Cotton plants were genetically modified through the introduction of a synthetic gene that encodes a Bacillus thuringiensis insecticidal protoxin referred to as Cry1F(synpro). This protoxin is a chimeric, full-length delta-endotoxin of 130 kDa, comprised of the core toxin of Cry1Fa2 protein and parts of the nontoxic portions of Cry1Ca3 and Cry1Ab1 proteins, all of which originated from Bacillus thuringiensis. The Cry1F(synpro) expressed in cotton plants confers resistance to lepidopteran pests. The current study was conducted to characterize the Cry1F(synpro) protein expressed in the transgenic cotton event 281-24-236. Results showed that the full-length Cry1F(synpro) produced in the transgenic cotton plants was sensitive to the host cell protease cleavage, resulting in a truncated, biologically active form (core toxin) with an apparent molecular mass of 65 kDa. This truncated toxin was purified by immunoaffinity chromatography from the cotton leaf extract. N-terminal sequencing, peptide mass fingerprinting by MALDI-TOF MS, and internal peptide sequencing by MS/MS confirmed the identity of the truncated core toxin of Cry1F. The mechanism of truncation was explored with Cry1F(synpro) derived from a recombinant Pseudomonas fluorescens. The transgenic cotton-produced Cry1F showed equivalent insecticidal activity to that of Pseudomonas fluorescens-derived Cry1F.  相似文献

Live Mycoplasma gallisepticum (MG) vaccines have been USDA approved and licensed for use in commercial layer chickens since 1988; however, egg production and egg quality data exist only for the F strain of MG. Information pertinent to the effects of ts-11 MG on egg and eggshell quality parameters, as well as egg size distribution, is lacking. In this study, pullets were inoculated at 10 wk of age with ts-11 strain MG and placed in biological isolation units at 10 birds/unit. Hen-day egg production, eggshell strength, Haugh unit score, pimpling incidence, and blood/meat spot incidence were monitored and recorded in each trial through a 45-wk production cycle. Further, eggs from all treatments were collected daily, Monday-Thursday, and individually weighed. Results of this study indicate that no significant difference was observed between the treatments for the parameters measured or for egg size distribution. Therefore, these data should lessen producers' concerns pertaining to the impact of ts-11 strain MG on egg production, egg and eggshell quality parameters, and egg size distribution.  相似文献

The anorexic or dysphagic adult horse often requires nutritional support. Providing nutrients by the enteral route is the safest and most economic choice, but the dietary options available for use in horses are somewhat limited. The objective of this study was to compare the physiologic response of normal horses with a low-residue liquid or normal diet over a 10-day feeding period. Two groups of 6 normal adult horses were maintained on 1 of 2 diets for a 10-day period. Diets were formulated to meet the caloric needs of a horse maintained in a stall. The control group was fed 70% timothy hay and 30% textured concentrate for the test period, and the experimental group received the low-residue liquid diet, similar to liquid nutritional formulas designed for human use. Clinical parameters, body weight, packed cell volume, total plasma solids, blood glucose, and serum electrolytes were recorded daily for each horse during the dietary trial period. On days 1, 5, and 10 of the study, a complete blood count, serum biochemical profile, and urinalysis were performed.Horses' serum total bilirubin concentration and pattern of weight loss differed between groups. All other physical parameters, blood chemistry, complete blood count, and urinalysis results remained within the normal reference interval for the horses regardless of diet, although some statistical differences were observed. Horses returned to pasture and free-choice grass diet without complications at the end of the dietary trial period. These results demonstrate that few differences of biologic significance were observed between horses being fed low-residue diet and horses receiving a normal diet of hay and grain over a 10-day period. The objective of this study was to evaluate a convenient, low-cost modification to conventional transfer methods for Atlantic halibut juveniles. A series of wire mesh cages were stacked within transport tanks creating a stratified transport system (STS), increasing the surface area for settlement and facilitating a more homogeneous distribution of halibut throughout the tank compared with the conventional insulated box (Unstructured, UTS). A stochastic cost-benefit analysis determined investment into a STS to be cost-effective, generating a mean benefit-cost ratio of 1.31 (95% CI, 0.68?C2.00) after 2?years and a mean 5-year net present value of $85,176 (95% CI, $46,906?C$125,630). The implementation of a STS was found to be technically feasible and economically efficient method to improve Atlantic halibut transport.  相似文献

The mixing properties of the dough are critical in the production of bread and other food products derived from wheat. The high molecular weight glutenin subunits (HMW-GS) are major determinants of wheat dough processing qualities. The different alleles of the HMW-GS genes in hexaploid wheat vary in their effect on dough quality. To determine the contribution of the individual HMW-GS alleles, lines deficient in HMW-GS proteins were generated by chemical mutagenesis in the elite bread wheat Triticum aestivum cv. Summit. In this report we describe the identification and characterization of Dy10 and Ax1 deficient lines. Examination of the effect of Dy10 and Ax1 deficiency on dough rheological properties by mixography showed shorter mixing time to reach peak resistance, and weaker and less extensible doughs relative to the wild type control. This is the first time that the role of Dy10 in vivo has been examined apart from the Dx5 + Dy10 allelic pair combination.  相似文献

Relationships between lint yield and within-boll yield components are important for genetic improvement of lint yield in cotton (Gossypium hirsutum L.) cultivars. F2 plants derived from crosses between germplasm lines and high yielding cultivars were analyzed to determine the contributions of within-boll yield components to lint yield and to select parents with desirable combining ability for multiple within-boll yield components. Forty-five F2 hybrids were planted at two field sites in 2010 and 2011 with 4 and 3 replicates, respectively. There were a total of six yield components analyzed including lint percentage (LP), seed number per boll, lint weight per seed (LW_S), seed surface area per seed, lint weight per unit seed surface area (LW_SA), and lint number per unit seed surface area (LN_SA). The contributions of these yield components to lint yield were analyzed by commonality analysis that separated the contributions to lint yield into the unique contributions of single yield components and the common contributions of the single yield components with one or more other yield components. The unique contributions of the six yield components to lint yield ranged from 1.6 to 21 % of total variation for lint yield in the 2-year experiments. The greatest common contributions to lint yield among all combinations of the six yield components were identified for a combination of four components, LP, LW_S, LW_SA, and LN_SA with 67 and 44 % of the total variation of lint yield in 2010 and 2011, respectively. Results suggest that all four of these yield components should be considered simultaneously in breeding for genetic improvement of lint yield. The germplasm line SP225 was detected as a good combiner with positive general combining ability (GCA) for LP (1.4 %), LW_SA (0.03 mg mm?2), and LN_SA (14.3 no mm?2), and favorable GCA for fineness (?3.1 mg km?1).  相似文献
