This study was designed to determine the susceptibility in vitro and infectivity of 1 field isolate of Mycobacterium avium sbsp paratuberculosis after exposure to monensin sodium and tilmicosin phosphate. Minimum inhibitory concentrations (0.39 microg monensin sodium/mL; 1.60 microg tilmicosin phosphate/mL) were determined in quintuplicate. Organisms were then incubated with 3 different concentrations of each medication for 3 different lengths of time, then washed and resuspended in sterile physiologic saline and injected intraperitoneally into mice that were genetically susceptible to infection. Mice were euthanatized 50 d later and the number of hepatic granulomas was used as the indicator of infectivity. Neither time of incubation nor concentration of medication had any effect on the infectivity of the organisms. Monensin sodium significantly reduced the number of hepatic granulomas in genetically susceptible mice while tilmicosin phosphate did not. Antimycobacterial activity of monensin sodium suggests that the role of monensin in the control of bovine paratuberculosis should be evaluated further. 相似文献

Precise quantification of nitrogen (N) mineralization in animal manure from different productivity-level management zones (MZs) could result in efficient and safe utilization of manure as N fertilizer. The objective of this study was to compare N mineralization rates of dairy cattle manure within and across three productivity-level MZs. To accomplish this, a 120 day laboratory incubation study was conducted at the Natural Resources Ecology Laboratory at Colorado State University using a completely randomized design set as a 2 factor factorial. Treatments for the study, which included the field equivalent of, 22, 44, 67, and 134 Mg ha?1 of applied animal manure, were replicated four times. Soils used in the study were collected from the top 15 cm of high, medium and low MZs from Fort Collins, Colorado, USA on a continuous corn (Zea mays L.) field. A significant difference (P ≤ 0.05) in mineralized N across MZs was found across manure rates. The high, medium and low MZ N mineralization rates were compared and there was no significant difference in cumulative N mineralized between MZs over time. The lack of statistical difference in mineralized N between MZs is hypothesized to have been influenced by the lack of substantial difference in soil pH and particle sizes (only 6% clay difference between MZs). That being the case, a study involving spatially variable soils with significantly different soil particle size between MZs may result in a different conclusion. The results of the study support the hypothesis that variable rates of manure mineralize differently within MZs; however, the results do not support the hypothesis that variable rates of manure in soil may mineralize differently across MZs. 相似文献

Changes in tree species distributions are a potential impact of climate change on forest ecosystems. The examination of tree species shifts in forests of the eastern United States largely has been limited to simulation activities due to a lack of consistent, long-term forest inventory datasets. The goal of this study was to compare current geographic distributions of tree seedlings (trees with a diameter at breast height ≤2.5 cm) with biomass (trees with a diameter at breast height > 2.5 cm) for sets of northern, southern, and general tree species in the eastern United States using a spatially balanced, region-wide forest inventory. Compared to mean latitude of tree biomass, mean latitude of seedlings was significantly farther north (>20 km) for the northern study species, while southern species had no shift, and general species demonstrated southern expansion. Density of seedlings relative to tree biomass of northern tree species was nearly 10 times higher in northern latitudes compared to southern latitudes. For forest inventory plots between 44° and 47° north latitude where southern tree species were identified, their biomass averaged 0.46 tonnes/ha while their seedling counts averaged 2600 ha−1. It is hypothesized that as northern and southern tree species together move northward due to greater regeneration success at higher latitudes, general species may fill their vacated niches in southern locations. The results of this study suggest that the process of northward tree migration in the eastern United States is currently underway with rates approaching 100 km/century for many species. 相似文献

The resorcylic acid lactones zearalenone ( 1), alpha-zearalenol ( 2), beta-zearalenol ( 3), alpha-zearalanol (zeranol) ( 4), beta-zearalanol (taleranol) ( 5), and zearalanone ( 6) were converted to their glucuronides on a preparative scale in good yields. Reactions were conducted with bovine uridine 5'-diphosphoglucuronyl transferase (UDPGT) as catalyst and uridine 5'-diphosphoglucuronic acid (UDPGA) as cofactor. The glucuronides were isolated by column chromatography and characterized by NMR spectroscopy and mass spectrometry. Although the principal products were 4- O-glucuronides (i.e., linkage through a phenolic hydroxyl), significant quantities of the 6'- O-glucuronides (i.e., linkage through the aliphatic hydroxyl) of alcohols 2, 4, and 5 were also isolated. In the case of 3, the 2- O-glucuronide was isolated as the minor product. Overall isolated yields of glucuronides, performed on a 20-50 mg scale, were typically ca. 80% based on the resorcylic acid lactone starting material. LC-UV-MS (2) analysis of purified specimens revealed MS (2) fragmentations useful for defining the point of attachment of the glucuronide moiety to the zearalenone nucleus. 相似文献

Management of rangelands has largely operated under the paradigm of minimizing spatially discrete disturbances, often under the objective of reducing inherent heterogeneity within managed ecosystems. This has led to a simplified understanding of rangelands and in many cases simplified rangelands. We argue that this type of management focus is incapable of maintaining biodiversity. An evolutionary model of disturbance (pyric-herbivory) suggests that grazing and fire interact through a series of feedbacks to cause a shifting mosaic of vegetation patterns across the landscape and has potential to serve as a model for management of grasslands with an evolutionary history of grazing. Our study demonstrates that the spatially controlled interaction of fire and grazing can be used to create heterogeneity in grassland ecosystems and the resulting heterogeneity in vegetation is expressed through other trophic levels, specifically small mammals in this study. Discrete fires were applied to patches, and patchy grazing by herbivores promoted a shifting vegetation mosaic across the landscape that created unique habitat structures for various small mammal species. Peromyscus maniculatus was about 10 times more abundant on recently burned patches (1–2 mo) than the uniform treatment or unburned patches within the shifting mosaic treatment. Chaetodipus hispidus was about 10 times greater in patches that were 15–20 mo post-fire in the shifting mosaic treatment than in the uniform treatment. Sigmodon hispidus, Microtus ochrogaster, and Reithrodontomys fluvescens became dominant in the shifting mosaic in patches that were more than 2 yr post-fire. This study, along with others, suggests that by managing transient focal patches, heterogeneity has the potential to be a new central paradigm for conservation of rangeland ecosystems and can enhance biological diversity and maintain livestock production across broad scales. 相似文献

A 20-year-old Thoroughbred mare was evaluated because of a 2-year history of infertility. The mare had normal estrous cycles and had been bred 7 times by different stallions. Ultrasonographic examination revealed a homogeneous hyperechoic intramural mass in the tip of the right uterine horn; the mass was also detected via hysteroscopy Unilateral ovariectomy and partial hysterectomy were performed by use of a hand-assisted laparoscopic technique. Leiomyoma was diagnosed via histologic examination of the mass. Unilateral ovariectomy and partial hysterectomy are recommended in mares with leiomyoma in a uterine horn, especially if the tumor is associated with infertility. The hand-assisted laparoscopic technique allows direct visualization of abdominal structures and accurate placement of ligatures without applying tension on the broad ligament, and eliminates the risks and costs of general anesthesia. 相似文献
