Porcine alveolar macrophages were found to be highly susceptible to the cytolytic effects of a toxin (Shiga toxin [Stx]) produced by certain strains of Escherichia coli and sometimes associated with clinical disease in pigs and other animals. In comparison with the cells that are most commonly used for Stx detection and titration in vitro (namely, Vero cells), porcine alveolar macrophages appeared to be generally more sensitive and test results could be obtained in less time. Moreover, unlike Vero cells, porcine alveolar macrophages need not be continuously propagated to ensure immediate availability. They can simply be removed from a low-temperature repository, thawed, seeded, and shortly thereafter exposed to the sample in question. These characteristics suggest that porcine alveolar macrophages may be useful in developing a highly sensitive and timely diagnostic test for Stx.  相似文献

A study was conducted to determine the effect of two levels of linoleic acid (LA) intake at either high or low α‐linolenic acid (ALA) intake on their conversion and subsequent deposition into long‐chain (20–22 C‐atoms) polyunsaturated fatty acids (LC PUFA) in muscle and backfat in growing pigs. In a 2 × 2 factorial arrangement, 32 gilts from 8 litters were assigned to one of four dietary treatments, varying in LA and ALA intakes. Low ALA and LA intakes were 0.15 and 1.31 g/(kg BW0.75/day), respectively, and high ALA and LA intakes were 1.48 and 2.65 g/(kg BW0.75/day) respectively. There was a close positive relation between intake of ALA and the concentration of ALA in backfat and in intramuscular fat. Dietary ALA did not affect the concentration of eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA), but increased docosapentaenoic acid (DPA) in backfat. High ALA intake did not significantly affect DHA but significantly increased EPA, 20:3 n‐3 and DPA concentrations in intramuscular fat. The n‐3 LC PUFA proportion in backfat was increased from approximately 1–3%, which may be useful to enrich meat with these fatty acids. The effect of ALA intake on n‐3 LC PUFA was suppressed by LA intake. Dietary ALA suppressed the concentration of n‐6 LC PUFA in blood plasma by more than 50%. When compared at equal incremental dose, the inhibiting effect of ALA on blood arachidonic acid was stronger than the stimulating effect of LA as precursor.  相似文献

The aim of the study was to evaluate if thymidine kinase (TK) correlated with duration of first remission (DFR) or survival in dogs with lymphoma and if initial TK levels correlated with stage and substage; and also to assess if TK level at diagnosis correlated with immunophenotype. TK was assayed in 73 dogs with treatment naïve lymphoma, then again after treatment; 47% had an initial TK above the reference interval. Dogs with B‐cell lymphoma had higher initial TK activities than dogs with T‐cell lymphoma. TK levels were not higher in dogs with higher stage disease and TK activity prior to treatment was not associated with DFR or survival. Where TK was elevated at diagnosis, it fell into the reference range during remission. TK was normal in 53% dogs at diagnosis, which is higher than previously reported. Further studies are warranted to assess the utility of TK in dogs with lymphoma.  相似文献

Increased numbers of tumour‐associated macrophages correlate with rapid tumour growth and metastasis in tumours. Thus, macrophage depletion has potential as a novel cancer therapy and positive responses have been reported in rodent tumour models. To investigate the effectiveness of this approach in dogs with cancer, we evaluated the effects of the macrophage‐depleting agent liposomal clodronate (LC) in dogs with soft‐tissue sarcoma (STS). To this end, we conducted a clinical trial of LC therapy in 13 dogs with STS. Repeated LC administration was well tolerated clinically. Preliminary examination of tumour biopsy sets from 5 of the 13 dogs demonstrated that the density of CD11b+ macrophages was significantly decreased after LC treatment. Circulating concentrations of interleukin‐8 were also significantly reduced. These preliminary studies are the first to suggest that LC can be used as a systemic macrophage‐depleting agent in dogs to reduce numbers of tumour‐associated macrophages.  相似文献

The reproductive system of the male agouti is not well documented. This study describes the specific anatomical features of the free part of the penis occurring during penile erection in the agouti. Electro-ejaculation was used to induce erection in three male agoutis that had previously produced offspring. Results proved that there were four stages in the erection process. Stage 1 involved protrusion of the penis from the preputial orifice. The lateral penile cartilages were then spread (stage 2). During stage 3, there was the blooming of the head of the glans penis (penile flower) and eversion of the intromittent sac. The protrusion of the keratinaceous styles and ejaculation occurred during stage 4. This information could assist in semen collection for use in reproductive techniques for the agouti such as artificial insemination.  相似文献

Taenia solium causes cysticercosis in pigs and taeniasis and neurocysticercosis in humans. Oncosphere antigens have proven to be effective as vaccines to protect pigs against an experimental infection with T. solium. A pair-matched vaccination trial field, using a combination of two recombinant antigens, TSOL16 and TSOL18, was undertaken in rural villages of Peru to evaluate the efficacy of this vaccine under natural conditions. Pairs of pigs (n=137) comprising one vaccinated and one control animal, were allocated to local villagers. Animals received two vaccinations with 200 μg of each of TSOL16 and TSOL18, plus 5mg Quil-A. Necropsies were performed 7 months after the animals were distributed to the farmers. Vaccination reduced 99.7% and 99.9% (p<0.01) the total number of cysts and the number of viable cysts, respectively. Immunization with the TSOL16-TSOL18 vaccines has the potential to control T. solium transmission in areas where the disease is endemic, reducing the source for tapeworm infections in humans.  相似文献

A randomised clinical trial from the North Island of New Zealand was conducted to assess the effect of repeated anthelmintic treatment on milk production, and to assess factors that affect treatment response. Nine hundred and twenty three multiparous, lactating dairy cattle from three pasture-based, spring-calving dairy herds were enrolled in this trial. Within each herd, cattle were stratified on age and calving date, and were randomly allocated to treatment (n=319) or control (n=604) groups. The treatment group received ≥0.05mg/kg of topical eprinomectin every 28 days for eight treatments during lactation. Pooled-milk from treated cows and bulk-milk samples were obtained at each treatment and analysed with an Ostertagia antibody ELISA, expressed as optical density ratios (ODR). Bi-monthly milk data were collected and expressed as energy-corrected milk (kg/day; ECM). A linear mixed model was used to analyse ECM, with cow as the random effect. The effect of anthelmintic treatment on days from calving, and start-of-mating, to conception were analysed with Cox-proportional hazard models. ODR values ranged from 0.6 to 1.3; there were no differences in ODR between herds (p=0.12), or between pooled-milk from treated cows and bulk-milk (p=0.26). Repeated treatments had no effect on daily ECM yields (p=0.74). However, there was a significant treatment×herd interaction (p=0.03); treatment increased ECM in one herd by 0.781kg/cow/day (p=0.015), but resulted in a non-significant decrease in the other two herds. A curvilinear interaction existed between days-in-milk and treatment response (p=0.039); the greatest treatment effect occurred during mid-lactation. Previous year milk production (p=0.46) and age (p=0.11) did not influence the effect of treatment on ECM. Treatment had no effect on any reproductive parameter. In conclusion, under New Zealand pastoral conditions, anthelmintic treatment increased milk production in one herd, but had no effect in two other herds. Further work is needed to identify why this variation in gastro-intestinal parasitism occurs.  相似文献

Objective-To determine whether administration of 2 doses of a multivalent, modified-live virus vaccine prior to breeding of heifers would provide protection against abortion and fetal infection following exposure of pregnant heifers to cattle persistently infected (PI) with bovine viral diarrhea virus (BVDV) and cattle with acute bovine herpesvirus 1 (BHV1) infection. Design-Randomized controlled clinical trial. Animals-33 crossbred beef heifers, 3 steers, 6 bulls, and 25 calves. Procedures-20 of 22 vaccinated and 10 of 11 unvaccinated heifers became pregnant and were commingled with 3 steers PI with BVDV type 1a, 1b, or 2 for 56 days beginning 102 days after the second vaccination (administered 30 days after the first vaccination). Eighty days following removal of BVDV-PI steers, heifers were commingled with 3 bulls with acute BHV1 infection for 14 days. Results-After BVDV exposure, 1 fetus (not evaluated) was aborted by a vaccinated heifer; BVDV was detected in 0 of 19 calves from vaccinated heifers and in all 4 fetuses (aborted after BHV1 exposure) and 6 calves from unvaccinated heifers. Bovine herpesvirus 1 was not detected in any fetus or calf and associated fetal membranes in either treatment group. Vaccinated heifers had longer gestation periods and calves with greater birth weights, weaning weights, average daily gains, and market value at weaning, compared with those for calves born to unvaccinated heifers. Conclusions and Clinical Relevance-Prebreeding administration of a modified-live virus vaccine to heifers resulted in fewer abortions and BVDV-PI offspring and improved growth and increased market value of weaned calves.  相似文献

A large commercial catfish enterprise encompassing over 500 food fish ponds from five farms covering multiple counties in the Mississippi Delta was included in this analysis of columnaris risk factors. A gram-negative bacterium, Flavobacterium columnare, is the cause of columnaris disease and is considered the second-most prevalent bacterial disease in farm-raised catfish. The objective of this study was to determine if pond-level risk factors reported by farm personnel were associated with columnaris disease mortalities. To identify risk factors affecting susceptibility of farm-raised channel catfish Ictalurus punctatus to columnaris disease, a Catfish Management database was developed. Logistic regression was used to model the relationships between probability of columnaris in ponds and risk factors examined. Generalized linear mixed models incorporating hierarchically structured random effects of ponds and one or more fixed-effects risk factors were fitted. In the screening process, each risk factor was evaluated in the basic model as a single fixed-effects factor, and if associated with the outcome (P ≤ 0.20), was retained for development of multivariable models. Two multivariable logistic regression models were constructed from data collected at the pond level by producers. The first was constructed from data in which water quality was not considered. Pond depth and reduced feed consumption for a 14-d period prior to disease outbreaks measured on a per hectare basis were significantly (P ≤ 0.05) associated with columnaris disease. The second, in which water quality variables were also considered, pond depth, reduced feed consumption, shorter intervals from stocking to disease outbreaks, and total ammonia nitrogen were significantly (P ≤ 0.05) associated with columnaris occurrence. This study showed some commonly recorded production variables were associated with columnaris disease outbreaks and, if monitored, could help identify "at risk" ponds before disease outbreaks occur.  相似文献

Three experiments were conducted to determine the effects of increasing dietary standardized ileal digestible (SID) Lys on growing and finishing gilts. Diets in all 3 experiments were corn-soybean meal-based and contained 0.15% l-Lys?HCl and 3% added fat from choice white grease. Desired SID Lys concentrations were achieved by altering levels of corn and soybean meal in the diet. Each experiment consisted of 6 treatments with 7 pens per treatment and approximately 27 gilts (PIC 337 × 1050) per pen. In Exp. 1, 1,085 gilts (initially 38.2 kg) were fed diets formulated to contain SID Lys concentrations of 0.7, 0.8, 0.9, 1.0, 1.1, or 1.2% for 28 d, which were analyzed to be total Lys concentrations of 0.78, 0.86, 0.99, 1.06, 1.14, and 1.24%, respectively. As SID Lys increased, ADG and G:F improved (quadratic, P < 0.003) with optimal performance reached at the SID Lys level of 1.1% or SID Lys:ME ratio of 3.16 g/Mcal. Broken-line analysis indicated breakpoints of 1.03 and 1.05% SID Lys for ADG and G:F, respectively. Gilts in this trial required approximately 21.8 g of SID Lys intake per kilogram of BW gain from 38 to 65 kg. In Exp. 2, 1,092 (initially 55.2 kg) gilts were fed diets formulated to contain SID Lys concentrations of 0.66, 0.74, 0.82, 0.90, 0.98, or 1.06% for 28 d, which were analyzed to be total Lys concentrations of 0.75, 0.73, 0.84, 0.90, 0.95, and 0.97%, respectively. Both ADG (quadratic, P = 0.12) and G:F improved (linear, P < 0.001) as SID Lys increased, with broken-line analysis of ADG indicating a requirement estimate of 0.90%, which corresponds to a SID Lys:ME ratio of 2.58 g/Mcal. Gilts in this trial required approximately 19.6 g of SID Lys per kilogram of BW gain from 55 to 80 kg. In Exp. 3, 1,080 gilts (initially 84.1 kg) were fed diets formulated to contain SID Lys concentrations of 0.54, 0.61, 0.68, 0.75, 0.82, or 0.89% for 29 d, which were analyzed to be total Lys concentrations of 0.62, 0.92, 0.79, 0.99, 0.93, and 1.07%, respectively. As the SID Lys concentration increased, ADG and G:F improved (linear, P < 0.001), and performance responses were maximized at the greatest SID Lys level of 0.89% or SID Lys:ME ratio of 2.55 g/Mcal of ME. Gilts in this trial required 23.0 g of SID Lys per kg of BW gain from 85 to 110 kg. The ideal SID Lys:ME ratio was based on the requirement determined by broken-line analysis in Exp. 1, 2, and 3, with the greatest level being tested in Exp. 3. This equation, SID Lys:ME ratio = -0.011 × BW, kg + 3.617, estimates the optimal SID Lys:ME ratios for growth of gilts (PIC 337 × 1050) in this commercial finishing environment. These studies showed growth performance advantages to increasing SID Lys for growing and finishing gilts over previously reported optimal levels, particularly in the later finishing stages.  相似文献
