Drought is a major constraint to productivity of Solanum aethiopicum ‘Shum’ group due to loss in market and nutrient value of stressed plants. This study evaluated S. aethiopicum Shum group accessions to identify genotypes (G) that excel across moisture deficit stress levels (WLs). A split-plot arrangement composed of four WLs and twenty accessions of S. aethiopicum as main plot and sub-plot factors, respectively, was implemented in a screenhouse, and repeated for two experiments. In each experiment, there was a highly significant effect of at least two WLs on mean performance among at least two accessions for most of the traits at p < 0.05. Further, very highly significant WL × G interactions were obtained for leaf relative water content (LRWC), leaves per plant (LPP) and plant height (PH), and non-significant for leaf blade length and leaf blade width. The order of priority as breeding traits for stability superiority across WLs was suggested as LRWC > PH > LPP. Consequently, based on LRWC, the most superiorly stable accessions were identified as accession 160 followed by accessions 145, 137, 108P and 184G while the least stable ones were identified as accessions 163G, 141, 163 and 108. The broad sense heritability (H 2) for each of the three recommended traits for drought resistance breeding was above 0.9 thus supportive for a good response to selection. Drought stress negatively affected the performance of S. aethiopicum Shum group but the exhibited variation allowed for selection of superiorly stable genotypes.  相似文献

Both plant species and CO2 concentration can potentially affect rhizodeposition and consequently soil microbial activity and community composition. However, the effect differs based on plant developmental stage. We focused on the effect of three plant species (forbs, grasses, and N2‐fixers) at an early stage of development on root C deposition and fate, soil organic matter (SOM) mineralization and soil microbial community composition at ambient (aCO2) and elevated (eCO2) CO2 levels. Plants were grown from seed, under continuous 13C‐labelling atmospheres (400 and 800 µmol mol?1 CO2), in grassland soil for three weeks. At the end of the growth period, soil respiration, dissolved organic C (DOC) and phospholipid fatty acid (PLFA) profiles were quantified and isotopically partitioned into root‐ and soil‐derived components. Root‐derived DOC (0.53 ± 0.34 and 0.26 ± 0.29 µg mL soil solution?1) and soil‐derived CO2 (6.14 ± 0.55 and 5.04 ± 0.44 µg CO2‐C h?1) were on average two times and 22% higher at eCO2 than at aCO2, respectively. Plant species differed in exudate production at aCO2 (0.11 ± 0.11, 0.10 ± 0.18, and 0.58 ± 0.58 µg mL soil solution?1 for Plantago, Festuca, and Lotus, respectively) but not at eCO2 (0.20 ± 0.28, 0.66 ± 0.32, and 0.75 ± 0.15 µg mL soil solution?1 for Plantago, Festuca, and Lotus, respectively). However, no differences among plant species or CO2 levels were apparent when DOC was expressed per gram of roots. Relative abundance of PLFAs did not differ between the two CO2 levels. A higher abundance of actinobacteria and G‐positive bacteria occurred in unplanted (8.07 ± 0.48 and 24.36 ± 1.18 mol%) and Festuca‐affected (7.63 ± 0.31 and 23.62 ± 0.69 mol%) soil than in Plantago‐ (7.04 ± 0.36 and 23.41 ± 1.13 mol%) and Lotus‐affected (7.24 ± 0.17 and 23.13 ± 0.52 mol%) soil. In conclusion, the differences in root exudate production and soil respiration are mainly caused by differences in root biomass at an early stage of development. However, plant species evidently produce root exudates of varying quality affecting associated microbial community composition.  相似文献

Safe and economical disposal of paper mill sludge is a key consideration for forest products industry. A study was conducted to examine the effects of amendments of sludge and nutrients on soil surface CO2 flux (Rs) in northern hardwood forests and to quantify the relat among R5, soil temperature, and moisture in these stands. The experiment was a randomized complete block design that included sludge-amended, fertilized, and control treatments in sugar maple (Acer saccharum Marsh) dominated hardwood forests in the Upper Peninsula of Michigan, USA. Results showed that Rs was positively correlated to soil temperature (R2=0.80, p<0.001), but was poorly correlated to soil moisture. Soil moisture positively affected the Rs only in the sludge-amended treatment. The Rs was significantly greater in the sludge-amended treatment than in the fertilized (p=0.033) and the control (p=0.048) treatments. The maximum Rs in the sludge-amended treatment was 8.8 μmol CO2·m−2·s−1, 91% and 126% greater than those in the fertilized (4.6 μmol CO2·m−2·s−1) and control (3.9 μmol CO2·m−2·s−1) treatments, respectively. The Rs did not differ significantly between the fertilized and control treatments. The difference in Rs between sludge-amended and the other treatments decreased with time following treatment. OBJECTIVE: To characterize and purify covalent complexes of matrix metalloproteinase-9 (MMP-9) and haptoglobin released by bovine granulocytes in vitro. SAMPLE POPULATION: Blood samples obtained from healthy cows and cows with acute and chronic inflammation to obtain WBCs and sera. PROCEDURES: WBCs were isolated by differential centrifugation, hypotonic lysis of RBCs, and degranulated by stimulation with phorbol ester (20 ng/mL). Cell-conditioned medium was subjected to affinity and gel chromatography and purified proteins subjected to SDS- PAGE gelatin zymography, western blot analysis, Coomassie blue staining, and peptide mass spectrometry for protein identification. Sera of cows hospitalized for acute and chronic septic conditions and of clinically normal cows were analyzed with similar methods. RESULTS: Matrix metalloproteinase-9 was released from neutrophils in vitro and migrated to a molecular mass of approximately 220 kd (prodimer), approximately 105 kd (promonomer), and > 220 kd (high-molecular mass complexes). These high-molecular mass complexes were composed of alpha- and beta-haptoglobin and MMP-9 (ratio13:13:1). Complexes of MMP-9 and haptoglobin had biochemical properties of both its protein constituents (i.e., enzymatic activity toward gelatin and hemoglobin binding). Complexes of MMP-9 and haptoglobin were also detected in sera of cows with acute inflammation, but not in clinically normal cows or cows with chronic disease. CONCLUSIONS AND CLINICAL RELEVANCE: A fraction of neutrophil MMP-9 is released in complex with haptoglobin. The complex is present in granules and retains biological activity of its components. Detection of the complex in serum may provide an indicator of acute inflammation.  相似文献

The antioxidant activities of aqueous phase beta-lactoglobulin (beta-Lg) and its chymotryptic hydrolysates (CTH) were compared in this study. Proteins and peptides have been shown to inhibit lipid oxidation reactions in oil-in-water emulsions; however, a more fundamental understanding of the antioxidant activity of these compounds in dispersed food lipid systems is lacking. CTH was more effective than an equivalent concentration of beta-Lg in retarding lipid oxidation reactions when dispersed in the continuous phase of Brij-stabilized oil-in-water emulsions (pH 7). Furthermore, it was observed that CTH had higher peroxyl radical scavenging and iron-binding values than beta-Lg. Liquid chromatography-mass spectrometry (LC-MS) was used to measure the rate of oxidation of three oxidatively labile amino acid residues (Tyr, Met, and Phe) in certain CTH peptide fragments. Significant oxidation of specific Tyr and Met residues present in two separate 12 amino acid peptide fragments was observed in the days preceding lipid oxidation (39 and 55% of Tyr and Met were oxidized, respectively, by day 4 of the study); however, no significant oxidation of the Phe residue present in a specific 14 amino acid peptide fragment could be observed during the same time period. These data could suggest that Met and Tyr residues are capable of scavenging radical species and have the potential to improve the oxidative stability dispersed food lipids.  相似文献

Tobacco (Nicotiana tabacum) plants grown from seed obtained by crossing a tobacco line that expressed an activated maize ribosome-inactivating protein (RIP) with a line that overexpressed tobacco anionic peroxidase were tested for their effects on corn earworm Helicoverpa zea and cigarette beetle Lasioderma serricorne larvae as compared to the wild-type plant cross. Significant feeding reductions were noted for transgenic plants expressing both resistance proteins as compared to wild-type plants for both H. zea and L. serricorne. Significant increases in mortality were also noted for those insects fed on the transgenic cross as compared to wild-type plants in some cases. Levels of both peroxidase and maize RIP were significantly higher in transgenic as compared to wild-type plants (which did not produce maize RIP). The degree of feeding was significantly negatively correlated with the level of RIP or peroxidase individually.  相似文献

At periods greater than 1000 seconds, Earth's seismic free oscillations have anomalously large amplitude when referenced to the Harvard Centroid Moment Tensor fault mechanism, which is estimated from 300- to 500-second surface waves. By using more realistic rupture models on a steeper fault derived from seismic body and surface waves, we approximated free oscillation amplitudes with a seismic moment (6.5 x 10(22) Newton.meters) that corresponds to a moment magnitude of 9.15. With a rupture duration of 600 seconds, the fault-rupture models represent seismic observations adequately but underpredict geodetic displacements that argue for slow fault motion beneath the Nicobar and Andaman islands.  相似文献

During 2004, 49 states and Puerto Rico reported 6,836 cases of rabies in nonhuman animals and 8 cases in human beings to the CDC, representing a 4.6% decrease from the 7,170 cases in nonhuman animals and 3 cases in human beings reported in 2003. Approximately 92% of the cases were in wildlife, and 8% were in domestic animals (compared with 91% and 9%, respectively, in 2003). Relative contributions by the major animal groups were as follows: 2,564 raccoons (37.5%), 1,856 skunks (27.1%), 1,361 bats (19.9%), 389 foxes (5.7%), 281 cats (4.1%), 115 cattle (1.7%), and 94 dogs (1.4%). Compared with the numbers of reported cases in 2003, cases in 2004 decreased among all groups, except bats, cattle, human beings, and "other domestics" (1 llama). Decreases in numbers of rabid raccoons during 2004 were reported by 12 of the 20 eastern states in which raccoon rabies was enzootic. In the East, Massachusetts reported the first cases of raccoon rabies detected beyond the Cape Cod oral rabies vaccine barrier. Along the western edge of the raccoon rabies epizootic (Ohio in the north and Tennessee in the south), cases of rabies were reported from unexpected new foci beyond oral rabies vaccine zones. On a national level, the number of rabies cases in skunks during 2004 decreased by 12.1% from the number reported in 2003. Once again, Texas reported the greatest number (n = 534) of rabid skunks and the greatest overall state total of rabies cases (913). Texas reported only 1 case of rabies in a dog that was infected with the dog/coyote rabies virus variant and only 22 cases associated with theTexas gray fox rabies virus variant (compared with 61 cases in 2003). The total number of cases of rabies reported nationally in foxes and raccoons declined 14.7% and 2.7%, respectively, during 2004. The 1,361 cases of rabies reported in bats during 2004 represented a 12.3% increase over the previous year's total of 1,212 cases for this group of mammals. Cases of rabies reported in cats, dogs, horses and mules, and sheep and goats decreased 12.5%, 19.7%, 31.8%, and 16.7%, respectively, whereas cases reported in cattle increased 174%. In Puerto Rico, reported cases of rabies in mongooses decreased 4.1% and rabies in dogs (9 cases) remained unchanged from those reported in 2003. Among the 8 cases of rabies in human beings, 1 person from Oklahoma and 3 from Texas died following receipt of infected organs and tissues from an Arkansas donor. In California, a person originally from El Salvador and, in Florida, a person originally from Haiti both died of canine rabies infections acquired outside the United States. In Wisconsin, a teenager contracted rabies from a bat bite and became the first known person to survive rabies despite not having received rabies vaccine prior to symptom onset.  相似文献
