A new bioactive macrolactone, nuiapolide (1) was identified from a marine cyanobacterium collected off the coast of Niihau, near Lehua Rock. The natural product exhibits anti-chemotactic activity at concentrations as low as 1.3 μM against Jurkat cells, cancerous T lymphocytes, and induces a G2/M phase cell cycle shift. Structural characterization of the natural product revealed the compound to be a 40-membered macrolactone with nine hydroxyl functional groups and a rare tert-butyl carbinol residue.  相似文献

Continuous application of organic fertilizers can cause accumulation of organic phosphorus(P) in soil, especially in the lowmolecular-weight organic phosphorus(LMWOP) forms. This organic P pool represents a potentially important source of P for both plants and microorganisms. To understand the effect of long-term fertilization(30 years)(P-rich soil) vs. fallowing(P-poor soil) on the bioavailability and fate of LMWOP in subtropical paddy soils, we determined the sorption and mineralization of 14 C-labeled adenosine, adenosine monophosphate(AMP), adenosine diphosphate(ADP), and adenosine triphosphate(ATP) in each soil. The contents of carbon, nitrogen, and P in the P-rich soil were more than two times greater than those in the P-poor soil. The mineralization rates of the LMWOP compounds were faster in the P-rich soil compared to the P-poor soil, and followed the order AMPADPATP. Using sterilized soil, all forms of adenosine-P were strongly sorbed to the solid phase and reached saturation in a short time, with the adsorbance increasing with the number of phosphate groups. We concluded that the mineralization of LMWOP compounds was repressed slightly by sorption to the solid phase, but only in the short term. Thus, LMWOP compounds serve as readily available sources of C for microorganisms, making P available for themselves as well as for the plants. However, P accumulation and the progressive saturation of the P sorption sites in highly fertile soils may increase the potential risk of P runoff.  相似文献

Four groups of cattle infested with Boophilus microplus (Canestrini) were each dipped in a different concentration of amitraz diluted from a 12.5% EC formulation to determine the efficacy and performance of the product in an 11 400 l dipping vat. Except for the period when heifers were dipped, animals were restrained in stanchions placed individually inside 3.3×3.3 m2 stalls within an open-sided barn. The amitraz in the vat was stabilized with hydrated lime to maintain a pH of ca. 12. Analyses of vat samples showed that concentrations of amitraz in the vat were 7.6 to 13% lower than the targeted concentrations of 0.010, 0.015, 0.020, and 0.025% active ingredient (AI) for dilutions prepared according to instructions on the manufacturer's label. The large quantity of hydrated lime added to the vat (10 kg/1000 l) interfered with the HPLC analysis of vat samples. Therapeutic efficacy of each of the four observed concentrations (0.0088, 0.0131, 0.0174, and 0.0231% AI) of amitraz was excellent (>99% control). However, the rapid detachment of all ticks from an animal within a few hours after treatment with amitraz, that has been frequently observed, was not pronounced in the present study. Only 47% of the B. microplus detached in the first 4 h post-treatment, and 84% detached within the first 24 h. All of the treatments, except the lowest concentration, provided protection of cattle against re-infestation by B. microplus larvae for 14 days post-treatment. Possibly as a result of the formation of a compact layer of lime and amitraz on the bottom after the vat was undisturbed for six weeks, intense agitation was required to re-suspend the active ingredient.  相似文献

An esterase which hydrolyzes the ester bond of trans-permethrin has been partially purified from homogenates of larvae of the cattle tick. The final (gel filtration) step showed two distinct peaks of p-nitrophenylbutyrate-hydrolyzing activity. The trans-permethrin-hydrolyzing activity of the lower-molecular-weight enzyme cochromatographed with the p-nitrophenylbutyrate-hydrolyzing activity of that enzyme. Little trans-permethrin hydrolysis was observed in the high-molecular-weight peak. The yield of the low-molecular-weight enzyme increased on extraction of the homogenates with Triton X-100. Inhibition studies using the low-molecular-weight enzyme and trans-permethrin as substrate indicated that hydrolysis was due largely to a carboxylesterase (EC 3.1.1.1).  相似文献

The final stages of cervical ripening and parturition resemble an inflammatory process. Although the role of cytokines in both spontaneous and experimentally induced parturitions has been described in several small laboratory animals and humans, the involvement of pro‐inflammatory and regulatory cytokines in physiologic parturition in cows has not been determined. In this study, the cytokine expression profiles were assessed in bovine cervical tissue at several stages of pregnancy and at parturition. Serial biopsy samples of the cervix were obtained from 10 cows on day 185 and day 275 of pregnancy (which was on average 5.4 days before parturition) and at parturition. Messenger RNA expression levels of interleukin (IL)‐1β, IL‐6, IL‐8, IL‐10 and tumour necrosis factor (TNF)α were determined using real‐time polymerase chain reaction and the number of neutrophils and eosinophils was estimated by Luna and Sirius Red staining. At parturition, IL‐8 expression had increased 430‐fold (p < 0.001) when compared with that of the day 185 of pregnancy, large numbers of neutrophils had invaded the cervix while eosinophils remained scarce, IL‐1β had increased eightfold (p < 0.05) and IL‐6 had not changed significantly. Additionally, IL‐10 was increased by 10‐fold (p < 0.001) and TNFα decreased by 57% (p < 0.05) when compared with that of the day 185 of pregnancy. The large increase in expression of IL‐8, enabling the influx of neutrophils, is indicative of its important role in the final stage of cervical ripening and at parturition. As previous studies have shown that neutrophils excrete matrix metalloproteinases (MMP), this might contribute to softening of the cervix. In contrast, the only slightly increased levels of IL‐1, steady concentrations of IL‐6 and decreased TNFα, the potential consequences of increased IL‐10 expression, indicate that final cervical of cows in ripening at term parturition is an inflammatory process influenced by regulatory cytokines.  相似文献

The reproductive fitness of Rhipicephalus (Boophilus) microplus (Canestrini) strains resistant to organophosphate (OP), pyrethroid (P), or formamidine (F) acaricides was compared to an acaricide-susceptible (SUS) strain to determine whether the acquisition of resistance affected reproductive fitness in the resistant strains. The SUS strain females had a 3.0 days preoviposition period, a 12.1 days oviposition period, a 22.5 days egg incubation period, a mean of 3670 eggs per female, and a mean percentage egg hatch of 78.1%, which were all remarkably similar to these same parameters reported for this species throughout the world. The reproductive biology of the P-resistant strain (PYR) and the F-resistant strain (FOR) were, for the most part, similar to those of the SUS strain. In the few instances where statistical differences did occur there was little evidence that the variation had any biological basis that could be attributed to a reduction in fitness related to resistance to P or F acaricides. Although the comparison of reproductive parameters of the OP-resistant strain (OPR) and the SUS strain identified statistical differences between the mean egg incubation and oviposition periods, the magnitude of the differences was not sufficient to conclude that the OPR strain was biologically less fit than the SUS strain. However, the OPR strain produced 30% fewer eggs (2562 eggs per female) than the SUS strain (3670 eggs per female) indicating the acquisition of resistance placed the OPR at a selective disadvantage relative to the SUS strain. This coupled with a lower, though non-significant, egg hatch was used to predict there would be a reduction of at least 34.1% in larval numbers available to potentially re-infest subsequent cattle than were available from the SUS strain. These data may aid the development of management strategies that can be used to control OP-resistant ticks.  相似文献
