Endocrine-disrupting chemicals (EDCs) are widespread used and can interfere on hormone regulation with adverse consequences for both biota and human. Vitellogenin (vtg) is a yolk precursor protein synthesized by the liver in response to estrogen. In order to characterize the vtg of tropical fish Rhamdia quelen and establish a molecular biomarker, adult male individuals were exposed to 17-β-estradiol (E2) for vtg induction and anti-R. quelen vtg polyclonal antibodies production. Vitellogenic female fish were used as positive control group. E2-induced vtg was characterized as a glycolipophosphoprotein of high molecular mass with peptide mass fingerprint very similar in E2-exposed male and vitellogenic female fish. A polyclonal serum containing anti-R. quelen vtg antibodies was produced and showed high specificity and sensibility to detect the vtg of three fish species: R. quelen, Piaractus mesopotamicus and Prochilodus lineatus. Wildlife and laboratory studies reported that EDCs released into the environment may alter the levels of plasma vtg in male fish, making this protein a valuable biomarker of xenoestrogens exposure. Then, we propose the use of anti-R. quelen vtg as a tool for biomonitoring studies and water quality assessment in Brazil and South American countries where the three fish species occur.  相似文献

To evaluate a new technology - integrated weight longlines (IW) - as a viable seabird mitigation technology for demersal longline fisheries, we compared three experimental mitigation treatments, IW line alone, IW with paired streamer lines (IWPS) and unweighted longlines (UW) with PS (UWPS), to a control of no deterrent (UW alone). Trials took place on two vessels targeting Pacific cod (Gadus macrocephalus) over a five-month period in the Bering Sea, Alaska, USA. We used multiple criteria for evaluations - catch rates of all taxa, seabird abundance and attack rate, and gear sink rate and performance - making this study the largest and most comprehensive experiment of its kind.All mitigation technologies dramatically decreased seabird bycatch rates while having little to no effect on fish catch rates. Mitigation was more effective for surface foraging seabirds (Fulmarus glacialis and Larus spp.) than for diving seabirds (short-tailed shearwaters, Puffinus tenuirostris), reducing mortality rates by 91-100% and 80-97%, respectively. Based on multiple criteria, IWPS performed best, sinking gear within the protection of streamer lines eliminating surface forager catch completely and reducing shearwater catch by 97%, relative to the control. UWPS, status quo in the Alaska fishery, and IW alone performed similarly reducing surface forager catch rates by 98% and 91%, respectively, and shearwater rates by 87% and 80%, respectively. Seabird abundance and attack rate were poor proxies of seabird mortality, especially for IW gear. IW lines reduced the distance astern that birds have access to sinking baits by near half and its handling qualities proved superior to UW. We conclude that IW longlines and paired streamer lines are the core mitigation techniques and when deployed together, constitute the best management practice for seabird conservation in demersal longline fisheries using autoline systems.  相似文献

Soil‐frost events may influence the dynamics of fine roots and therefore affect root‐derived C fluxes to the soil. We studied the impact of soil frost on the fine‐root dynamics of Norway spruce (Picea abies [L.] Karst.) by a replicated snow‐removal experiment in a mature forest in SE Germany. Snow removal in the three treatment plots reduced soil temperature significantly with minima <–5.5°C in the O layer while the snow‐covered control plots never reached temperatures below the freezing point. Sequential soil coring in the O layer at the beginning and at the end of the soil‐frost period as well as after thawing revealed that the soil frost treatment increased fine‐root mortality by 29%. However, enhanced fine‐root production in the snow‐removal plots nearly compensated for the fine‐root losses caused by low temperatures. These findings were confirmed by minirhizotron observations in the O layer and the upper 25 cm of the mineral soil showing that relative fine‐root loss was by far higher in the snow‐removal plots than in the control plots. Compensatory fine‐root production in the snow‐removal plots exceeded fine‐root production in the control plots during a period of 8 weeks after the soil frost application by 39% in the O layer while it was similar in both plot types in the mineral soil. Sequential coring and minirhizotron observations led to substantially different fine root–longevity estimates for the soil frost period. However, in both cases, the snow‐removal treatment was characterized by a significant reduction in root longevity indicating a faster fine‐root turnover. As a consequence, experimental soil frost enhanced the C input to the soil via root death at our study site by approx. 42 g m–2 and stimulated the C investment towards the root system of the spruce trees due to a higher sink activity.  相似文献

OBJECTIVE: To determine the effects of treatment with and without adjuvant radiation therapy on recurrence of ocular and adnexal squamous cell carcinoma (SCC) at specific anatomic locations in horses. DESIGN: Retrospective study. ANIMALS: 91 horses. PROCEDURES: Medical records of horses with histologically confirmed ocular and adnexal SCC evaluated from 1985 to 2002 were reviewed. Sex, breed, age, type of treatment, location, and recurrence of SCC were recorded. Two treatment groups determined by recurrence of SCCs treated with and without adjuvant radiation therapy were established. RESULTS: The anatomic site with the highest recurrence rate was the limbus (junction of the cornea and sclera) or bulbar conjunctiva (477%), independent of treatment group. There was a significant difference in recurrence rates of ocular and adnexal SCCs between the 2 treatment groups, independent of anatomic location. Recurrence rates of SCCs treated with and without adjuvant radiation therapy were 11.9% and 44.1%, respectively. Recurrence rates for SCCs of the eyelid, limbus or bulbar conjunctiva, and cornea treated with adjuvant radiation therapy were significantly different from those for SCCs treated without adjuvant radiation therapy. The most frequently represented anatomic site for ocular and adnexal SCCs was the eyelid (28.7%). Coat color, breed, and the interaction of age and breed had a significant effect on tumor recurrence regardless of treatment type and anatomic location. CONCLUSIONS AND CLINICAL RELEVANCE: Results indicated that ocular and adnexal SCCs treated with adjuvant radiation therapy had a significantly lower recurrence rate, compared with SCCs treated without adjuvant radiation therapy, independent of anatomic location.  相似文献

Cultivation machinery applies large amounts of mechanical energy to the soil and often brings about a decrease in soil organic carbon (SOC). New experiments on the effects of mechanical energy inputs on soil respiration are reported and the results discussed. In the laboratory, a specific energy, K, of 150 J kg−1, similar to that experienced during typical cultivation operations, was applied to soil aggregates using a falling weight. Respiration (carbon dioxide, CO2 emission) of the samples was then measured by an electrical conductimetric method. Basal respiration (when K=0) measured on Chromic Luvisol aggregates, was found to increase with increasing SOC, from 1.88 μg CO2 g−1 h−1 for a permanent fallow soil (SOC=11 g kg−1) to 8.25 μg CO2 g−1 h−1 for a permanent grassland soil (SOC=32 g kg−1). Basal respiration of a Calcic Cambisol, more than doubled (2.0–5.2 μg CO2 g−1 h−1) with increasing gravimetric soil water contents. Mechanical energy inputs caused an initial burst of increased respiration, which lasted up to 4 h. Over the following 4–24 h period, arable soils with lower SOC contents, (11–21 g kg−1), respiration rates dropped back to a level, approximately 1.14 times higher than the basal value. However, grassland soils with higher SOC contents (28–32 g kg−1), increases in this longer-term respiration rate following 150 J kg−1 of energy, were negligible. A field experiment, in which CO2 was measured by infra-red absorption, also showed that tillage stimulated increased levels of soil respiration for periods ranging from 12 h to more than one week. The highest respiration rates, 80 mg CO2 m−2 h−1 were associated with high energy, powered tillage on clay soils. On the same soil, low energy draught tillage resulted in a respiration rate of approximately half this value. The results of these experiments are discussed in relation to equilibrium levels of soil organic matter. The application of known quantities of mechanical energy to soil aggregates under laboratory conditions, in order to simulate the effect of different cultivation practices, when combined with the subsequent measurement of soil respiration, can provide useful indication of the likely consequences of soil management on SOC.  相似文献

Today rapid survey methods of proximal soil sensing (PSS) provide an increasing number of different and highly resolved data. These multidimensional data sets can lead to multilayered and complex maps of parameters which are only indirectly related to soil properties and soil functions. However, in applications usually just one clear elementary map is required. It is of increasing importance to tackle this problem utilizing a cluster algorithm for the synthesis and reduction of multidimensional input variables. The cluster algorithm provides a partitioning of the investigated site whereby the units are characterized by the statistics of the PSS data. Therefore, the question that arises is how suitable is the suggested partitioning in terms of the delineation of different soil units. In this study, we investigate the suitability of cluster partitioning through a case study at a medium‐scale test site (≈ 50 000 m2). Two common PSS methods: electromagnetic induction (EMI) and gamma spectrometry (GS) will be employed to create a data set for partitioning by a K‐means cluster. The result of the cluster analysis is a delineation of three different parts. In contrast to previous studies, we evaluate the generated partitions by independent soil properties such as grain size, horizon thickness, and color of stratified randomly taken soil samples. The soil analyses show that one of three clusters significantly differs from the others in terms of grain‐size distribution and horizon thickness. The partitioning of the other two clusters could not be confirmed by the considered soil parameters. Nevertheless, the case study demonstrates the combination of different PSS data by K‐means clustering as a potential approach for site partitioning. An evaluation of the results of the cluster analysis through the collection and analysis of soil samples is highly recommended.  相似文献

A food-borne origin of the transmission of bovine spongiform encephalopathy (BSE) to cattle is commonly assumed. However, the fate of infectious prion protein during polygastric digestion remains unclear. It is unknown at present, whether infectious prion proteins, considered to be very stable, are degraded or inactivated by microbial processes in the gastrointestinal tract of cattle. In this study, rumen and colon contents from healthy cattle, taken immediately after slaughter, were used to assess the ability of these microbial consortia to degrade PrP(Sc). Therefore, the consortia were incubated with brain homogenates of scrapie (strain 263K) infected hamsters under physiological anaerobic conditions at 37 degrees C. Within 20 h, PrP(Sc) was digested both with ruminal and colonic microbiota up to immunochemically undetectable levels. Especially polymyxin resistant (mainly gram-positive) bacteria expressed PrP(Sc) degrading activity. These data demonstrate the ability of bovine gastrointestinal microbiota to degrade PrP(Sc) during digestion.  相似文献
