Changes in semen quality and selected biochemical markers were analyzed during a week of spawning season of common carp Cyprinus carpio L. Semen was obtained twice, on May 30 and on June 7, and each time it was collected 24 h after hormonal stimulation using Ovopel [(d-Ala6, Pro9 NEt)-mGnRH + metoclopramide] in 1 pellet kg?1. The total volume of semen (ml), volume of semen per kg of body weight (ml kg?1 b.w.), sperm concentration (×109 ml?1), total number of sperm per kg of body weight (×109 kg?1 b.w.), pH of semen, pH of seminal plasma, seminal plasma osmotic pressure (mOsm kg?1) and the total protein content in seminal plasma (mg ml?1) were determined. A 10 mM Tris buffer containing 100 mM NaCl with 0.5 % BSA (pH 9.0, osmolality 200 mOsm kg?1) was used to activate sperm. The following computer-assisted sperm analysis (CASA) parameters were determined: percentage of motile sperm (MOT, %), progressively motile sperm (PRG, %), curvilinear velocity (VCL, μm s?1), straight-line velocity (VSL, μm s?1), movement linearity (LIN, %), wobbling index (WOB, %), amplitude of lateral head displacement (ALH, μm) and beat cross-frequency (BCF, Hz). The volume of semen per kg of BW, total number of sperm per kg of BW and semen pH were significantly lower at the second semen sampling compared to the first semen sampling. Volume of semen at the second sampling correlated positively with CASA parameters. A lack of differences among CASA parameters between both collection periods indicates good quality of carp sperm hormonally stimulated with Ovopel twice at a 1-week interval.  相似文献

Termite activities are known to significantly influence small-scale soil properties in tropical savannas. The lateral and vertical extent of the alterations to the nest's surrounding, and particularly resulting impacts on diagnostic soil horizons remain largely unresolved until today. We examined the effects of mound-building termites on soil genesis and constitutive chemical soil properties in and below their nests. Two transects to a soil depth of 100 cm were dug below three younger mounds of Cornitermes silvestrii (the primary nest builder), three older mounds in which C. silvestrii had died out and which were secondarily colonized mainly by Nasutitermes kemneri, and three reference sites in the Brazilian Cerrado. The samples were characterized by standard procedures for soil classification; in addition, phosphorus extractions were conducted on selected samples using NaHCO3 for labile P forms, and concentrated HCl for stable P forms. This data set was then used to build calibration models for the prediction of labile and stable inorganic (Pi) and organic (Po) P forms, as well as for contents of organic carbon (OC), for the remaining samples applying mid-infrared spectroscopy in combination with partial least squares regression (MIRS-PLSR). We can show that the termite influence on the soil was sufficiently large to change diagnostic characteristics of the soils under termite mounds. The MIRS-PLSR predictions were suitable for quantifying organic carbon and most of the labile and stable phosphorus fractions. They showed an enrichment of OC, NaHCO3-Po and NaHCO3-Pi contents in nests inhabited by primary and secondary termites by factors of 1.6–2.0 and 1.4–1.5, respectively. The soils surrounding the nests had higher contents of OC and NaHCO3-P under both nest types vertically down to 30 cm below the lower nest border, and OC and NaHCO3-Pi contents were elevated at minimum to a lateral distance of 60 cm away from the nest border. As the pattern of HClconc-Pi, which comprised 95% of total P, showed no variations, we conclude that the higher NaHCO3-Pi amount was formed in termite nests by changing the availability of the more stable HClconc-Pi. In contrast to the contents, the OC and NaHCO3-P stocks below the mounds inhabited by primary termites were comparable to those inhabited by secondary ones, because the bulk density of the secondarily inhabited nests was elevated. This was due to a transport of clay-rich material from the subsurface argic horizons into the nests. Here, the secondary termites even reverted the lessivation observed in the reference soils and under mounds inhabited by primary termites, thus causing the soil types to change from Alisols and Acrisols to the properties of Umbrisols.  相似文献

Within-field variability in wheat grain yield and its quality always exists in production fields and depends, among other factors, mainly on various soil properties related to nutrients and water availability. The aim of the research was to examine the relationships between selected soil properties such as texture; pH; content of the available phosphorus (P), potassium (K), magnesium (Mg), and organic carbon; and winter wheat grain yield and quality under rainfed conditions. Six crop fields with winter wheat (Triticum aestivum, L.) in three sites located in different regions of Poland were examined during two seasons. The grain yield was mainly determined by the soil texture, and the majority of the chemical soil properties did not have a significant effect on grain yield. The grain quality traits were determined by the examined soil properties to a smaller degree than grain yield. The relationships were not consistent across sites and years.  相似文献

Stimulation of macrophages by a variety fatty acids causes activation of MAP kinases (MAPKs). The consequences arising from down-regulation of MAPKs may be a limitation in the activity of PPARγ, which is modulated by a modification catalyzed by these kinases. Phosphorylation of MAP kinases-ERK1/2 and p38 as well as PPARγ was determined by real-time polymerase chain reaction and Western blotting in human macrophages cultured with conjugated linoleic acids (CLAs). We demonstrated that CLA isomers alter MAP kinase phosphorylation and PPARγ activation. Phosphorylation of ERK1/2 was diminished in cells cultivated with cis-9,trans-11 CLA, whereas phosphorylation of p38 was reduced by trans-10,cis-12 CLA. PPARγ was phosphorylated mainly by ERK1/2, and consequently, PPARγ phosphorylation was suppressed mainly by cis-9,trans-11 isomer. In human adipocytes, cis-9,trans-11 C 18:2 raised the activation of PPAR and several of its downstream target genes. We suggest that a similar process may also occur in human macrophages.  相似文献

Soil texture (ST) is relatively stable over time, although it may change due to erosion, clay eluviation, and other processes. Soil texture affects soil quality, productivity and management. Therefore, indirect, accurate methods for assessing of soil texture classes (STCs) are needed in agricultural practice. A study was performed on four production fields in northern and central Poland to compare the fitting performance of STC models based on apparent electrical conductivity (ECa), topographic properties (elevation, slope gradient and wetness index) and Amber NDVI measurements. One common and accurate indicator of STCs was not found for all study fields. On average, ECa was most accurate in indicating areas of different STCs within the fields, but it tended to overestimate the size of sandy areas on loamy fields and vice versa. The accuracy of STC assessment using ECa measurements may be biased due to imperfect soil drainage, high elevations, which increase evaporation and STC variation with depth. STC assessment using Amber NDVI measurements may be useful, particularly on flat and sandy fields, but the results are affected by the same factors as ECa, and additionally by crop growth stages and by the weather conditions in the period preceding the measurements. Despite the good quantitative results of the STC assessment by elevation (one field) and by the topographic wetness index (another field), both terrain attributes failed to accurately indicate the distribution of some STC areas within each field. Therefore, in landscapes developed from deposits of the last glaciation relevant ST differences might not sufficiently be detected by the analysis of terrain attributes alone. The selection of STC predictors and evaluation of the assessment quality must consider both the quantitative indicators such as correlation and determination coefficients describing relationships between ST and ECa, NDVI and topography and percentage of a field area with accurately indicated STC and the distribution of areas with different STCs within a field. The use of ECa, NDVI values, and topographic properties for STC assessment may be useful in reducing costs of soil sampling and analysis, but cannot replace it.  相似文献

Purpose

The Matylda catchment, in southern Poland, was polluted by the discharge of mine waters from a lead and zinc mine that inundated parts of a valley floor and caused the accumulation of metal-polluted sediments. After a partial reclamation of the mine site in the early 1980s, polluted sediments continue to accumulate on downstream floodplains and in fishponds. The aim of this study was to reconstruct the changes in metal dispersal during 100?years of mining and during the 40-year post-mining period and to propose a strategy for pollution mitigation in the area.

Materials and methods

Analyses of Cu, Cd, Pb, Zn, Mn, Ca, Mg and Fe concentrations, speciation of heavy metals and mineralogical analyses were undertaken on overbank sediment cores and in stream sediments. Concentrations of the same elements and macro-ions soluble in stream waters were also determined.

Results and discussion

Concentrations of Zn, Cd and Pb in the sediment profiles vary between 40,000 and 55,000, 300 and 600 and 30,000 and 50,000?mg?kg-1, respectively. Changes of metal concentrations and the stratigraphy of sediments from the floodplains, stream channels and fishponds suggest rapid changes of metal loads migrating downstream during both the mining and post-mining periods. Since the time of mine closure, fine-grained, mine-derived sediments (ca. 12?cm thick) have been the main source of pollution of post-mining sediments and surface waters. Closure of the mine was followed by a relatively short period of rapid redistribution of sediment-associated heavy metals in the stream channel. Since the 1980s, the floodplain and fishponds have received a constant supply of metals. It contrasts with the slow sediment accretion rate and a rapid decrease of metal concentrations in floodplain pools due to dilution by decomposed leaf litter. A fivefold increase of Cd content in waters over the 4.6?km reach of the Matylda stream indicates continuous leaching of this element from the contaminated valley floor.

Conclusions

Unsuccessful mine site rehabilitation is due to leaching of mine-originated sediments dispersed over the valley bottom. However, the rate of metal remobilization over the last 40?years is low because of the small thickness and widespread anoxic conditions that prevail within both recent and mine-originated sediments and the alkaline pH of stream water, which reduces metal mobility. Distribution of the contaminated layer over a large area of the valley bottom precludes cost-efficient catchment rehabilitation.  相似文献

Although prostaglandin (PG) F is considered as the principal luteolytic factor, its action on the bovine corpus luteum (CL) is mediated by other intraovarian factors. Among them, nitric oxide (NO) seems to play a mandatory role in luteolysis. In this article we review the background and current status of work on possible roles of NO in the CL function, based on available information and our own experimental data. NO is produced in all three main types of bovine CL cells: steroidogenic, endothelial and immune cells. PGF and some luteolytic cytokines (tumor necrosis factor, interferon) increase NO production and stimulate NO synthase expression in the bovine CL. NO inhibits progesterone production, stimulates the secretion of PGF and leukotriene C4, reduces the number of viable luteal cells and, finally, participates in functional luteolysis. NO induces the apoptotic death of CL cells by the modulation of bcl‐2 family gene expression and the stimulation of caspase‐3 gene expression and activity. However, this simple molecule shows both luteolytic and luteotropic actions during the estrous cycle in ruminants. The aim of this overview is to present and discuss the recent findings crucial for understanding NO role in the process of CL regression in cattle.  相似文献

We previously showed in in vivo and in vitro studies that nitric oxide (NO) is engaged in luteolysis in cattle. Nitric oxide produced locally in the bovine corpus luteum (CL) inhibits progesterone (P4) synthesis and is suggested to be a component of the luteolytic cascade induced by uterine prostaglandin (PG) F2alpha. In the present study, the molecular mechanisms of NO action during structural luteolysis were studied in cultured bovine luteal cells (Days 15-17 of the estrous cycle). The effects of the NO donor (NONOate; 10(-4)M) on DNA fragmentation, cell viability, P4 production and caspase-3 activity were compared with those of PGF2alpha (10(-6)M). Moreover, mobilization of intracellular calcium [Ca2+]i and gene expressions of Fas-L, Fas, bcl-2, bax, and caspase-3 in the cells were determined by semi-quantitative RT-PCR after NONOate treatment. Caspase-3 activity was examined calorimetrically. Contrary to PGF2alpha NONOate decreased cell viability. DNA fragmentation after NONOate treatment increased by more than with PGF22alpha. NONOate increased mobilization of [Ca2+]i in the cells. Although the NO donor did not affect Fas-L and bcl-2 gene expression, it stimulated Fas and bax mRNA and caspase-3 expression. The ratio of bcl-2 to bax mRNA level decreased in the cells treated with NONOate. Moreover, NONOate stimulated caspase-3 activity more effectively than PGF2alpha. The overall results suggest that NO is a luteolytic factor that plays a crucial role in regulation of the estrous cycle in structural luteolysis by inducing apoptosis of luteal cells in cattle.  相似文献
