The use of copper (Cu)-based fungicides to control diseases in vineyards can cause excessive Cu accumulation in soils. Greater Cu availability is expected in acid sandy soils with low organic matter (OM), where the risk of toxicity to plants and environment contamination is high. This study aimed to study the evolution of Cu in acid, sandy soils planted with grapevines for a period of time between 2 and 32 years. Two studies were carried out in South Brazil: in the first study, soil samples, from 19 vineyards were collected and analyzed in the first 0.2 m depth-layer for the total and available Cu. In the second study, 3 soils were selected among the previous 19 according to the age of vineyard (5, 11, and 30 year-old), and their 0.60 m-depth profile was analyzed for 5 fractions of Cu (soluble, exchangeable, mineral, organic, and residual). All the soils were classified as Ultisols. The vineyards with more than 25 years under cultivation had a mean total Cu concentration of 90 mg kg?1 in the 0–0.10 m layer. Approximately 80% of it was potentially available to plants. The greatest total and available Cu were found in the soil surface layers (0.2 m depth), where up to 75% of the total Cu is adsorbed in clay minerals and only 20% is complexed in the OM.  相似文献

Purpose

San Luis Potosí is one of the largest metal producers; mining activity has been responsible for metal emissions for over 100 years, from several sources (deposits, tailings, effluents, and dusts) generating effects in human and ecosystem health. The objective of this study was to evaluate the effect of the concentrations of heavy metals in the soil health of four municipalities of San Luis Potosí contaminated with mine tailings, using enzyme activity as a biochemical endpoint.

Materials and methods

Four municipalities contaminated with residues of historical mining activity were analyzed (25 topsoil samples per type of site contaminated and reference). The parameters that were analyzed included pH; organic matter (OM); electrical conductivity (EC); percentage of clay, As, Cd, Cr, Cu, Hg, Pb, and Zn; and arylsulfatase (ARS), β-glucosidase (BG), urease (UR), and fluorescein diacetate hydrolysis (FDA) activities in soil. Differences among the parameters per municipality and type of site were evaluated using a factorial analysis of variance. The relationships were analyzed by Pearson’s correlation and a stepwise distance-based linear model permutation test (DistLM). Results were visualized using a distance-based redundancy analysis (dbRDA). A hazard quotient (HQ) for metals was calculated in order to estimate the effects on soil microbial processes.

Results and discussion

A concentration gradient (mg/kg) of Zn (4744.5–65,585.7), Pb (1321.0–31,932.2), As (ND-8736.7), and Cu (130.9–8475.4) was found in the contaminated sites. The HQ showed a very high hazard level for the elements detected in all contaminated sites (1.4–655.8). The pattern of enzymatic inhibition found was ARS (95.8 %), UR (90.6 %), FDA (86.9 %), and BG (76.0 %). Strong negative relationships were observed among enzymatic activities and heavy metals in the following inhibitory effect Cu > As > Zn > Pb. Metals and covariables explained from 84 to 86 % of variability in enzyme activity. EC, Cu, and As showed a strong inhibitory effect; and parameters such as OM, pH, and clay were found to have a slightly inducing effect.

Conclusions

In this study, the heavy metal concentrations were higher than the ones obtained in other reports for this region. The HQ reveals the presence of possible risks for the health of life in the region. The decrease of enzyme activities in soil could trigger adverse changes in the flow of matter and energy in ecosystems. This study provides a field baseline that could be part of a long-term monitoring program for these locations.

Bluetongue virus (BTV) is a double-stranded RNA virus that induces apoptosis both in mammalian cell cultures and in target tissues. Based on information that members of the mitogen-activated protein kinase family (MAPKs) are mediators of apoptosis, we have examined in detail the MAPK-dependent apoptosis in BTV infection. Previously, we have shown that apoptosis in BTV infection requires the participation of mitochondrial apoptotic pathways. In addition, we demonstrated that NF-κB is activated and that its inhibition substantially reduces cellular apoptosis. For the first time, here we demonstrated the activation of MAPKs after BTV infection. Moreover, by pre-treatment with MAPK inhibitors, c-Jun N-terminal kinases (JNKs) and p38 MAPK, but not extracellular signal-related kinase (ERK), significantly decreased the induction of apoptosis. JNK and p38 activation regulated the cytochrome c released from mitochondria and caspase 3 activation. These results strengthen the understanding of BTV infection and contribute to our previous data confirming that BTV infection induces robust apoptosis in mammalian cells and is likely to play a primary role in BTV pathophysiology.  相似文献

Growth hormone (GH) is an important hypophyseal hormone that is primarily involved in body growth and metabolism. In mammals, control of Trypanosoma cruzi parasitism during the acute phase of infection is considered to be critically dependent on direct macrophage activation by cytokines. To explore the possibility that GH might be effective in the treatment of Chagas’ disease, we investigated its effects on the course of T. cruzi infection in rats, focusing our analyses on its influences on parasitemia, NO, TNF-α and IFN-γ concentration and on histopathological alterations and parasite burden in heart tissue. T. cruzi-infected male Wistar rats were intraperitoneally treated with 5 ng/10 g body weight/day of GH. Animals treated with GH showed a significant reduction in the number of blood trypomastigotes during the acute phase of infection compared with untreated animals (P < 0.05). For all experimental days (7, 14 and 21 post infection) of the acute phase, infected and GH treated animals reached higher concentrations of TNF-α, IFN-γ and nitric oxide as compared to untreated and infected counterparts (P < 0.05) Histopathological observations of heart tissue revealed that GH administration also resulted in fewer and smaller amastigote burdens, and less inflammatory infiltrate and tissue disorganization, indicating a reduced parasitism of this tissue. These results show that GH can be considered as an immunomodulator substance for controlling parasite replication and combined with the current drug used may represent in the future a new therapeutic tool to reduce the harmful effects of Chagas’ disease.  相似文献

This study was designed to evaluate the validity of PCR for the direct detection of Mycoplasma (M.) agalactiae and Mycoplasma mycoides subsp. capri (Mmc), as the two species most frequently causing contagious agalactia (CA) in goats. The PCR method was compared with the traditional culture technique to determine which method was most efficient at identifying all auricular carriers present in herds. The samples analyzed were 307 ear swabs taken from goats reared in a CA endemic area. We assessed the validity of each technique to detect each species and agreement between both methods. For each species, the result was taken as true-positive when at least one of the two tests was positive. Of the swabs tested, 246 were scored positive by PCR (235 and 11 for Mmc and M. agalactiae, respectively) and 117 showed a positive culture result (113 for Mmc and 4 for M. agalactiae). 133 of the PCR-positive samples (124 and 9 for Mmc and M. agalactiae, respectively) yielded negative culture results and 4 culture-positive samples tested negative using PCR (2 for each species). Sensitivity and negative predictive values for PCR were 84.62 and 99.32 (for M. agalactiae) and 99.16 and 97.22% (for Mmc) respectively, and for culture were 30.77 and 97.03 (for M. agalactiae) and 47.08 and 36.08% (for Mmc), respectively. PCR proved to be a rapid and sensitive method for the detection of mycoplasmas in the external ear of asymptomatic carriers. Tools such as this are needed to adopt efficient control measures against CA.  相似文献

The aim of this study was to validate 2 commercially available enzyme-linked immunosorbent assays (ELISAs) for adiponectin in dogs, 1 canine-specific and 1 originally designed for measurements in humans. Intra-assay and interassay precision was evaluated by multiple measurements in canine serum samples, and assay accuracy was indirectly determined by linearity under dilution. Interference caused by hemolysis and lipemia was also studied. Both assays were subsequently used for measuring adiponectin concentrations in clinically healthy dogs and those with different grades of obesity. The intra-assay and inter-assay precision was less than 7.5% and 13.5% in serum samples with low and high adiponectin concentrations, respectively. Lipemia and hemolysis did not affect the results of any of the assays. Both assays were able to differentiate lean dogs from those that were overweight or obese on the basis of the measured adiponectin concentrations. From these results it can be concluded that canine adiponectin concentrations can be measured reliably by means of the 2 ELISAs evaluated in this study.  相似文献

Six single‐flow continuous culture fermenters were used to determine fermentation profile, microbial growth and amino acid (AA) flow promoted by olive leaves supplemented with barley grains and faba beans (OLSUP), and alfalfa hay (AH). Two incubation runs were carried out with three fermenters inoculated with ruminal fluid from wethers and three from goats. The inoculum source did not affect (p = 0.059 to 0.980) any of the parameters. Daily volatile fatty acid (VFA) production and carbohydrate digestibility were greater (p = 0.009 and 0.024, respectively) for AH, therefore the pH values were lower (p = 0.015) than for OLSUP. Acetate was greater (p < 0.001) and isobutyrate, isovalerate and caproate lower (p < 0.001 to 0.006) for AH with greater acetate/propionate (p = 0.014) and ‘VFA/digested carbohydrate’ (p = 0.026) ratios. Daily microbial N flow and efficiency were greater (p = 0.016 and p = 0.041) for diet AH. Individual AA flows were greater (p < 0.001 to 0.016) for AH, but microbial essential AA proportion was greater for OLSUP (p = 0.015). The results indicate that OLSUP promoted lower bacterial growth and AA flow than AH, which could have been partially due to a limitation of N availability to ruminal microbes.  相似文献

Electrons in graphene behave like Dirac fermions, permitting phenomena from high-energy physics to be studied in a solid-state setting. A key question is whether or not these fermions are critically influenced by Coulomb correlations. We performed inelastic x-ray scattering experiments on crystals of graphite and applied reconstruction algorithms to image the dynamical screening of charge in a freestanding graphene sheet. We found that the polarizability of the Dirac fermions is amplified by excitonic effects, improving screening of interactions between quasiparticles. The strength of interactions is characterized by a scale-dependent, effective fine-structure constant, α(g)* (k,ω), the value of which approaches 0.14 ± 0.092 ~ 1/7 at low energy and large distances. This value is substantially smaller than the nominal α(g) = 2.2, suggesting that, on the whole, graphene is more weakly interacting than previously believed.  相似文献

Yeasts and organic minerals are used in diets to improve health, productive performance and some carcass characteristics of ruminants and non-ruminants. Thirty-two lambs(Pelibuey×Katahdin; BW=(30.55±1.67) kg; n=8) were used in a 56-d feeding experiment to study the effects of different levels of live yeast(Saccharomyces cerevisiae; yeast), selenium(Se) and chromium(Cr) mixed(Se-Cr), and a mixture of yeast-Se-Cr on growth performance and carcass traits. Animals were stratified by body weight(BW) and randomly assigned to one of four treatments: 1) control group(0.0 g kg–1 yeast); 2) yeast(1.50 g kg–1 dry matter intake(DMI) d–1); 3) Se-Cr premix(1.5 mg kg–1 DMI d–1 for each mineral); and 4) yeast-Se-Cr mixture. There were no treatment effects on final BW; whereas lambs fed Se-Cr or yeast-Se-Cr had higher(P0.05) DMI than animals supplemented with only yeast. Average daily gain(ADG), gain:feed ratio, chop area, dorsal fat and carcass yield were similar(P0.05) among treatment groups. In conclusion, supplementation with yeast, Se-Cr mixed or yeast-Se-Cr did not improve ADG, final BW, back fat content and carcass yield of growing of Pelibuey×Katahdin lambs. Supplementation with Se-Cr and yeast-Se-Cr increased DMI, and approximately 250 g ADG animal–1 d–1 was produced with no negative effects on growth and health of the animals.  相似文献
