trans-Resveratrol is a polyphenol found in blueberries, grapes, and wine with cancer chemopreventive properties. The low bioavailability of this compound enhances its concentration in the luminal content and becomes a potential chemopreventive agent against colon cancer. In the present study, the antiproliferative and pro-apoptotic effects on the human colorectal carcinoma HT-29 cells as well as the mechanisms underlying these effects were examined. Proliferation, cytotoxicity, and apoptosis were measured by fluorescence-based techniques. Studies of dose-dependent effects of trans-resveratrol showed antiproliferative activity with an EC 50 value of 78.9 +/- 5.4 microM. Caspase-3 was activated in a dose-dependent manner after incubation for 24 h giving an EC 50 value of 276.1 +/- 1.7 microM. Apoptosis was also confirmed with microscopic observation of changes in membrane permeability and detection of DNA fragmentation. The activity of trans-resveratrol on the mitochondria apoptosis pathway was evidenced by the production of superoxide anions in the mitochondria of cells undergoing apoptosis. In conclusion, trans-resveratrol inhibits cell proliferation without cytotoxicity and induces apoptosis in HT-29. Results of the present study provide evidence demonstrating the antitumor effect of trans-resveratrol via a ROS-dependent apoptosis pathway in colorectal carcinoma. 相似文献

Bedrock surfaces in the Ouachita Mountains, Arkansas, exposed by spillway construction and which had not previously been subjected to surface weathering environments, developed 15–20 cm thick soil covers in less than three decades. All open bedrock joints showed evidence of weathering and biological activity. Rock surfaces and fragments also showed evidence of significant weathering alteration. The results suggest a soil production function whereby weathering and increases in thickness are initially rapid. The rapid initial rate (5 to 10 mm year− 1) is facilitated by a weathering-favorable regional climate, local topography favoring moisture and sediment accumulation, and aggressive vegetation colonization. The ages of the trees on the bedrock benches suggests that a short period (< 10 years) of pedogenic site preparation is necessary before trees can become established. Initial chemical weathering within newly-exposed rock fractures in resistant sandstone strata and chemical weathering of weak shale layers, coupled with accumulation of organic and mineral debris in fractures and microtopographic depressions facilitates plant establishment, which accelerates local weathering rates. 相似文献

Peach lipid transfer protein (LTP) can cause severe allergic reactions to peach-allergic patients. It belongs to the nonspecific LTPs family, a class of proteins extremely resistant both to proteolytic digestion and to high temperatures. Food processing can either drop or increase the allergenicity, depending on the process and on the food. As far as peach-derived products (pulp, juice) are concerned, it has been previously shown how thermal treatment performed in an autoclave does not decrease LTP allergenicity. In this work, it was attempted to investigate whether sequential microwave and ultrasound processing could affect the allergenicity of peach juice. Incubation with specific anti-Pru p 3 serum showed how treating peach peel with microwave at 140 °C and with ultrasound does not eliminate Pru p 3 IgE binding properties. Nuisance odors generation from waste and wastewater treatment plants are a cause of public discomfort and complaints. This situation impairs the air quality and represents a growing social and public health problem, especially in developing countries. Several modeling approaches have been developed and successfully implemented in the frame of a wastewater treatment plant for both the biological treatment and physicochemical processes. The mathematical modeling of the odor generation process is still considered a quite complex issue, mainly due to the fact that olfactory nuisance can be caused by many different chemical compounds and the perception of odors is influenced by subjective thresholds. Moreover, the impact of odor sources on air quality is highly conditioned by complex atmospheric dispersion processes. Prioritizing new areas for conservation in the Rocky Mountains of North America is important because the current intensity and scale of human development poses an immediate threat to biodiversity. We identified priority areas for avian biodiversity within a 3200-km corridor from Yellowstone National Park in Wyoming, US to the Yukon in Canada (the Y2Y region). We applied the conservation planning tool, MARXAN, to summarize 21 avian values. MARXAN minimizes the area delineated, while simultaneously incorporating multiple criteria (species richness representation, spatial clustering) and biodiversity targets into a single mappable solution. We prioritized avian biodiversity ‘hotspots’ at continental and ecoprovincial scales based on: (1) avian species richness; and (2) habitat associations of 20 focal species. At the continental scale, the single best solution represented 19% of the Y2Y region; 29% of this solution overlapped with existing protected areas. In northern Y2Y, large contiguous areas with high avian value were concentrated on the western edge of the continental divide. In southern Y2Y, contiguous areas were smaller and more numerous than in the north. In contrast to the majority of studies prioritizing conservation areas, we explored the effect of varying the extent of the target region by analyzing data at the scale of the entire Y2Y region and for eight ecoprovinces separately. We found that (1) large contiguous patches characterized only three ecoprovinces, while for the remaining ecoprovinces, numerous single scattered habitat patches of varying sizes were required to meet conservation goals; and (2) generally, only a small percentage of sites was already protected within the existing protected areas network. Our results are important for conservation planners and resource managers in the Y2Y region for incorporating areas of high conservation value for birds at regional and ecoprovincial scales during conservation project design and adaptive planning. 相似文献

To define the best grazing management strategy, it is important to assess animals' responses to variations in the structure and composition of tropical forages. This 2-year study evaluated animal response to Marandu palisadegrass (Urochloa brizantha)-calopo (Calopogonium mucunoides) mixed pastures managed under rotational grazing. Treatments consisted of three defoliation frequencies defined by rest periods interrupted at 90% (90LI), 95% (95LI) and 100% (100LI) of photosynthetically active radiation interception (LI). The stubble post-grazing height target was 15 cm. Statistical difference was declared at p < .10. The 100LI had lowest crude protein (CP) and in vitro dry matter digestibility (IVDDM) for the Marandu palisadegrass (p = <.001 both) and calopo (p = .003 and p = .067, respectively). Also, the OM digestibility decreased 7.0% in the 100LI condition than 90LI and 95LI treatment (p = .005). There was no difference in forage, grass, and legume intakes between the treatments (p > .10). The 100LI treatment decreased CP intake from grass in 33.3% (p = <.001) compared to other treatments. Greatest production of microbial N (p = .093) occurred with the 90LI treatment. The CP/digestible organic matter (DOM) ratio, urinary N excretion and retained N were lowest in the 100LI treatment (p = <.001, p = .007 and p = .014, respectively). The recommendation for grazing between 90 and 95% of LI is recommended because of greater CP intake and N utilization for the animals and improved the nutritive value of Marandu palisadegrass and calopo mixed pastures. 相似文献

The use of forage legumes to contribute biologically fixed nitrogen (N) to pastures is an alternative to increase beef cattle production in tropical regions. The objective was to compare the impact of the introduction of a legume with that of N fertilizer application on forage and animal production in Brachiaria pastures. This two-year study assessed three pasture treatments: (1) mixed Marandu palisadegrass (Brachiaria brizantha [syn. Urochloa brizantha] cv. Marandu) and the legume “ovalifolium” (Desmodium ovalifolium) cv. Itabela (Mixed), (2) Marandu palisadegrass pastures with 150 kg N ha−1 (Fertilized), and (3) Marandu palisadegrass without N fertilizer (Unfertilized). Rotational stocking with a variable stocking rate was used with a target herbage allowance of 1.0 kg forage kg body weight−1. The pre-grazing green herbage mass was similar for Fertilized and Mixed pastures, with 54% and 63% more mass than Unfertilized pasture, respectively (p < .001). Cattle that grazed the fertilized pasture had the greatest average daily gain (ADG; p = .017). The stocking rate and liveweight gain per area were greatest for the Fertilized and Mixed pastures (p < .001 and p < .001, respectively). No differences between treatments were found for DM forage intake (p = .555). Organic matter digestibility was lowest (p < .001) for the Mixed pasture. The inclusion of the ovalifolium legume in the Marandu pasture had the same impact on beef cattle production as annual fertilization with 150 kg N ha−1. The effects of nitrogen (N) fertiliser on grain size and shape, starch and protein concentration, vitreosity, storage protein composition, and alcohol yield of two winter wheat varieties contrasting in endosperm texture were studied in a field trial in Herefordshire, UK in 2004. Averaged across varieties, the alcohol yield was 439 L/tonne for grain with a protein concentration of 11.5 g/100 g. The soft endosperm wheat variety Riband produced on average 7.7 L more alcohol per tonne of grain at a given protein concentration than the hard endosperm variety, Option. At the same time, N fertiliser was shown to have significant effects on alcohol production through its major influence on grain protein concentration. Averaged over both varieties, there was a reduction in alcohol yield of 5.7 L for each 10 kg increase in protein content per tonne of grain. The starch concentration of Riband was 2.9 g/100 g higher than Option at a given grain protein concentration, supporting its higher observed alcohol yields. A low conversion of starch to alcohol in this study (6.30 L/10 kg starch) compared to the theoretical value (6.61 L/10 kg starch) indicated that there is potential for improvement of this character. The traits relating to grain size and shape were principally influenced by genotype, and were not influenced by N fertiliser. Conversely, there were only minor genotypic effects on grain protein concentration and vitreosity. An important finding was that there were no interactions between variety and N treatment for any of the variables considered, indicating that the response of the two varieties to changes in applied N was the same, resulting in consistent differences in starch concentration and alcohol yield between genotypes at different levels of grain protein. An analysis of the composition of the wheat storage proteins by size-exclusion chromatography showed that the gliadins increased on average by 0.56 g per g increase in total grain protein and were quantitatively the major protein fraction, suggesting that selection for low gliadin content may be a desirable means by which to reduce grain protein, and thereby increase alcohol yield in wheat. The relationship between alcohol yield per unit area and applied N rate was described by a quadratic function and the maximum alcohol yield per unit area was ca. 3630 L/ha. Statistical analysis suggested that the economic optimum rate of N applied for grain yield was close to the optimum N rate for maximum alcohol productivity. 相似文献

Factors associated with the severity with which different challenge models (CMs) compromise growth performance in pigs were investigated using hierarchical clustering on principal components (HCPC) analysis. One hundred seventy-eight studies reporting growth performance variables (average daily gain [ADG], average daily feed intake [ADFI], gain:feed [GF], and final body weight [FBW]) of a Control (Ct) vs. a Challenged (Ch) group of pigs using different CMs (enteric [ENT], environmental [ENV], lipopolysaccharide [LPS], respiratory [RES], or sanitary condition [SAN] challenges) were included. Studies were grouped by similarity in performance in three clusters (C1, C2, and C3) by HCPC. The effects of CM, cluster, and sex (males [M], females [F], mixed [Mi]) were investigated. Linear (LRP) and quadratic (QRP) response plateau models were fitted to assess the interrelationships between the change in ADG (∆ADG) and ADFI (∆ADFI) and the duration of challenge. All variables increased from C1 through C3, except for GF, which decreased (P < 0.05). LPS was more detrimental to ADG than ENV, RES, and SAN models (P < 0.05). Furthermore, LPS also lowered GF more than all the other CMs (P < 0.05). The ∆ADG independent of ∆ADFI was significant in LPS and SAN (P < 0.05), showed a trend toward the significance in ENT and RES (P < 0.10), and was not significant in ENV (P > 0.10), while the ∆ADG dependent on ∆ADFI was significant in ENT, ENV, and LPS only (P < 0.05). The critical value of ∆ADFI influencing the ∆ADG was significant in pigs belonging to C1 (P < 0.05) but not C2 or C3 (P > 0.10). The ∆ADG independent of duration post-Ch (irreparable portion of growth) was significant in C1 and C2 pigs, whereas the ∆ADFI independent of duration post-Ch (irreparable portion of feed intake) was significant in C1 pigs only (P < 0.05). Moreover, the time for recovery of ADG and ADFI after Ch was significant in pigs belonging to C1 and C2 (P < 0.05). Control F showed reduced ADG compared with Ct-M, and Ch-F showed reduced ADFI compared with Ch-M (P < 0.05). Moreover, the irreparable portion of ΔADG was 4.8 higher in F (−187.7; P < 0.05) compared with M (−39.1; P < 0.05). There are significant differences in growth performance response to CM based on cluster and sex. Furthermore, bacterial lipopolysaccharide appears to be an appropriate noninfectious model for immune stimulation and growth impairment in pigs. 相似文献
