The development of obesity in porcine fetuses was investigated using a lean and obese strain of pigs at 80, 90, 100 and 110 d of gestation. In absolute terms, fetuses of obese gilts (FO) generally had lower carcass weight and contained less total protein, dry matter and ash than fetuses of lean gilts (FL). In relative terms (percentage of wet carcass weight) FO, compared with FL, generally had decreased percentages of water and increased percentages of protein and lipid. Comparisons based on absolute terms revealed body composition of the strains to be different at 90 d, indicating that factors responsible for obese-type growth were active before that time. Both body composition and hormone concentration differences were most pronounced at later gestation ages. Depressed growth hormone, elevated cortisol, and a tendency toward elevated insulin concentrations in fetal plasma were apparent in late gestation for FO compared with FL. These hormonal patterns are consistent with onset of obesity in FO in late gestation. Greater weights of semitendinosus and longissimus muscles were observed in FL vs FO at 90, 100 and 110 d of gestation (P less than .05). These greater muscle weights were generally accompanied by greater contents of RNA, DNA and protein in FL muscles at these same ages. However, at 80 d, FL had greater absolute DNA content in semitendinosus muscle whereas muscle weight was similar between the strains. This suggests that greater muscle weights for FL than FO were caused by more nuclei in muscle of FL. In general, indices of hypertrophy (protein/DNA) and protein synthetic capacity (RNA/DNA) of muscle were usually similar for both strains at all gestation ages. It is concluded that decreased muscle growth in late gestation of FO compared with FL is more related to fewer total nuclei and perhaps fewer myofibers than to an impaired cellular capacity for protein synthesis.  相似文献

1. A new homologous radioimmunoassay has been developed for the measurement of turkey prolactin.

2. A 25000 kDa purified recombinant derived turkey prolactin (rtPRL), the biological activity of which was tested using a crop sac assay, was used as immunogen for the production of rabbit antiserum. In this biological test, the rtPRL was as active as the ovinePRL.

3. The radioligand (rtPRL) was labelled with 125I and the assay allowed the detection of standard doses of rtPRL ranging from 400 pg/tube to 50 ng/tube.

4. No cross reaction with chicken luteinising hormone and recombinant chicken growth hormone was detected.

5. The within and between assay coefficients of variability were 5.0 ± 2.7% and 16.3%, respectively. The overall mean recovery ratio was 1.01.

6. The dose‐response curves obtained with serial dilution of plasma and pituitary from turkey hens at different physiological stages and from male turkeys were parallel to those obtained with standard rtPRL.

7. The measured concentration of prolactin was 5 times higher in plasma from incubating than laying turkey hens, and the pituitaries from incubating hens contained 2 and 4 times more prolactin than those of laying and out of lay hens or males, respectively.

8. To further assess the validity of the assay, we measured changes in plasma concentration of prolactin in turkeys following stimulation with chicken vasointestinal peptide (cVIP). A single injection of 1 or 10 μg/kg body weight of cVIP to laying hens produced a large and rapid increase in plasma prolactin.

BACKGROUND: This investigation was prompted by the referral of increasing numbers of young Texel and Beltex rams with ataxia and weakness, or wobbler syndrome. HYPOTHESIS: The study aims were to describe the clinical and pathologic findings in affected sheep. ANIMALS: The animals evaluated in this study included 7 Texel sheep (6 male and 1 female) and 3 Beltex sheep (2 male and 1 female) referred from pedigree flocks. Typically, the sheep were 15-18 months of age at referral. METHODS: Diagnostic investigations included radiographic and computed tomographic (CT) myelography followed by gross postmortem and histopathologic examinations. RESULTS: Clinical findings typical of cervical spinal cord compression were present in all sheep but varied in severity. Myelography confirmed dorsal spinal cord compression in the region of C6-C7. No bony abnormalities were identified as described in cases of canine and equine wobbler syndrome. Postmortem examinations revealed discrete, smooth, nodular to polypoid projections of adipose tissue apparently prolapsing through the dorsolateral intervertebral space at C6-C7 and causing localized spinal cord compression. Histopathology of the nodules confirmed that they were composed of well-differentiated adipocytes typical of fatty tissue. Spinal cord lesions were similar in all sheep with marked Wallerian degeneration at the site of compression and mild Wallerian degeneration present cranial and caudal to the lesion. CONCLUSIONS AND CLINICAL IMPORTANCE: The findings of this study suggest a novel cervical myelopathy in these sheep breeds caused by the presence of fatty nodules encroaching into the dorsal vertebral canal at C6-C7. Additional investigations are required to establish the etiology and possible hereditary risk factors for this unique clinicopathologic syndrome.  相似文献

It is generally assumed that animal species differ in physiological characteristics of their digestive tract. Because investigating digestive processes is often labour‐intensive, requiring lengthy adaptation and collection periods, comparisons are often made on the basis of data collated from different studies. We added a new data set on dietary crude fibre (CF) and macromineral (Na, K, Ca, P, Mg) composition and apparent digestibility (aD) of organic matter (OM) and macrominerals in rabbits (Oryctolagus cuniculus; a total of 180 measurements with 12 individuals and 10 different diets) to a literature data collection for rodents and domestic horses (Equus caballus). Significant negative relationships between dietary CF and aD OM were obtained, but the significant difference in digestive efficiency between rabbits and guinea pigs (Cavia porcellus) reported from studies where both species had been investigated under identical conditions were not detectable in the larger data collection. However, the 95% confidence interval of the negative slope of the CF–aD OM relationship did not overlap between domestic horses on the one hand, and rabbits and guinea pigs on the other hand, indicating a less depressing effect of CF in horses. Akaike's information criterion indicated that aD of a macromineral did not only depend on its concentration in the diet, but also on the individual and various other diet characteristics and the presence of other macrominerals, indicating complex interactions. The data indicate similar mechanisms in the absorption of macrominerals amongst rabbits, rodents and horses, with the exception of Na and K in guinea pigs and Ca in chinchillas (Chinchilla laniger) that should be further corroborated. In particular, the occurrence of high aD Ca in hindgut fermenters, regardless of whether they employ a digestive strategy of coprophagy or whether they have fossorial habits, suggests that this peculiarity might represent an adaptation to hindgut fermentation in general.  相似文献

Although use of corridors by some wildlife species has been extensively examined, use by bats is poorly understood. From 1 June to 31 August (2004–2005), we used Anabat II detectors to examine bat activity and species occupancy relative to forested corridors on an intensively managed forest landscape in southern South Carolina, USA. We compared bat activity among corridor interiors, corridor edges, and stands adjacent to corridors. We also compared models relating occupancy of bat species to site-level characteristics using an information theoretic approach. We identified 16,235 call sequences of 8 species and detected bat presence at 89% (n = 320) of sites sampled. Our results indicate higher occupancy rates for bats along corridor edges compared to interior corridor or adjacent stands. Although we found few differences among species with respect to site-level characteristics, occupancy of all bat species was positively associated with corridor overstory height and negatively associated with adjacent stand age. The presence of roads adjacent to corridors positively influenced occupancy of Eptesicus fuscus, Lasiurus seminolus, and Perimyotis subflavus. Our results suggest management practices designed to create and enhance corridors may represent an ecologically sound method for maintaining important bat habitat features (i.e., edge) across managed forest landscapes.  相似文献

In this study, the effect of polymorphisms in the leptin gene on the hematological variables in periparturient dairy cows was investigated. The hematological profile of 67 Holstein cows was assessed for 6 wk around calving. The DNA of the cows was genotyped at 6 polymorphic loci within the leptin gene, and 7 haplotypes were reconstructed. Significant haplotype substitution effects were found, for haplotype 1, on total white blood cell count for 2 wk around calving (+0.70 10(3)/μL, P = 0.05; +1.38 10(3)/μL, P = 0.0001); on neutrophil cell count in the first week after calving (+0.94 10(3)/μL, P = 0.001); on lymphocyte count during the 3 wk before and the first week after calving (+0.32 10(3)/μL, P = 0.05; +0.27 10(3)/μL, P = 0.03; +0.26 10(3)/μL, P = 0.04; +0.34 10(3)/μL, P = 0.01); on red blood cell count during the last week before calving and wk 1 and 2 after calving (+0.21 10(6)/μL, P = 0.02; +0.23 10(6)/μL, P = 0.01; +0.20 10(6)/μL, P = 0.03); on mean corpuscular volume (-1.35 fL, P = 0.01; -1.29 fL, P = 0.002; -1.18 fL, P = 0.004; -1.09 fL, P = 0.008; -1.23 fL, P = 0.003; -1.31 fL, P = 0.003); and on mean corpuscular hemoglobin (-0.37 pg, P = 0.05; -0.38 pg, P = 0.02; -0.39 pg, P = 0.01; -0.34 pg, P = 0.03; -0.40 pg, P = 0.01; -0.40 pg, P = 0.01) during all 6 wk analyzed. Significant haplotype substitution effects, but opposite those of haplotype-1, were found for haplotype-2 on white blood cell count (-1.10 10(3)/μL, P = 0.01; -1.30 10(3)/μL, P = 0.002; -1.09 10(3)/μL, P = 0.01) and neutrophil count (-0.82 10(3)/μL, P = 0.02; -0.95 10(3)/μL, P = 0.005; -0.92 10(3)/μL, P = 0.01). Haplotype-3 influenced red blood cell count (-0.23 10(6)/μL, P = 0.03; -0.28 10(6)/μL, P = 0.01; -0.34 10(6)/μL, P = 0.002) during the last 2 wk before and the first week after calving, and also, with effects evident only in wk 3 and 2 before calving, mean corpuscular volume (+1.38 fL, P = 0.03; +0.97 fL, P = 0.05; +1.08 fL, P = 0.05), mean corpuscular hemoglobin (+0.58 pg, P = 0.02; +0.38 pg, P = 0.04; 0.51 pg, P = 0.01), and red blood cell distribution width (-0.56% P = 0.02; -0.47%, P = 0.05). The current study provided evidence that several polymorphisms in the leptin gene play a role in the variability of hematological variables during the peripartum period, and might be used as genetic markers for improving the immunological conditions of dairy cows in critical productive periods.  相似文献

A mixed breed dog presented with diffuse unilateral hind limb swelling, which ultrasound and cytology confirmed to be caused by severe haematoma formation. Multi-detector computed tomography (MDCT) angiography allowed distinct visualisation of an anomalous segment of the proximal popliteal artery, the presumed origin of the self-sustaining haematoma. Histopathology classified the malformed vessel as a haemangioendothelioma, a neoplasia of intermediate malignancy. Considering this as differential diagnosis to a neoplastic vascular alteration of high malignancy (such as haemangiosarcoma) might alter choice of treatment in future cases with similar clinical and imaging findings.  相似文献
