An international CCQM-P60 pilot study involving eight national metrological institutes was organized to investigate if the quantification of genetically modified (GM) corn powder by real-time PCR was affected by the DNA extraction method applied. Four commonly used extraction methods were compared for the extraction of DNA from a GM Bt176 corn powder. The CTAB-based method yielded the highest DNA template quantity and quality. A difference in the 260 nm/230 nm absorbance ratio was observed among the different extraction methods. Real-time amplification of sequences specific for endogenous genes zein and hmg as well as transgenic sequences within the cryIA(b) gene and a fragment covering the junction between the transformed DNA and the plant genome were used to determine the GM percentage. The detection of the transgenic gene was affected by the quantity and quality of template used for the PCR reaction. The Bt176 percentages measured on diluted or purified templates were statistically different depending on the extraction method applied.  相似文献

Maxillary incisor pseudo‐odontomas are common in pet prairie dogs and can cause progressive respiratory obstruction, while mandibular pseudo‐odontomas are rarely clinically significant. The aim of this retrospective cross‐sectional study was to describe CT features of maxillary and mandibular incisor pseudo‐odontomas vs. normal incisors in a group of pet prairie dogs. All pet prairie dogs with head CT scans acquired during the period of 2013–2015 were included. A veterinary radiologist who was aware of final diagnosis reviewed CT scans and recorded qualitative features of affected and normal incisors. Mean density values for the pulp cavity and palatal and buccal dentin were also recorded. A total of 16 prairie dogs were sampled (12 normal maxillary incisors, 20 confirmed maxillary incisor pseudo‐odontomas, 20 normal mandibular incisors, 12 presumed mandibular incisor pseudo‐odontomas). Maxillary incisors with confirmed pseudo‐odontomas had a significantly hyperattenuating pulp and dentin in the reserve crown and apical zone, when compared to normal maxillary incisors. Pseudo‐odontomas appeared as enlargements of the apical zone with a globular/multilobular hyperattenuating mass formation haphazardly arranged, encroaching on midline and growing caudally and ventrally. Presumed mandibular incisor pseudo‐odontomas had similar CT characteristics. In 60% of prairie dogs with maxillary incisor pseudo‐odontomas, the hard palate was deformed and the mass bulged into the oral cavity causing loss of the palatine bone. The common nasal meatus was partially or totally obliterated in 81.8% of prairie dogs with maxillary pseudo‐odontomas. Findings supported the use of CT for characterizing extent of involvement and surgical planning in prairie dogs with pseudo‐odontomas.  相似文献

We determined for the first time the reproductive biology of Piabina argentea through macroscopic and microscopic analysis of ovaries and evaluated the morphological changes in hepatocytes. Two hundred and 46 specimens were collected, 204 females and 42 males, between March 2014 and February 2015. Biometrics data were obtained. From females, gonad and liver samples were conducted to histological, histochemical and immunohistochemical techniques. Mature ovaries were used to determine absolute and relative fecundity. Total length and body weight values indicated that females were larger than males. The estimated weight–length ratio showed negative allometric growth. The absolute fecundity average was 171.83 ± 59.89 oocytes per ovary. In addition, females spawning capable and regressing stages were found throughout the sampling period and the presence of all oocyte types in regressing stage ovaries indicated asynchronous oocyte development and multiple spawning. From regenerating to spawning capable stage the oocytes accumulated yolk in cytoplasm became bigger. While in the liver hepatocytes with a larger cell area during regenerating stage and proliferative activity in the spawning capable stage were observed. Thus, our results indicate that P. argentea had an opportunistic reproductive strategy and cyclic morphological changes of hepatocytes occurred during the oogenesis.  相似文献

Cardiorespiratory and blood gas alterations were evaluated in 6 healthy dogs that underwent a laparoscopic procedure using isoflurane anesthesia and carbon dioxide (CO(2)) pneumoperitoneum for 30 min. Heart rate, respiratory rate, body temperature, venous blood pH, partial pressure of CO(2) and oxygen, oxygen saturation, total carbon dioxide (TCO(2)) and bicarbonate were monitored. Significant alterations were hypercapnia, hypoventilation, and respiratory acidosis.  相似文献

We characterised the among species variability in leaf gas exchange and morphological traits under controlled conditions of seedlings of 22 tropical rainforest canopy species to understand the origin of the variability in leaf carbon isotope discrimination (Δ) among species with different growth and dynamic characteristics (successional gradient). Our results first suggest that these species pursue a consistent strategy in terms of Δ throughout their ontogeny (juveniles grown here versus canopy adult trees from the natural forest). Second, leaf Δ was negatively correlated with WUE and N, and positively correlated with gs, but among species differences in Δ were mainly explained by differences in WUE. Finally, species belonging to different successional groups display distinct leaf functional and morphological traits. We confirmed that fast growing early successional species maximise carbon assimilation with high stomatal conductance. In contrast, fast and slow growing late successional species are both characterised by low carbon assimilation values, but by distinct stomatal conductance and leaf morphological features. Along the successional gradient, these differences result in much lower Δ for the intermediate species (i.e. fast growing late successional) as compared to the two other groups.  相似文献

The Influence of Landscape Structure on Female Roe Deer Home-range Size   总被引:5,自引:0,他引:5  
Animal distribution and abundance are greatly affected by the availability of their food resources, which also depends on landscape structure. Lothar hurricane in 1999 had profoundly modified the structure of the forests in France, affecting the habitat quality of ungulates. We tested whether the variations in home-range size of 23 female roe deer were influenced by the fragmentation of the landscape caused by Lothar in the Chizé forest, namely by the increase in heterogeneity associated with the localized massive tree felling. Home-range size was studied in the summers of 2001 and 2002 and we found that variation in home-range size was mainly explained by only one landscape variable: edge density. Home-range size decreased as edge density increased, which is consistent with the fact that edges are good browsing habitats for roe deer. The result of this study suggests that, after 2 years, the hurricane had improved the quality of the home ranges by creating more forest heterogeneity and increasing the contacts between the different vegetation patches within the home range. These results highlight the fact that spatial heterogeneity is likely to be a key factor influencing the distribution and local population density.  相似文献
