Microbial biofertilizers are becoming an effective tool for sustainable agriculture by means of the reduction of the use of chemical fertilizers. However, the knowledge of each specific plant–microorganism interaction is essential for a correct application. In this study, we analyzed the in vitro plant‐growth‐promotion mechanisms of a Rhizobium leguminosarum strain named PEPV16 isolated from Phaseolus vulgaris nodules. This strain was able to produce siderophores and indole acetic acid and to solubilize phosphate. Confocal microscopy showed that this strain was able to colonize the roots of two horticultural crops, Lactuca sativa L. (lettuce) and Daucus carota L. (carrot). Strain PEPV16 was also able to promote the plant growth of both plant species increasing the dry matter of shoots and roots of lettuce and carrots, respectively, as well as to increase the uptake of N and P in the edible parts of both plant species. These data confirmed the suitability of Rhizobium as biofertilizer for nonlegumes.  相似文献

Tomato leaf curl New Delhi virus is an emerging whitefly-borne species of begomovirus in Mediterranean regions that poses a severe threat to cucurbit crops of the genus Cucurbita. Until now, only two sources of resistance have been identified in Cucurbita spp., these being PI604506 (cv. Large Cheese) and PI381814 (Indian landrace), both of C. moschata. The resistance of cv. Large Cheese is conferred by a single recessive gene located on chromosome 8. The objective of the present investigation was to screen for tomato leaf curl New Delhi virus (ToLCNDV) resistance among 105 accessions drawn from five species of Cucurbita and, if high resistance was found in any of them, determine the mode of inheritance. Screening was conducted using whitefly-mediated inoculation on all 105 accessions. The accessions showing some resistance were further screened by mechanical inoculation as well as by quantitative PCR-based diagnostics. The results showed that, overall, the accessions of C. pepo and C. maxima were the most susceptible, those of C. argyrosperma and C. ecuadorensis intermediate, and those of C. moschata most resistant to ToLCNDV. Only one accession of C. moschata, BSUAL-252, originating from Japan, was highly resistant to ToLCNDV, showing no symptoms after either method of inoculation, and absence of virus accumulation. Upon crossing BSUAL-252 with a susceptible accession of C. moschata, BSUAL-265, the resistance was observed to be conferred by a single dominant gene. This gene is not linked to the genomic region on chromosome 8 where the locus of the previously identified recessive gene for ToLCNDV resistance resides.  相似文献

Background

The Andes-Amazon basin of Peru and Bolivia is one of the most data-poor, biologically rich, and rapidly changing areas of the world. Conservation scientists agree that this area hosts extremely high endemism, perhaps the highest in the world, yet we know little about the geographic distributions of these species and ecosystems within country boundaries. To address this need, we have developed conservation data on endemic biodiversity (~800 species of birds, mammals, amphibians, and plants) and terrestrial ecological systems (~90; groups of vegetation communities resulting from the action of ecological processes, substrates, and/or environmental gradients) with which we conduct a fine scale conservation prioritization across the Amazon watershed of Peru and Bolivia. We modelled the geographic distributions of 435 endemic plants and all 347 endemic vertebrate species, from existing museum and herbaria specimens at a regional conservation practitioner's scale (1:250,000-1:1,000,000), based on the best available tools and geographic data. We mapped ecological systems, endemic species concentrations, and irreplaceable areas with respect to national level protected areas.

Results

We found that sizes of endemic species distributions ranged widely (< 20 km2 to > 200,000 km2) across the study area. Bird and mammal endemic species richness was greatest within a narrow 2500-3000 m elevation band along the length of the Andes Mountains. Endemic amphibian richness was highest at 1000-1500 m elevation and concentrated in the southern half of the study area. Geographical distribution of plant endemism was highly taxon-dependent. Irreplaceable areas, defined as locations with the highest number of species with narrow ranges, overlapped slightly with areas of high endemism, yet generally exhibited unique patterns across the study area by species group. We found that many endemic species and ecological systems are lacking national-level protection; a third of endemic species have distributions completely outside of national protected areas. Protected areas cover only 20% of areas of high endemism and 20% of irreplaceable areas. Almost 40% of the 91 ecological systems are in serious need of protection (= < 2% of their ranges protected).

Conclusions

We identify for the first time, areas of high endemic species concentrations and high irreplaceability that have only been roughly indicated in the past at the continental scale. We conclude that new complementary protected areas are needed to safeguard these endemics and ecosystems. An expansion in protected areas will be challenged by geographically isolated micro-endemics, varied endemic patterns among taxa, increasing deforestation, resource extraction, and changes in climate. Relying on pre-existing collections, publically accessible datasets and tools, this working framework is exportable to other regions plagued by incomplete conservation data.  相似文献

Purpose

The aim of this study was to evaluate in the medium term (5 years) the effect of two organic amendments, which were spiked to a degraded soil as a strategy for bioremediation, on the amount and characteristics of soil humic acids (HAs) and their ability to associate with certain extracellular enzymes.

Materials and methods

Soil samples were collected in an experimental field where 5 years earlier, a mixture of the organic fraction of household waste and sewage sludge (2:1 ratio), both composted (composted residue, CR) and non-composted (fresh residue, FR), had been added in triplicate at rates equivalent to 1 % (D1) and 3 % of organic carbon (D2) to 30-m2 plots as a strategy for degraded soil restoration. Humic substances (HSs) and HAs were extracted from the collected soil samples and submitted to chemical, biochemical, spectroscopic (FTIR), and chemical-structural (CPMAS 13C NMR) analyses.

Results and discussion

After 5 years, the amended soils showed significantly higher HS and HA content than did the control soil, and the differences with respect to the control were greater with compost addition than with FR addition. The HA from the amended soils had higher H, N, and S contents than the HA from the non-amended soil in addition to a lower oxygen content and lower O/C ratio values. Furthermore, the FTIR spectra of the HA from the amended soils showed a higher absorption intensity in bands corresponding to aliphatic and amide-carboxylic groups and polysaccharide structures and a lower absorption intensity in bands corresponding to carbonyls and carboxylic groups than the HA from the control. These results were confirmed by 13C-NMR spectra, which showed a clear increase of aliphatic compounds in the HA from the amended soils with respect to the HA from the control. HA spectra were not greatly influenced by the maturity of the amendment or by the application dose.

Conclusions

In general, the addition of organic amendments increased the quantity of enzymes immobilized in the humic colloid. Furthermore, the addition of the composted residues favored to a greater extent the immobilization of the abovementioned enzymes, which represent a biological reservoir in the soil. This is of great importance since these enzymes possess functional capacity even when the soils are under conditions that are stressful or unfavorable for microbial life. An increase in the quantity of immobilized enzymes such as that observed in amended soils supposes an important improvement in soil quality.

Ninety-nine isolates of Fusarium oxysporum f. sp. ciceris (Foc), representative of the two pathotypes (yellowing and wilt) and the eight races described (races 0, 1A, 1B/C, 2, 3, 4, 5, and 6), were used in this study. Sixty isolates were analyzed by the RAPD technique using DNA bulks for each race and 40 primers. Bands presumably specific for a DNA bulk were identified and this specificity was confirmed by further RAPD analysis of individual isolates in each DNA bulk. Primers OPI-09, OPI-18, OPF-06, OPF-10, and OPF-12 generated RAPD marker bands for races 0, 1B/C, 2, 3, 4, 5, and 6. The reliability and utility of this procedure was validated in blind trials using 39 new Foc isolates. Ten of the 39 isolates had already been typed to race by pathogenicity tests and 29 were typed both by pathogenicity and RAPD testing in this study. In these blind trials, we assigned the 39 new isolates to a race solely on the basis of their RAPD haplotype. Thus, we concluded that Foc races 0, 1B/C, 5, and 6 can be characterized by the RAPD markers. Cluster analysis of the RAPD data set resulted in three clusters of isolates within Foc. The yellowing isolates were grouped in two distinct clusters which correspond to races 0 and 1B/C. The wilt isolates constitute a third cluster that included races 1A, 2, 3, 4, 5, and 6. These results provide a means of studying the distribution of Foc races, to assist in the early detection of introduced race(s) and to facilitate the efficient deployment of available host resistance.  相似文献

The amino acid composition of 53 honey samples from Spain, consisting of 39 floral, 5 honeydew, and 9 blend honeys, has been determined. Physicochemical characteristics, polyphenolic content, amino acid composition, and estimation of the radical scavenging capacity against the stable free radical DPPH of the honey samples were analyzed. The resulting data have been statistically evaluated. The results showed that pH, acidity, net absorbance, electrical conductivity, and total polyphenolic contents of the honeys showed a strong correlation with the radical scavenging capacity. The correlation between the radical scavenging capacity of honey and amino acid contents was high with 18 of the 20 amino acids detected, with correlation values higher than those obtained for polyphenolic content. These results suggest that the amino acid composition of honey is an indicator of the sample's scavenging capacity.  相似文献
