle sera (n = 143) and sera representative of the target population (n = 351) was tested by seven in-house indirect fluorescent antibody tests (IFATs 1–7) and three enzyme-linked immunosorbent assays (ELISAs 1–3; two in-house and one commercial). Diagnostic performance of the serological tests was calculated and compared according to the following criteria: (1) the “Pre-test information,” which uses previous epidemiological and serological data; (2) the “Majority of tests,” which classifies a serum as positive or negative according to the results obtained by most tests evaluated. Unexpectedly, six tests showed either sensitivity (Se) or specificity (Sp) values lower than 90%. In contrast, the best tests in terms of Se, Sp, and area under the ROC curve (AUC) values were IFAT 1 and optimized ELISA 1 and ELISA 2. We evaluated a high number of IFATs, which are the most widely used tests in Ibero-America. The significant discordances observed among the tests regardless of the criteria employed hinder control programs and urge the use of a common test or with similar performances to either the optimized IFAT 1 and ELISAs 1 and 2. 相似文献
The aim of this study was to evaluate the effect of supplementation of pig diets in the growth and termination phases with different calcium sources. In experiment I, 36 whole males were distributed in randomized blocks in six groups, with six replications. A basal diet was formulated to meet the animals’ nutritional requirements except for calcium (0.09%), and the sources evaluated (calcitic limestone, monodicalcium phosphate, calcinated bone flour, and oyster flour) replaced the basal diet to provide 0.59% of total calcium. To determine the endogenous calcium, a diet containing low calcium (0.019%) was given simultaneously to another group of animals. Feces and urine were collected for determination the coefficients of apparent and true digestibility. In experiment II, 160 piglets were distributed in randomized blocks in four treatments, with five replications and four animals per experimental unit. Carcass and performance parameters, calcium concentration in bone and serum, and bone parameters were evaluated. The data were submitted to analysis of variance and factorial. The calcium source did not influence the digestibility coefficients determined by total collection (P > 0.05). The digestibility of Ca from oyster flour estimated by collection with an indicator was higher than that from the other sources (P < 0.05). Calcium sources did not interfere in the evaluated parameters (P > 0.05). The sources studied in this work can be used to supplement growing pigs’ diets. 相似文献
This study aims to evaluate the effect of the Tifton 85 hay (Cynodon dactylon L. Pers) replacement by Orelha de Elefante Mexicana spineless cactus (Opuntia stricta [Haw.] Haw) on the nutrient intake and digestibility, growth performance, microbial protein synthesis, and efficiency of dietary nitrogen compound utilization in the diets of Girolando heifers. Twenty-four (3.5 months and 100 kg ± 3.5 kg) Girolando heifers (5/8 Holstein x Gyr) were used and arranged in a completely randomized design. Dry matter, organic matter, crude protein, and neutral detergent fiber decreased linearly as a function of the levels of Tifton hay (TH) replacement by Orelha de Elefante Mexicana spineless cactus (OEM). The non-fiber carbohydrate intake and coefficient of dry matter digestibility increased, while the neutral detergent fiber (NDF) digestibility decreased linearly with the OEM inclusion. After evaluating the growth performance by observing weight gain and morphometric measurements, there were no significant alterations. The urinary volume, nitrogen balance as well as the efficiency of nitrogen retention had increased, while the urinary and plasma urea concentrations had decreased linearly with the OEM inclusion. The results obtained in this study show that Orelha de Elefante Mexicana spineless cactus should replace Tifton hay in post-weaned heifer diets. 相似文献

The aim of this study was to evaluate the histological alterations in the digestive and renal systems of 35 male ovines in Paraíba, Brazil, at around 16?± 1.87 kg submitted to the following treatments for 90 days (15 of adaptation +?75 of experimental period): 1- Tifton hay (TH); 2- Cassava foliage hay (CH); 3–35% TH and 35% CH; 4–35% TH and 35% forage palm (FP), and 5–35% CH and 35% FP, based on dry matter (DM). Samples of the liver, kidney, rumen, and small intestine were histomorphometrically evaluated and the data were submitted to ANOVA analysis and Tukey post test at p ≤?0.05. The use of alternative foods in the voluminous fraction (CH?+?FP) in the ovine diets did not compromise the performance of the animals; on the contrary, promoted better gain when compared with TH treatment (18.55 kg vs 9.42 kg). The CH?+?FP diet had a positive effect on papilla height. The papilla width decreased with the diets CH?+?FP and TH?+?FP. The thickness of the ruminal muscular layer were higher in the diets containing TH?+?CH and CH?+?FP. The keratinized layer of the ruminal epithelium was larger in the diet containing CH and FP. The villi of the intestinal crypts were deeper in the diets with FP and the villus/crypt ratio was the same in all diets. Goblet cells decreased with the CH or FP diet. Some renal changes were observed according to the diets, mainly when using FP. It is recommended to use CH and FP as a large fraction of the ovines diet, which will not alter animal performance.

The aim of this study was to evaluate the caprine preantral follicles enclosed on vitrified/warmed ovarian cortex grafted to nude BALB/mice during 1 month. The ovarian cortex from goats was fragmented (3 × 3 × 0.5 mm) and divided into four groups: fresh control, vitrified control, fresh transplant and vitrified transplant. Follicular morphology, development and density, fibrosis as well as apoptosis, and tissue revascularization were evaluated. It was also observed a significant decrease in morphologically normal preantral (primordial, transition, primary and secondary) follicles in both vitrified control and vitrified transplant treatments when compared with both fresh control and fresh transplant. However, fresh control and fresh transplant exhibited a similar percentage of developing follicles. Additionally, Vitrified control showed a significant increase in developing follicles in comparison with both fresh control and fresh transplant. Follicular density significantly decreased in all treatments in comparison with fresh control. We observed high fibrosis in both fresh transplant and vitrified transplant. The mRNA expression of caspase 3 was lower in both fresh transplant and vitrified transplant in comparison with vitrified control. In conclusion, xenotransplantation is an excellent strategy to maintain normal preantral follicle morphology after vitrification/warming of goat ovarian tissue. Yet, in order to ensure the survival and development of these follicles, it is essential to improve the revascularization of the graft. 相似文献
The aim of the study was to determine the effect of dietary supplementation of a probiotic and a synbiotic on the morphometric parameters of the small intestine of broiler chickens. The experiment was conducted on three hundred sixty, one‐day‐old female Ross 308 chicks, which were randomly selected from 20,000 birds and divided into three treatment groups (n = 120) with ten replicates per treatment. The control group (C) was fed a commercial diet, the probiotic group (PRO) was fed the same diet with an added 1% of the probiotic Lavipan® (Lactococcus lactis, Carnobacterium divergens, Lactobacillus casei, Lactobacillus plantarum and Saccharomyces cerevisiae), and the synbiotic group (SYN) was fed the commercial diet with an added synbiotic: 0.8% of the prebiotic RFO (extracted from lupin seeds) and 1% Lavipan®. According to the manufacturer's data, apart from the typical probiotic action,microorganisms contained in the preparation release anti‐bacterial substances (hydrogen peroxide and bacteriocins) and, therefore, are antagonistic towards pathogenic bacteria present in the gut of animals. Supplementation took place in the first seven days of rearing, and all birds had ad libitum access to water and feed during the 42 days of the experiment. On the last day, all birds were slaughtered and samples from three segments of the small intestine were taken. Villi area, height, width and crypt depth ratios were read using Multiscan software. Synbiotic supplementation increased the BWG of broilers from first to tenth day of rearing, compared to the control group. The PRO group had improved villi morphometric parameters of the duodenum. In the jejunum and ileum, both bioactive substances improved villus width and villus surface area. Crypts were deeper in the small intestine of birds supplemented with bioactive substances, which allows greater renewal of the villi. As expected, the intestinal morphometric parameters of broiler chickens benefited from bioactive substance supplementation. 相似文献
