BACKGROUND: Fifteen novel derivatives of D‐DIBOA, including aromatic ring modifications and the addition of side chains in positions C‐2 and N‐4, had previously been synthesised and their phytotoxicity on standard target species (STS) evaluated. This strategy combined steric, electronic, solubility and lipophilicity requirements to achieve the maximum phytotoxic activity. An evaluation of the bioactivity of these compounds on the systems Oryza sativa–Echinochloa crus‐galli and Triticum aestivum–Avena fatua is reported here. RESULTS: All compounds showed inhibition profiles on the two species Echinochloa crus‐galli (L.) Beauv. and Avena fatua L. The most marked effects were caused by 6F‐4Pr‐D‐DIBOA, 6F‐4Val‐D‐DIBOA, 6Cl‐4Pr‐D‐DIBOA and 6Cl‐4Val‐D‐DIBOA. The IC50 values for the systems Echinochloa crus‐galli–Oryza sativa and Avena fatua–Triticum aestivum for all compounds were compared. The compound that showed the greatest selectivity for the system Echinochloa crus‐galli–Oryza sativa was 8Cl‐4Pr‐D‐DIBOA, which was 15 times more selective than the commercial herbicide propanil (Cotanil‐35). With regard to the system Avena fatua–Triticum aestivum, the compounds that showed the highest selectivities were 8Cl‐4Val‐D‐DIBOA and 6F‐4Pr‐D‐DIBOA. The results obtained for 6F‐4Pr‐D‐DIBOA are of great interest because of the high phytotoxicity to Avena fatua (IC50 = 6 µM , r2 = 0.9616). CONCLUSION: The in vitro phytotoxicity profiles and selectivities shown by the compounds described here make them candidates for higher‐level studies. 8Cl‐4Pr‐D‐DIBOA for the system Echinochloa crus‐galli‐Oryza sativa and 6F‐4Pr‐D‐DIBOA for Avena fatua‐Triticum aestivum were the most interesting compounds. In Ethiopia, inoculation of soybean with rhizobial inoculants is not common practice, but could provide an option to increase grain yields in low nitrogen (N) acidic soils. In these acid soils, the selection of acid tolerant rhizobia is one strategy that may increase the performance of soybean. In this study, rhizobial strains isolated from Ethiopian soils were evaluated for their acid tolerance and symbiotic N fixation efficiency with soybean, in controlled environments. Following this, four isolated rhizobial strains were evaluated in six field experiments in major soybean growing areas of Ethiopia. Inoculation with the commercial strain or with one of two locally sourced isolates, that were developed as inoculants, improved soybean yield. The yield increase due to inoculation with the commercial strain was consistent and greater than other treatments, while the increase due to the two locally sourced strains was comparable to, or greater than, application of 46 kg N/ha in soils, where the resident rhizobial population was ≤1.4 × 103 cfu/g soil. For soils with high background rhizobial populations, there was no response to inoculation. In one of the experimental sites (Bako), the percentage of N fixed (%Ndfa) was 55 for the commercial strain and 35 for the local strain, ES3. This study demonstrated that field validation is a necessary step in the selection of acid-tolerant strains of rhizobia to increase soybean production for Ethiopia. 相似文献

Microbes have developed high-affinity uptake mechanisms to assimilate iron (Fe) and other metals such as aluminum (Al), gallium (Ga), chromium (Cr), and copper (Cu). Siderophores, which are metal chelating compounds, and membrane receptor proteins are involved in these specialized mechanisms. A few siderophore-producing microorganisms associated with plant roots also influence the uptake of some metals. In this study, the potential microbial-assisted Cu and Fe uptake by Phaseolus vulgaris (common bean) plants was evaluated. Seedlings of cultivated common bean varieties Bayo-INIFAP (B) and Negro-150 (N) and wild types yellowish (WY) and black (WB) were developed in the presence of a Cu and Fe solution and associated with the siderophore-producing microorganisms R. leguminosarumbv. Phaseoli (strains 19, 44, and 46); Pseudomonas fluorescens(strain Avm), and Azospirillum brasilense (strain 154). Seedlings of cultivated variety N and black wild type WB inoculated with the strain CPMex.44 accumulated 71% and 30% more Fe than the un-inoculated plants, respectively; however, the wild black bean accumulated the highest absolute amount of Fe (221.56 mg/kg of dry matter) as compared with the cultivated black variety N (126.16 mg/kg of dry matter) (P < 0.05). In the wild type WY seedlings, the highest Fe accumulation was observed when the seeds were inoculated with the Pseudomonas strain Avm (206 mg/kg of dry matter) (P < 0.05). The interaction of Pseudomonas strain Avm with seedlings of the cultivated B variety and the wild type WB promoted the highest accumulation of Cu (51 and 54 mg/kg of dry matter, respectively), 7 and 14 mg more than in the respective non-inoculated seedlings. No promotion of Fe accumulation was observed in the seedlings of the cultivated B variety and in roots; instead, less Fe was accumulated. The wild type WY did not show any improvement in Cu accumulation. In this study, Rhizobiumstrains promoted Fe but not Cu uptake in P. vulgaris seedlings while Pseudomonas strains promoted the uptake of both Cu and Fe. 相似文献

Chestnut (Castanea sativa) is an important basic food in rural diets and a major starch crop used in a similar way to potatoes. Chestnuts are a fundamental economic resource in the "chestnut regions" not only for the fruit but also for the chestnut wood. Chestnuts have become increasingly important with respect to human health, for example, as an alternative gluten-free flour source. Chestnuts are also a rich source of other beneficial compounds, but there have been few studies on the composition during processing. In this study, we analyzed the chemical composition of three Portuguese cultivars at different stages of industrial processing. The chestnut cultivars were Longal, Judia, and Martaínha. All three cultivars had high moisture contents but were low in ash, crude fat, and crude protein contents, with high starch and low fiber contents. The free amino acid contents, including various essential amino acids, varied depending on the cultivar. All three cultivars also had a significant content of polyphenolics with gallic acid; ellagic acid was predominant among hydrolyzable and condensed tannins. Many of these compounds are known to exert significant positive effects on human health. The one-way analysis of variance for fresh chestnut shows significant differences among the three cultivars for most of the studied parameters. The same statistical analysis applied to each one of the two cultivars (Judia and Longal) sampled for the four processing steps analyzed indicates a significant effect of this factor in practically all of the constituents. On the other hand, the two-way analysis of variance shows that, besides the residual, the processing step and the interaction cultivar x processing step were the factors that more contributed for the total variation observed in the constituents analyzed, while the contribution of cultivar was much less significant. 相似文献

Enterobacter cloacae is a plant-beneficial bacterium that shows promise for suppression of damping-off of cucumber and other crops caused by Pythium ultimum. We have been using a mutational approach to determine the E. cloacae genes important in bacterial-plant and bacterial-pathogen interactions in the spermosphere and rhizosphere. E. cloacae M43 is a transposon mutant of E. cloacae 501R3 that was significantly impaired in colonization of seeds and roots of diverse crop plants. Strain M43 did not increase in population on cucumber, sunflower, and wheat seeds and was significantly reduced in growth on pea seeds relative to strain 501R3. Populations of M43 were also dramatically lower than those of strain 501R3 in cucumber, pea, sunflower, and wheat rhizosphere in 42 d experiments. Molecular characterization of M43 demonstrated that there was a single transposon insertion in the genome of this strain and that this insertion was in a region of the E. cloacae genome with a high degree of DNA sequence identity with aceF. aceF encodes the dihydrolipoamide acetyltransferase subunit of the pyruvate dehydrogenase complex (PDHC). Cell lysates from strain 501R3 grown on minimal medium plus 50 mM glycerol and 2 mM acetate contained 0.011±0.0036 U pyruvate dehydrogenase activity while cell lysates from M43 grown under identical conditions contained no detectable pyruvate dehydrogenase activity. Additionally, the nutritional use profile of M43 under aerobic and anaerobic conditions was as expected for an ace mutant. Experiments reported here strongly suggest a role for aceF and the PDHC in colonization of seeds and roots of diverse crop plants by E. cloacae. 相似文献

The effect of different conditions of pea germination on dietary fiber (DF) composition was studied. Insoluble dietary fiber (IDF) and soluble dietary fiber (SDF) were subjected to acid hydrolysis, and the resultant neutral sugars, uronic acids, and Klason lignin were quantified. Germinated peas exhibited significantly higher contents of total dietary fiber (TDF) than the raw sample, due to the increases of both DF fractions. Under darkness conditions, germination exhibited the highest contents of IDF and SDF. Decreasing IDF/SDF ratios showed that the carbohydrate changes did not take place to the same extent during germination, the SDF fraction being the most affected. The detailed chemical composition of fiber fractions reveals increases of cellulose in the IDF of germinated samples, whereas SDF exhibits a decrease of pectic polysaccharides and also increases of polysaccharides rich in glucose and mannose. The DF results were corroborated by a comparative examination of the cell wall carbohydrate composition. 相似文献

This study describes the purification of an N-acetylglucosamine-binding lectin from Koelreuteria paniculata seeds and its effects on the larval development of Callobruchus maculatus and Anagasta kuehniella. The lectin (KpLec) was characterized and isolated by gel filtration, affinity column, and reverse phase chromatography. SDS-PAGE indicated that this lectin is a dimer composed of subunits of 22 and 44 kDa. The N terminus exhibited 40% similarity with Urtiga dioica agglutinin. KpLec was tested for anti-insect activity against C. maculatus and A. kuehniella. With regard to C. maculatus, an artificial diet containing 0.7 and 1% KpLec produced LD(50) and ED(50) value, respectively. However, for A. kuenhiella, an artificial diet containing 0.65% KpLec produced an LD(50), whereas 0.2% KpLec produced an ED(50). The transformation of genes coding for this lectin could be useful in the development of insect resistance in important agricultural crops. 相似文献

A study was conducted to measure residues of 13 organophosphorus (OP) pesticides, widely used as dairy cattle ectoparasiticides or in crops used for animal feed, in homogenized and pasteurized Mexican milk samples. Four different milk brands with high distribution were collected biweekly during a 12 month period (n = 96) in supermarkets. OP pesticide residues were measured by gas chromatography with a flame photometric detector. Approximately 39.6% of the samples contained detectable levels of OP pesticide residues. Eight samples contained residues exceeding established maximum residue limits (MRL), and the OP pesticides present in these samples were dichlorvos (five samples), phorate, chlorpyrifos, and chlorfenvinphos (one sample, respectively). Average residues of 13 OP pesticides measured were below established MRLs ranging between 0.0051 and 0.0203 ppm. 相似文献

The present study examines the ability of isoflavone extracts from whole soy bread and two soy bread fractions, crumb and crust, to modulate the proliferation of human prostate cancer PC-3 cells. Total isoflavone content in the two fractions of soy bread were similar (3.17 micromol/g dry basis). However, their conjugate patterns were altered. Both fractions of soy bread contained a similar level of isoflavone aglycones ( approximately 24%). Low concentrations of soy bread extracts increased PC-3 cell proliferation as much as 47% compared to untreated control. This proliferative effect in cell growth was reduced at higher extract concentration. Soy bread crust extract (10 mg/mL) reduced PC-3 cell proliferation by 15% compared to untreated control. Interestingly, wheat bread extracts increased cell proliferation at all concentrations tested. Although extracts from both breads possessed biological activity, only soy bread crust extract reduced PC-3 cell proliferation.
