The characteristics of dissolved organic matter (DOM) in soils are often determined through laboratory experiments. Many different protocols can be used to extract organic matter from soil. In this study, we used five air‐dried soils to compare three extraction methods for water‐extractable organic matter (WEOM) as follows: (i) pressurised hot‐water‐extractable organic carbon (PH‐WEOC), a percolation at high pressure and temperature; (ii) water‐extractable organic carbon (WEOC), a 1‐hour end‐over shaking; and (iii) leaching‐extractable organic carbon (LEOC), a leaching of soil columns at ambient conditions. We quantified the extraction yield of organic carbon; the quality of WEOM was characterized by UV absorbance, potential biodegradability (48‐day incubation) and parallel factor analysis (PARAFAC) modelling of fluorescence excitation emission matrices (FEEMs). Biodegradation of dissolved organic carbon (DOC) was described by two pools of organic C. The proportions of labile and stable DOC pools differed only slightly between the WEOC and LEOC methods, while PH‐WEOC contains more stable DOC. The mineralization rate constants of both labile and stable DOC pools were similar for the three methods. The FEEMs were decomposed into three components: two humic‐like fluorophores and a tryptophan‐like fluorophore. The effect of extraction method was poorly discriminant and the most similar procedures were PH‐WEOC and LEOC while WEOC extracts were depleted in humic‐like fluorophores. This study demonstrates that WEOM quality is primarily determined by soil characteristics and that the extraction method has a smaller, but still significant, impact on WEOM quality. Furthermore, we observed considerable interaction between extraction procedure and soil type, showing that method‐induced differences in WEOM quality vary with soil characteristics.  相似文献

Phosphorus (P) deficiency is one of the most yield limiting factors for crop production in South American soils. Upland rice (Oryza sativa L.) is an important crop in South American cropping systems, including Brazil. A field experiment was conducted with the objective to evaluate 20 upland rice genotypes for phosphorus (P) use efficiency. The P rate used was low (0 kg P ha?1) and high [87 kg P ha?1 or 200 kg phosphorus pentoxide (P2O5) ha?1]. Plant height, shoot dry weight, grain yield, panicle number, 1000 grain weight, spikelet sterility, and grain harvest index were significantly influenced by P and genotype treatments. The P X genotype interaction was significant for grain yield, indicating that genotypes responded differently under two P rates. Overall, grain yield increased by 12% with the addition of P fertilization. Based on grain yield efficiency index, genotypes were classified into efficient, moderately efficient, and inefficient group. The genotypes that were classified as efficient in P use were BRA032048, BRA042094, BRA02601, BRA032051, BRA032033, BRA052015, BRA042156, BRA01600, BRA01506, BRA052023 and BRA042160. The inefficient genotypes in P us efficiency were BRS Primavera, BRA052045, BRA01596, and BRS Sertaneja. Grain harvest index had a significant positive association with grain yield and spikelet sterility had a significant negative association with grain yield, as expected. Average, P-use efficiency of five genotypes was about 17 kg kg?1 (kg grain yield per kg P applied).  相似文献

Soybeans contain oil bodies that are coated by a layer of oleosin proteins. In nature, this protein coating protects the oil bodies from environmental stresses and may be utilized by food manufacturers for the same purpose. In this study, oil bodies were extracted from soybean using an aqueous extraction method that involved blending, dispersion (pH 8.6), filtration, and centrifugation steps. The influence of NaCl (0-250 mM), thermal processing (30-90 degrees C, 20 min) and pH (2-8) on the properties and stability of the oil bodies was analyzed using zeta-potential, particle size, and creaming stability measurements. The extracted oil bodies were relatively small ( d 32 approximately 250 nm), and their zeta-potential went from around +12 mV to -20 mV as the pH was increased from 2 to 8, with an isoelectric point around pH 4. Yessotoxins from a large-scale culture (226 L) of Protoceratium reticulatum strain CAWD129 were harvested by filtration followed by solid-phase extraction. The extract was purified by column chromatography over basic alumina and reverse-phase flash chromatography to afford pure yessotoxin (193 mg). Isolation of yessotoxin was greatly facilitated by selection of a strain which did not produce analogues that interfered with yessotoxin isolation. In addition to yessotoxin, numerous minor yessotoxins were detected by LC-MS in other fractions. From one of these, an early eluting minor analogue with the same molecular weight as yessotoxin and a similar mass spectrometric fragmentation pattern was isolated. This analogue was identified by NMR and mass spectrometry as a novel yessotoxin analogue containing a furan ring in the side chain. This finding reveals biosynthetic flexibility of the yessotoxin pathway in P. reticulatum and confirms earlier findings of production of many minor yessotoxin analogues by this alga. Production of these analogues appeared to be a constitutive trait of P. reticulatum CAWD129.  相似文献

The greenhouse gases CO2 and N2O emissions were quantified in a long-term experiment in northern France, in which no-till (NT) and conventional tillage (CT) had been differentiated during 32 years in plots under a maize–wheat rotation. Continuous CO2 and periodical N2O soil emission measurements were performed during two periods: under maize cultivation (April 2003–July 2003) and during the fallow period after wheat harvest (August 2003–March 2004). In order to document the dynamics and importance of these emissions, soil organic C and mineral N, residue decomposition, soil potential for CO2 emission and climatic data were measured. CO2 emissions were significantly larger in NT on 53% and in CT on 6% of the days. From April to July 2003 and from November 2003 to March 2004, the cumulated CO2 emissions did not differ significantly between CT and NT. However, the cumulated CO2 emissions from August to November 2003 were considerably larger for NT than for CT. Over the entire 331 days of measurement, CT and NT emitted 3160 ± 269 and 4064 ± 138 kg CO2-C ha−1, respectively. The differences in CO2 emissions in the two tillage systems resulted from the soil climatic conditions and the amounts and location of crop residues and SOM. A large proportion of the CO2 emissions in NT over the entire measurement period was probably due to the decomposition of old weathered residues. NT tended to emit more N2O than CT over the entire measurement period. However differences were statistically significant in only half of the cases due to important variability. N2O emissions were generally less than 5 g N ha−1 day−1, except for a few dates where emission increased up to 21 g N ha−1 day−1. These N2O fluxes represented 0.80 ± 0.15 and 1.32 ± 0.52 kg N2O-N ha−1 year−1 for CT and NT, respectively. Depending on the periods, a large part of the N2O emissions occurred was probably induced by nitrification, since soil conditions were not favorable for denitrification. Finally, for the period of measurement after 32 years of tillage treatments, the NT system emitted more greenhouses gases (CO2 and N2O) to the atmosphere on an annual basis than the CT system.  相似文献

In this study, sn-1,2-, sn-2,3-, and sn-1,3-diacylglycerols were isolated from olive oil, and their urethane derivatives (urethanes) were prepared. Normal-phase high-performance liquid chromatography (NP-HPLC) separation of the urethane isomers was performed and the separate classes were studied by nuclear magnetic resonance (NMR). The use of 1H NMR and homo- and heteronuclear 2D techniques provided a great amount of information in a very short time, particularly when a high-field NMR instrument (700 MHz) was used. Particularly diagnostic for this kind of compound was the glyceridic moiety that presents typical chemical shifts both for carbon and hydrogen. These studies show the usefulness of NMR spectroscopy to recognize clearly the sn-1,3- and, moreover, sn-1,2- with respect to sn-2,3-diacylglycerols, although very minor differences occur between them.  相似文献

Nematicides are widely used to control plant-parasitic nematodes in intensive export banana (Musa spp.) cropping systems. Data show that the concentration of fosthiazate in banana fruits varies from zero to 0.035 g kg-1, under the maximal residue limit (MRL=0.05 mg kg-1). The fosthiazate concentration in fruit is described by a Gaussian envelope curve function of the interval between pesticide application and fruit harvest (preharvest interval). The heterogeneity of phenological stages in a banana population increases over time, and thus the preharvest interval of fruits harvested after a pesticide application varies over time. A phenological model was used to simulate the long-term harvest dynamics of banana at field scale. Simulations show that the mean fosthiazate concentration in fruits varies according to nematicide application program, climate (temperature), and planting date of the banana field. This method is used to assess the percentage of harvested bunches that exceed a residue threshold and to help farmers minimize fosthiazate residues in bananas.  相似文献
