In order to elucidate the effect of stress on reproductive hormones, the present study was designed to investigate the effect of adrenocorticotropic hormone (ACTH) on the plasma levels of cortisol, progesterone, oestradiol-17 beta and prostaglandin F2 alpha metabolite in ovariectomized gilts. Ovariectomy and cannulation of the jugular vein were performed within 1 week of oestrous detection, under general anaesthesia. Approximately 1 week after surgery, two gilts were each administered ACTH (Synacthen Depot) intravenously, at a dose of 0.01 mg/kg body weight, and one gilt was given saline solution (5 ml). The reverse was performed on the following day. The administration of ACTH was followed by a concomitant elevation of cortisol, progesterone and prostaglandin F2 alpha metabolite but not of oestradiol-17 beta. Peak cortisol, progesterone and prostaglandin F2 alpha metabolite levels were reached at 80 +/- 10.0, 80 +/- 10.0 and 46.6 +/- 13.3 min after ACTH administration and the durations of the peaks were 181.8 +/- 19.8, 308.1 +/- 49.7 and 181.8 +/- 7.9 min, respectively. The total area under the curve for cortisol, progesterone and prostaglandin F2 alpha metabolite was significantly higher in the ACTH than in the control group. The present results indicate that during stress, cortisol, progesterone and prostaglandin F2 alpha levels are elevated while the level of oestradiol-17 beta is less affected. It can be concluded that the administration of ACTH to ovariectomized gilts, results in the elevation of cortisol, progesterone and prostaglandin F2 alpha metabolite but not of oestradiol-17 beta.  相似文献

The pituitary hormones prolactin and oxytocin play important roles in the production and ejection of milk. In addition, some gastrointestinal peptides are released in response to suckling. During suckling, the piglets massage the udder of the sow both before and after let-down and the duration of suckling is correlated to the amount of milk produced by the sow. The aim of this study was to investigate whether there is a quantitative relation between the release of prolactin, gastrin, somatostatin, insulin, glucagon and vasoactive intestinal polypeptide (VIP) and the amount of stimulation of the sow's teats by the piglets.Repeated blood samples were drawn from three Swedish Landrace sows during three consecutive nursings by each sow on days 1, 3, 7 and 14 after parturition. The duration of massage by the piglets was noted, as was the number of piglets massaging. Hormone levels were quantified by radioimmunoassay.The releases of prolactin, somatostatin, insulin, glucagon and VIP but not of gastrin were found to be significantly related to the amount of teat massage performed by the piglets during the first 2 weeks of lactation. The release was related to the duration of piglet massage or to the combined effect of duration and the number of piglets massaging but not to the number of piglets massaging per se. The basal level of prolactin was found to decrease during this time.  相似文献

Androstenone is one of the main compounds responsible for boar taint, and 3beta-hydroxysteroid dehydrogenase (3betaHSD) might be involved in its metabolism. In this study, the gene expression of 3betaHSD and 17beta-hydroxysteroid dehydrogenase (17betaHSD) were determined by real-time PCR analysis and related to the concentrations of androstenone, testosterone, and estrone sulphate (E1S). The experiments were performed on gonadally intact male pigs classified based on high or low fat androstenone concentrations, as predetermined by HPLC, as well as on immunocastrated and surgically castrated male pigs. The male pigs with high androstenone concentrations in fat had low 3betaHSD gene expression in liver and testis. Moreover, the 17betaHSD gene expression in liver, but not in testis, varied negatively with fat androstenone concentrations. Immunocastrated and surgically castrated male pigs had nondetectable concentrations of fat androstenone and plasma testosterone and E1S, and the castration procedure induced a significant increase of 3betaHSD and 17betaHSD gene expression. The mRNA expression was generally much greater from the 3betaHSD than from the 17betaHSD gene. Furthermore, fat androstenone was negatively correlated with liver 3betaHSD gene expression (Pearson correlation, r = -0.69; P < 0.05), and the 17betaHSD gene expression in liver was negatively correlated with plasma E1S (r = -0.95; P < 0.001), indicating an important role of liver 17betaHSD in the estrogen metabolism of gonadally intact male pigs. Another strong correlation was found between 3betaHSD and 17betaHSD gene expression in liver of the gonadally intact male pigs (r = 0.86; P < 0.01), possibly reflecting similar regulation mechanisms of these genes.  相似文献

The present study was designed to evaluate the effects of synthetic ACTH (1-24, tetracosactid) and porcine CRH on the plasma levels of cortisol and PGF2alpha metabolite in cycling gilts (n = 3) and castrated boars (n = 3). The experiments were designed as crossover studies for each gender separately. Each animal received, during three consecutive days; 1) ACTH (Synacthen Depot) at a dose of 10 microg/kg body weight in 5 ml physiological saline, 2) porcine CRH at a dose 0.6 microg/kg body weight in 5 ml physiological saline or 3) physiological saline (5 ml). The test substances were administered via an indwelling jugular cannula in randomized order according to a Latin square. The administration of ACTH to cycling gilts resulted in concomitant elevations of cortisol and PGF2alpha metabolite with peak levels reached at 70.0 +/- 10.0 and 33.3 +/- 6.7 min, respectively. Similarly, the administration of ACTH to castrated boars resulted in concomitant elevation of cortisol and PGF2alpha metabolite with peak levels reached at 60.0 +/- 0.0 and 20.0 +/- 0.0 min, respectively. Cortisol peaked at 20 min after administration of CRH in both cycling gilts and castrated boars with maximum levels of 149.3 +/- 16.5 nmol/l and 138.3 +/- 10.1 nmol/l, respectively. It can be concluded that administration of synthetic ACTH (tetracosactid) to pigs caused a concomitant elevation of cortisol and PGF2alpha metabolite levels in both cycling gilts as well as castrated boars. The administration of CRH to pigs resulted in an elevation of cortisol levels in both cycling gilts and castrated boars. Conversely, PGF2alpha metabolite levels were not influenced by the administration of CRH either in cycling gilts or in castrated boars.  相似文献

The objective of this study, comprising two trials, was to evaluate the effect of a gonadotropin‐releasing hormone (GnRH‐vaccine ImprovacTM; Pfizer Ltd) in a sample of the Swedish pig population. The pigs (n = 120) were assigned to three groups: control (entire male pigs), surgical castration and immunization against GnRH. Surgically castrated pigs did not express detectable levels of either testosterone or estrone sulphate (E1S) in plasma, or androstenone in fat and had lower skatole and indole levels in fat than entire male pigs. Immunization significantly reduced testes weight and bulbourethral gland length, plasma levels of the testicular hormones testosterone and E1S, and fat levels of androstenone, skatole and indole. Skatole levels in plasma were significantly lower than in entire male pigs in the second trial, but not in the first due to overall low skatole levels. All immunized pigs and surgically castrated pigs expressed skatole concentrations in fat below the level of 0.2 μg/g, above which meat is regarded as tainted. In contrast, eight entire male pigs exceeded this level. Indole levels in plasma from immunized pigs were lower than those from entire male pigs. Surgical castration caused lower daily weight gain in the suckling period compared with piglets raised intact, whereas in the post‐weaning period no difference was observed. Immunization resulted in higher feed intake and daily weight gain after the second injection. The estimated lean meat content was improved in comparison with the castrated pigs, but was lower than for entire male pigs. Dressing percentage was lower in immunized pigs than in surgically castrated and entire male pigs. The frequency of skin damage did not differ between immunized and entire male pigs or between immunized and surgically castrated pigs.  相似文献

The studies aimed at identification of neoplastic cells at the S phase of mitotic cycle in mammary gland adenocarcinomas of bitches. The material was sampled from bitches of various races, aging 6 to 12 years, in which the mammary gland tumours developed spontaneously. The tumours were verified histopathologically and, then, immunohistochemical reactions were performed in order to detect cells which had incorporated BrdU (bromodeoxyuridine), contained Ki-67 or PCNA antigen. The histological preparations were photographed and obtained pictures were subjected to computer-assisted image analysis using Axiophot microscope (Carl Zeiss) coupled to a computer and the Multi-ScaneBase V 8.08 software, working under Windows. Fifty percent of sections from mammary gland adenocarcinomas demonstrated BrdU labelling index of 4-5%, 40% of 1-3%, while in the remaining 10% of examined tumours no BrdU incorporation could be demonstrated. No evident relationship could be detected between the presence of BrdU incorporation and Ki-67 or PCNA antigen presence but a significant correlation was demonstrated between the expression of Ki-67 and PCNA.  相似文献

Soya products containing phytooestrogens are widely used as feed for pigs. However, limited data are available on the effects of phytooestrogen on the endocrine status of pigs. The aim of this work was to study the impact of the phytooestrogen genistein added to a soya‐free diet on the hormonal pattern in gilts during oestrus and artificial insemination (AI). Ten gilts were fed a soya‐free diet and fitted with jugular vein catheter through vena auricularis. The gilts were randomly divided into two groups (G‐ and C‐group) where the G‐group was given pure genistein, 1 mg/kg body weight (BW) twice daily, per os. Blood samples were collected before, during and after AI. Oxytocin, prostaglandin E2, prostaglandin F2α, 13,14‐dihydro‐15‐keto‐prostaglandin F (PGFM), cortisol and LH concentrations in blood plasma were analysed. Oxytocin concentrations were almost twice as high in the G‐group as in C‐group after the AI. Prostaglandin E2 concentrations were higher in G‐group than in C‐group during the entire sampling period. After AI, the concentrations of prostaglandin E2 increased in G‐group but not in C‐group. Prostaglandin F concentration had a pulsatile pattern, with increasing pulses after AI in G‐group. Plasma PGFM concentrations increased after AI with a small variation between the groups. Plasma cortisol concentration increased after AI in C‐group. LH decreased after AI in G‐group. Genistein stimulated elevations of plasma oxytocin and prostaglandin E2 concentrations and a pulsative pattern in prostaglandin F concentration. The possible involvement of genistein in plasma cortisol and basal LH concentrations in gilts given genistein may also be suggested.  相似文献

Algae have an indisputable role in coastal ecosystems, but their accumulation and uncontrolled proliferation cause severe damage for the local municipalities. Fertilization with seaweed has been shown to increase soil fertility and crop production reducing ultimately the need for inorganic fertilizers. However, contradictory results of the compost effect have been reported. In the present work, we aimed at testing the suitability of three composted algae materials obtained in a previous study as soil amendments for vines. The composted materials consisted of pruning waste (P) and seaweed (S) mixed (henceforth, P2S1, P1S1 and P1S2, referring the number to the ratio of P to S). Overall, we observed an increase in soil organic matter, phosphorus (P) and potassium (K) in the soil treated in comparison with soils with inorganic fertilization. A moderate N soil enrichment (ca. 20%) was also detected. The leaf analysis reflected generally the greatest concentrations of NPK for the organic treatments, but this was remarkable only during the first year after seaweed application. A noticeable improvement in grape production was detected especially with the P1S1 compost without compromising grape quality although a decrease in sucrose content was noted with the compost with higher productivity (e.g. P2S1 and P1S1). This slight sucrose drop could be attributable to a greater water availability mediated by the compost or to a dilution factor of the sucrose content caused by a greater number of berries in those vines. These findings suggest that although monitoring of the long-term effects is needed, the use of seaweed amendments for agriculture could offer a cost-effective method for coastal municipalities to reduce excessive algae debris while also minimizing the impacts of inorganic fertilization.  相似文献
