Abstrac

The effects of 3 oxides (Fe, Al, and Mn oxides) and 3 clay minerals (kaolin, montmorillonite, and allophane) on the adsorption and subsequent kinetic properties of acid phosphatase were compared. The amount of enzyme adsorbed by the oxides and clay minerals followed the order: montmorillonite ? kaolin > Mn oxide > Fe oxide > Al oxide ? allophane. The adsorption isotherms of the enzyme on the oxides and clay minerals, except for montmorillonite and allophane, fitted the Langmuir equation. The activity of the enzyme immobilized by the inorganic components studied was in the order of allophane > kaolin > Fe oxide > montmorillonite > Al oxide ≒ Mn oxide. Compared to the free enzyme, the V max, Km, and V max / K m values of the immobilized enzyme decreased, increased, and decreased, respectively. Among the oxides or clay minerals, the higher the ability of the inorganic components to adsorb the enzyme, the lower the value of the V max / K m ratio of the immobilized enzyme. These findings suggest that the catalytic efficiency of the enzyme complexes formed is determined by the adsorbability of the inorganic components for the enzyme.  相似文献

The spatial distribution of Todarodes pacificus in and near the Kuroshio/Oyashio Transition Zone during its northward migration was examined by comparative surveys using two types of mid‐water trawl net and supplementary squid jigging from June to July 2000. The vertical and horizontal distribution patterns varied for different body sizes in relation to the oceanographic structure. Todarodes pacificus of 1–20 cm dorsal mantle length (ML) were widely distributed from the coastal waters of Japan to near 162°E longitude, probably due to transport by the Kuroshio Extension (KE). Todarodes pacificus smaller than 10 cm ML were mainly distributed in temperate surface layers at sea surface temperatures (SSTs) >15°C near the KE meander probably because of their poor tolerance to lower temperatures and limited swimming ability. Squid of 10–15 cm ML were distributed in the offshore waters of 10–15°C SST and in the coastal waters of northern Honshu, and underwent diel vertical migrations between the sea surface at night and deeper layers during the daytime. Squid larger than 15 cm ML were distributed in the coastal feeding grounds of northern Honshu and Hokkaido until they began their southward spawning migration. They also underwent diel vertical migrations, but remained deeper at night than the squid of 10–15 cm ML; this migration pattern closely matched that of their main prey such as euphausiids. We concluded that as T. pacificus grow, they shift their distribution range from the temperate surface layer around the KE toward the colder deeper layers, above 5°C, in the Oyashio and coastal areas.  相似文献

ABSTRACT:   The gonad of Spratelloides gracilis was not sexually differentiated in the yolk-sac, preflexion, flexion and postflexion stages. Sexual differentiation and development of the ovary and testis started in the transition stage from larva to juvenile. In juveniles at the fin ray completion stage, the ovary and testis could be distinguished because the ovary contained germ cells initiating meiosis and the testis had blood vessels and a high density of somatic cells. The ovary further developed in larger juveniles to have oocytes of perinucleolus stage together with those of the chromatin nucleolus stage, and oogonium. However, in the testis of larger juveniles, primary spermatogonium began proliferation by meiosis. Sexual differentiation may be regarded as one of morphological and functional changes accompanying metamorphosis in S. gracilis . Some fish larger than the mature size of 60 mm standard length had advanced germ cells and functional gonads, others did not have functional gonads. The distal end of the immature gonads did not connect with a genital duct near the anus. These observations indicate that S. gracilis has large variability in size-at-maturity. The variability in size-at-maturity in S. gracilis , together with large variability in age-at-maturity, may constitute an ecological basis for an extended spawning season in S. gracilis .  相似文献

Reducing sea turtle by-catch in pelagic longline fisheries   总被引:6,自引:0,他引:6  
Reducing by‐catch of sea turtles in pelagic longline fisheries, in concert with activities to reduce other anthropogenic sources of mortality, may contribute to the recovery of marine turtle populations. Here, we review research on strategies to reduce sea turtle by‐catch. Due to the state of management regimes in most longline fisheries, strategies to reduce turtle interactions must not only be effective but also must be commercially viable. Because most research has been initiated only recently, many results are not yet peer‐reviewed, published or readily accessible. Moreover, most experiments have small sample sizes and have been conducted over only a few seasons in a small number of fisheries; many study designs preclude drawing conclusions about the independent effect of single factors on turtle by‐catch and target catch rates; and few studies consider effects on other by‐catch species. In the US North Atlantic longline swordfish fishery, 4.9‐cm wide circle hooks with fish bait significantly reduced sea turtle by‐catch rates and the proportion of hard‐shell turtles that swallowed hooks vs. being hooked in the mouth compared to 4.0‐cm wide J hooks with squid bait without compromising commercial viability for some target species. But these large circle hooks might not be effective or economically viable in other longline fisheries. The effectiveness and commercial viability of a turtle avoidance strategy may be fishery‐specific, depending on the size and species of turtles and target fish and other differences between fleets. Testing of turtle avoidance methods in individual fleets may therefore be necessary. It is a priority to conduct trials in longline fleets that set gear shallow, those overlapping the most threatened turtle populations and fleets overlapping high densities of turtles such as those fishing near breeding colonies. In addition to trials using large 4.9‐cm wide circle hooks in place of smaller J and Japan tuna hooks, other fishing strategies are under assessment. These include: (i) using small circle hooks (≤ 4.6‐cm narrowest width) in place of smaller J and Japan tuna hooks; (ii) setting gear below turtle‐abundant depths; (iii) single hooking fish bait vs. multiple hook threading; (iv) reducing gear soak time and retrieval during daytime; and (v) avoiding by‐catch hotspots through fleet communication programmes and area and seasonal closures.  相似文献

Neonicotinoid pesticides (NNs) cause behavioral abnormalities in mammals, raising concerns about their effects on neural circuit activity. We herein examined the neurological effects of the NN clothianidin (CLO) by in vivo Ca2+ imaging using two-photon microscopy. Mice were fed the no-observed-adverse-effect-level (NOAEL) dose of CLO for 2 weeks and their neuronal activity in the primary somatosensory cortex (S1) was observed weekly for 2 weeks. CLO exposure caused a sustained influx of Ca2+ in neurons in the S1 2/3 layers, indicating hyperactivation of neurons. In addition, microarray gene expression analysis suggested the induction of neuroinflammation and changes in synaptic activity. These results demonstrate that exposure to the NOAEL dose of CLO can overactivate neurons and disrupt neuronal homeostasis.  相似文献

It was previously reported that (-)-epigallocatechin-3-O-gallate (EGCG) suppresses the expression of the high-affinity IgE receptor FcepsilonRI in human basophilic cells and that this suppressive effect is associated with EGCG binding to the cell surface. This study examined the effects of five methylated derivatives of EGCG, (-)-epigallocatechin-3-O-(3-O-methyl)gallate (EGCG 3' 'Me), (-)-epigallocatechin-3-O-(4-O-methyl)gallate (EGCG 4' 'Me), (-)-4'-O-methyl-epigallocatechin-3-O-gallate (EGCG 4'Me), (-)-epigallocatechin-3-O-(3,4-O-methyl)gallate (EGCG 3' '4' 'diMe), and (-)-4'-O-methyl-epigallocatechin-3-O-(4-O-methyl)gallate (EGCG 4'4' 'diMe) on FcepsilonRI expression and ERK1/2 phosphorylation, and each of their cell surface binding activities was measured. Of these five methylated derivatives, three that are methylated at the 3' '- and/or 4' '-position, EGCG 3' 'Me, EGCG 4' 'Me, and EGCG 3' '4' 'diMe, suppressed FcepsilonRI expression and ERK1/2 phosphorylation, although the suppressive effects were lower than that of EGCG. EGCG 4'Me and EGCG 4'4' 'diMe, both of which are methylated at the 4'-position, did not demonstrate a suppressive effect. Furthermore, it was found that EGCG 3' 'Me, EGCG 4' 'Me, EGCG 3' '4' 'diMe, and EGCG 4'Me, which are methylated at the 3' '- and/or 4' '-positions or the 4'-position, could bind to the cell surface even though their binding activities were lower than that of EGCG. Only EGCG 4'4' 'diMe, which is methylated at both the 4'- and 4' '-positions, could not bind. These results suggest that the trihydroxyl structure of the B ring is essential for EGCG to exert the suppressive effects and that the hydroxyl groups on both the 4'-position in the B ring and the 4' '-position in the gallate are crucial for the cell surface binding activity of EGCG.  相似文献
