Genotyping of Turkish grapevine (Vitis vinifera L.) germplasm was characterized by use of six highly polymorphic microsatellite loci (VVS2, VVMD5, VVMD7, VVMD27, VrZAG62, VrZAG79). In this study we aimed to clarify the relationships between homonymous varieties coming from different regions. Our results showed a large degree of genetic variability among most of the homonymous cultivars. The number of alleles per locus ranged from 10 to 21, and gene diversity (expected heterozygosity) values ranged from 0.85 to 0.93. Cultivars presenting the same names of Sergi karas? (sampled from ?anl?urfa and Gaziantep), Yediveren (sampled from ?anl?urfa, Gaziantep, and National Germplasm Repository Vineyard in Tekirda?) and Serpenek?ran (sampled from ?anl?urfa and Gaziantep) were clustered together, or very close to each other, in a phenogram. Moreover, the alleles at the six microsatellite loci analyzed were found to be similar in terms of base pairs within each of these three closely positioned varieties. However, all the other cultivars failed to show a suitable clustering pattern when comparing their DNA profiles and names. Similarly named cultivars were not generally grouped together in the phenogram. On the other hand, we detected a tendency for differently named homonymous grape cultivars to cluster together.  相似文献

To investigate the changes in characteristics (CIE L*, a* and b*) of liver colour as well as growth, carcass and digestive tract of broilers as influenced by stage and schedule of feed restriction, 400 two‐week‐old broilers (Ross 308) were randomly assigned to 20 floor pens. Four replicate pens of 20 equally mixed sex birds per pen were randomly allotted each of the five treatments. Birds in control were fed ad libitum. Birds in QFRM and QFRL were restricted 75% of ad libitum intake (quantitative feed restriction, QFR) for 10 days. Birds in FTRM and FTRL were fed ad libitum for 7 days per a period of 10 days with third, fifth and seventh days of feed withdrawal (feeding time restriction, FTR). The restrictions in the QFRM and FTRM started on day 14 (at middle age, M) while the restrictions in the QFRL and FTRL started on day 31 (at late age, L). The restricted broilers consumed less feed than the full‐fed birds (p < 0.05). The QFRM and FTRM birds consumed less feed than QFRL and FTRL birds (p < 0.05). The body weight gain decreased by the QFRL and FTRL, the feed efficiency increased by the QFRM and FTRM compared to other treatments (p < 0.05). The QFRL and FTRL increased a* and b* values of liver and the relative weights of gut and liver, and the FTRL increased the L* value of liver compared to other treatments (p < 0.05). The QFRL and FTRL decreased the relative weight of abdominal fat compared to the control (p < 0.05). In conclusion, (i) the restricted feeding at middle stage improved feed efficiency; (ii) the restricted feeding at later stage decreased growth rate and abdominal fat; and (iii) limited‐time feeding during later period caused a measurable variation in the liver colour.  相似文献

Specific plants can remove salts from the soil and contribute to saline remediation in orchard intercropping. Determining the level of highest salinity that a salt-removing crop can withstand without reducing its yield is important for management. It is also important to know the critical hazardous level of saline irrigation water for the fruit trees.The objective of this study was to investigate the salt-removing capacity of purslane by studying different stress criteria and by tracking its salt removal from germination to harvest. Therefore, a pot experiment was performed by enhanced salinity levels.The results showed that purslane could cumulatively remove considerable amounts of salt from the soil if practical to cultivate as an intercrop all year round. In this regard, 6.5 dS m−1 can be concluded as the reasonable salinity level for the purslane managed to be intercropped in fruit orchards.  相似文献

In this study, the electron paramagnetic resonance (EPR) spin probe technique is applied as a detection method in the differentiation of irradiated and unirradiated wheat seeds. Two wheat cultivars, Kunduru and Bezostaya, were used. Aqueous solutions of 4-hydroxy-TEMPO (TANOL) spin probe prepared with a line-broadening agent, potassium ferricyanide, were used in all experiments. The EPR spectra of the samples were recorded against time. A decrease in the signal intensity and a change in the shape of the intensity-time curve (rehydration curve) were observed, depending on the applied level of irradiation. The ratio of the lipid and aqueous regions at the high field (mI = -1) line changes, depending on the dose of irradiation.  相似文献

This study was designed to investigate the effect of feeding on the plasma disposition of triclabendazole (TCBZ) in goats following oral administration. A total of eight goats, aged 14–16 months and weighing 20–30 kg were used in this study. The animals were allocated into two groups (fasted and fed groups) of four animals each. The goats in fed group were fed ad libitum but the animals in fasted group were not fed 24 h before and 6 h after drug administration. Commercial oral drench formulation of TCBZ (Endex-K, 5%) was administered orally to animals in two groups at dose of 10 mg/kg bodyweight. Heparinized blood samples were collected between 1 and 192 h after treatment and the plasma samples were analysed by high performance liquid chromatography (HPLC) for TCBZ, TCBZ sulphoxide (TCBZ–SO), and TCBZ sulphone (TCBZ–SO2). Relatively very low concentration of TCBZ parent drug was detected between 2 and 48 h, but TCBZ–SO and TCBZ–SO2 metabolites were present between 2 and 192 h in the plasma samples of fed and fasted animals. Fasting significantly enhanced the plasma concentration of TCBZ and its metabolites. The availability of TCBZ, TCBZ–SO and TCBZ–SO2 in the plasma samples of fasted goats were markedly greater compared to those of fed goats. It was concluded that fasting decreases the digesta flow rate and prolongs the retention of the drug into the gastrointestinal tract, resulting in enhanced quantitative gastrointestinal absorption or systemic availability of TCBZ and its metabolites in fasted goats.  相似文献

Preharvest bug damage to wheat can cause significant losses in bread‐making quality. One of the most prevalent forms of bug damage which frequently occurs in most countries of the Middle East, Eastern Europe and North Africa can be attributed to Heteropterous insects, particularly Eurygaster spp. Intercultivar differences in the susceptibility of glutenin to proteolytic degradation by the bug Eurygaster integriceps were investigated using six breadwheat cultivars of Turkish origin. Crude enzyme extract was prepared with distilled water from bug‐damaged wheat. The freeze‐dried extract was blended with sound samples of ground wheat, and the mixture was incubated in distilled water for 30 and 60 min at 37°C and subsequently freeze‐dried. The proteolytic effects of bug damage were determined on large polymeric glutenin. The latter was measured as 50% 1‐propanol insoluble (50PI) glutenin extractable with 50% 1‐propanol in reductant dithiothreitol. The decreases in the amount of 50PI glutenin and the high and low molecular weight subunits were quantified using reversed‐phase HPLC. There was a substantial and progressive decrease in the quantity of 50PI glutenin and its subunits with increasing incubation time. Intercultivar differences were observed that were unrelated to intrinsic levels of proteolytic activity. After 60 min of incubation, the relative decrease in 50PI glutenin compared with control samples ranged from 43% (cv. Ankara) to 65 % (cv. Kirkpinar). Some cultivars (Lancer, Ankara and Gün) with similar levels of intrinsic proteolytic activity showed significantly different responses to bug protease. One cultivar (cv. Kirkpinar) with the lowest proteolytic activity was the most susceptible. High quality breadwheats (cvs. Bezostaya, Lancer, Kiraç and Gün) were generally more resistant to the bug protease, although Ankara, with both intermediate protease activity and breadmaking quality, was the most resistant cultivar. While the 50PI glutenin test was very effective in quantifying the damaging effects of bug protease on wheat protein quality, the nature of the intercultivar differences was unclear.  相似文献

Economic losses due to suni‐bug (Eurygaster spp. and Aelia spp.) damage are important for the cereal industry in East European and Middle East countries. Samples of five durum wheat cultivars (Diyarbakir, Firat, Ege, Svevo, and Zenith) with zero, medium, and high levels of suni‐bug damage were used to determine the effects of suni‐bug damage on milling properties and semolina quality. As the damage level increased, semolina yields of all cultivars decreased significantly. The loss of semolina yield was greater than decreases in total yield of semolina plus flour, indicating that semolina yields were affected to a higher extent than were flour yields. The ash contents of the semolina samples increased significantly in all cultivars with increasing suni‐bug damage. The falling number values were not correlated with suni‐bug damage level and amylase activities of all samples were quite low. The pasting properties did not differ to a great extent depending on the suni‐bug damage level. Gluten quality of semolina samples substantially deteriorated as suni‐bug damage level increased, as determined by SDS‐sedimentation and mixograph analyses. It was concluded that suni‐bug damage would decrease profits of durum wheat millers substantially by affecting semolina yield and quality.  相似文献

Genotoxic effects of Fenaminosulf, fungicide and micro-biocide, were examined by using mitotic index (MI), mitotic phase, and Comet assay on the root meristem cells of Allium cepa. In the Allium root growth inhibition test, EC50 value was firstly determined as 25 ppm and, 12.5 (0.5 × EC50), 25 (EC50) and 50 (2 × EC50) ppm concentrations of Fenaminosulf were introduced to onion tuber roots. Distilled water was used as a negative control. All obtained data were subjected to statistical analyses by using SPSS 15.0 for Windows software. For comparison purposes, Duncan multiple range tests by using one-way analysis of variance (ANOVA) were employed and p < 0.05 was accepted as significant value. While MI (except in 24 h at 12.5 and 50 ppm) and prophase index increased, metaphase, anaphase and telophase indexes decreased in all concentrations compared to control at each exposure time. A significant increase in DNA damage was also observed at the concentration of 25 ppm in 24 h, 25 and 50 ppm in 96 h by Comet assay.  相似文献
