To evaluate the relationship between the potassium (K) status in the microbial community and the exchangeable K concentration in soils, the effects of K addition on microbial activity were assessed in cultivated Andisols not having received K fertilizer. Potassium limitation was not observed in the microbial community, even in a soil amended with only nitrogen (N) and phosphorus (P) but not K since 1938, though crop plants in this soil showed severe K deficiency symptoms. Furthermore, in a soil amended with NP + compost, microbial activity was limited by K only after limitation of carbon (C) and N. These results suggest that soil microorganisms demand more C and N than K, even in soils with low K availability, and also that the soil microbial community is less susceptible to K deficiency than are crop plants.  相似文献

Reintroductions and translocations are increasingly used as conservation tools. However, despite guidelines recommending that health-screening be an integral part of such programmes, few reports are available. For most species, even baseline data, against which to compare future results, are not published. Here we report two case studies of health surveillance in wildlife reintroduction programmes - for water voles (Arvicola terrestris) in Australia, and for marsupial dibblers (Parantechinus apicalis) in the UK. These illustrate the potential importance of even basic screening strategies in helping to avoid disease transfer and identifying predictors of survival. Factors to be considered in the design of future monitoring schemes are also discussed.  相似文献

Studies on samples of 20 hard-grained wheat cultivars and a commercial flour that varied in starch and protein quality showed that both characteristics influenced the texture of Japanese alkaline noodles (ramen). Flour swelling volume (FSV) and flour pasting characteristics (peak viscosity and breakdown) determined with a Rapid-Visco Analyser (RVA) assessed independently of α-amylase effects, were negatively correlated with total texture score. Protein quality, as indicated by farinograph stability, was positively correlated with total texture score. RVA pasting characteristics were substantially affected by small levels of α-amylase, and inactivation by means of 1 mM AgNO3 was a critical requirement in characterizing the quality of the starch component of flour.  相似文献

The ICSI procedure is potentially of great value for felids, and it has not been extensively studied in these species. The objectives of this work were to determine the best conditions for ICSI in the domestic cat (DC) to generate interspecific embryos by injecting cheetah (Ch) and leopard (Leo) spermatozoa. Firstly, DC oocytes were matured with insulin–transferrin–selenium (ITS) or without it (MM) and cultured using atmospheric (21%) or low (5%) oxygen tension after ICSI. The group ITS‐5%O2 showed the highest blastocyst rate (p < 0.05), 20.9% vs 8.7%, 7% and 6.5%, for MM‐21%O2, MM‐5%O2 and ITS‐21%O2, respectively. The best conditions were used to generate the interspecific embryos, together with ionomycin activation (Io) after ICSI. Interspecific embryos resulted in high rates of blastocysts that were not positively affected by Io activation: 32.6% vs 21% for Ch and Ch‐Io, 9.8% vs 21% for Leo and Leo‐Io, and 20% vs 17.4% for DC and DC‐Io. We also evaluated DNA‐fragmented nuclei of experiment 1 and 2 blastocysts, using TUNEL assay. The fragmented nucleus proportion was higher in the ITS‐5%O2 group, 67.6%. Surprisingly, interspecific blastocysts showed the lowest fragmented nucleus proportion: 27% and 29.9% for Ch and Leo, respectively. We concluded that ITS and 5%O2 improve blastocyst formation in DC, although with a concomitant increase in DNA fragmentation. Most importantly, cheetah and leopard spermatozoa were able to generate blastocysts without artificial activation, which suggests that developmental capacity of wild felid spermatozoa can be evaluated by interspecific ICSI. Canine distemper virus (CDV) may induce multifocal demyelination in the central nervous system of infected dogs. The pathogenesis of this process is not clear. The present work identifies the presence of apoptotic cells in white and grey matter of dogs'cerebellum, naturally infected with CDV. Fifteen dogs with clinical signs of canine distemper that tested positive for CDV nucleoprotein were used. Brain specimens were processed and embedded in paraffin. Sections 5 microm thick were stained with hematoxylin-eosin and Shorr. Other sections were submitted to TUNEL reaction and to immunohistochemistry for CDV nucleoprotein detection. Acute and chronic demyelinated plaques were observed in the white matter, while apoptosis occurred particularly in the granular layer of grey matter. Apoptosis seems to play an important role in the pathogenesis of canine distemper demyelination.  相似文献

An epidemiological study was conducted in a highland rural community in Peru to determine risk factors for canine echinococcosis caused by Echinococcus granulosus. Dogs were diagnosed using a coproantigen enzyme-linked immunosorbent assay (ELISA). Dog owners were interviewed prior to stool collection and asked for attitudes, practices and beliefs likely to be associated with local patterns of E. granulosus transmission. Univariate and multivariate analysis were used to determine odds ratios (OR) and their 95% confidence intervals (CI). The main risk factors found to be significantly associated with canine echinococcosis by univariate analysis were dog age (3-25 months) (OR, 5.14; CI, 1.7-15.7), female sex (OR, 4.3; CI, 1.4-13.3) and having been fed hydatid infected offal (OR, 2.9; 95% CI, 1.0-8.6). There was complete lack of knowledge about echinococcosis transmission. In addition to periodic dog treatment, control programs need to emphasize education of the human population to increase knowledge of parasite transmission and to change human practices associated with high rates of infection.  相似文献

The industrial processing of precious stones is a source of revenue for several Brazilian towns, especially in the state of Rio Grande do Sul. Given the growing number of small-sized companies that process precious stones, wastewater production is inevitable and is a cause for concern inasmuch as preservation of nature is considered. The present study investigates the detoxification of the wastewater produced by the process of rhodamine B dyeing using oxidation processes. Ozonization (O3), ultraviolet irradiation (UV), and O3/UV methods were assessed. Some of the parameters used to measure the efficiency of the analyzed treatments included COD, ecotoxicity (Daphnia magna), cytotoxicity, and genotoxicity assays (Allium cepa assays). Results show predominance of negative and local environmental impacts, which are reversible in more than 70% of cases. The major proposed reversibility measures were the change in the process layout and dye wastewater segregation. Among the analyzed methods, ozonization proved to be more efficient in decolorization, with 60 min of treatment, pH = 9 and dosage of 5.705 mg O3/mg of rhodamine B. A pseudo first-order reaction, with a kinetic constant of 7.5 × 10−2 min−1, was observed. The cytotoxic and genotoxic effects were assessed for both raw and treated wastewaters. Despite complete decolorization, cytotoxicity and genotoxicity assays revealed an EC50 of 28.6, in addition to chromosome aberrations in 40% of dividing cells for the treated wastewater.  相似文献

We addressed the mechanistic basis for a negative correlation between soil pH and phosphomonoesterase activity, often found in various soils. Also in the present study, a significant negative correlation was observed between soil pH and phosphomonoesterase activity measured at pH 6.5 in Japanese acidic forest soils (3 Inceptisols, 3 non-allophanic Andisols, and 2 allophanic Andisols). A hypothesis that higher activity of phosphomonoesterase in acid forest soils results from increased synthesis of phosphomonoesterase by microorganisms in response to P limitation was tested. Soils with lower pH showed a lower optimum pH for phosphomonoesterase activity and greater activity at the optimum pH than other soils. To assess nutrient limitations of the soil microbial community, the effects of addition of C, N, or P on phosphomonoesterase and dehydrogenase activities, which is an intracellular enzyme and the activity of which reflects overall microbial activity, were examined in the soil samples. Addition of P increased dehydrogenase activity in some forest soils. Also, microorganisms in some soils were co-limited by C, N, and P. Response ratios (RR) of phosphomonoesterase and dehydrogenase activities in P-amended soil to their activity in non-amended soil were used to evaluate the response of soil microorganisms to P limitation. The ratio of RR-dehydrogenase to RR-phosphomonoesterase was strongly correlated with phosphomonoesterase activity at the optimum pH (P < 0.01). The results indicate that P limitation accounts for higher phosphomonoesterase activity in the more acid forest soils.  相似文献
