We previously showed that an intradermal (i.d.) FaeG DNA prime (2x)-oral F4 protein boost immunization induces a systemic response and weakly primes a mucosal IgG response in pigs, especially when plasmid vectors encoding the A and B subunit of the E. coli thermo-labile enterotoxin (LT) are added to the DNA vaccine. In the present study, we evaluated whether addition of 1alpha,25-dihydroxyvitamin D(3) (vitD(3)) to the DNA vaccine could further enhance this mucosal priming and/or modulate the antibody response towards IgA. To further clarify priming of systemic and mucosal responses by the i.d. DNA vaccination, we firstly compared the localization of the F4-specific antibody response in pigs that were orally boosted with F4 to that in pigs that received a third i.d. DNA immunization and secondly evaluated cytokine mRNA expression profiles after i.d. DNA vaccination. The i.d. DNA prime (2x)-oral F4 boost immunization as well as the 3 i.d. DNA vaccinations induced mainly a systemic response, with a higher response observed following the heterologous protocol. Co-administration of vitD(3), and especially of the LT vectors, enhanced this response. Furthermore, only the heterologous immunization resulted in a weak mucosal priming, which appeared to require the presence of the LT vectors or vitD(3) as adjuvants. In addition, the LT vectors strongly enhanced the FaeG-specific lymphocyte proliferation and this was accompanied by the absence of a clear IL-10 response. However, despite two DNA immunizations in the presence of these adjuvants and an oral F4 boost, we failed to demonstrate the secretory IgA response needed to be protective against enterotoxigenic E. coli. 相似文献

The wild tetraploid wheat species Tr$$ (Zhuk) Zhuk Var. araratieum is a source of pest resistance genes for T$$ aesti$$ L. Our objectives were to describe the breeding behaviour of T.arartuititm when backcrossed to common wheat and transfer resistance to leaf rust (caused by Pu$$) and powdery mildew (caused by Blumeria $$wheat. Crosses were made between five wheat genotypes and $$ accessions. Fertifity and chromosome numbers of BC$$; plants were determined. Resistance to leaf rust was transferred toBC2 -derived families from 10 different T’ararati$$an accessions. Leaf rust resistance genes in nine T. araratieum accessions can be assigned to at least four loci. Leaf rust resistance transferred from three accessions was inherited in the hexaploid derivatives as a single. $$ gene in each case. Resistance to powdery mildew was also detected in the T. araratie$$ backcross derivatives. The bottleneck restricting introgression of useful genes directly from diploid into hexaploid wheats is the low number of BC1F1 seeds obtained. In crosses between hexaploid wheat (Triticum aestivum L.; AABBDD) and Aegilops squarrosa L. (DD) or T. urartu Thum. (AA), this bottleneck may be overcome simply by pollinating a sufficient number of F1 spikes. However, hybrids between hexaploid wheat cultivars (T. aestivum) and T. monococcum L. (AA) generally are highly female-sterile, often having no pistils. One T. monococcum accession, PI 355520, when crossed with T. aestivum, produced hybrids with female fertility in the same range as that of T. aestivum/A. squarrosa or T. aestivum/T. urartu hybrids, ca. 0.5 to 1.0 backcross seed per spike. We found that female fertility was controlled by two duplicate genes in PI 355520, and that this accession can be used as a bridging parent to introgress genes from other T. monococcum accessions into hexaploid wheat. Pairing of homologous chromosomes was less frequent and weaker in such crosses than in T. aestivum/A. squarrosa crosses, but homoeologous bivalents occurred at a rate of almost 0.5 II per cell. Restitution division was detected in crosses involving all three diploid species and was confirmed cytologically in crosses with PI 355520. Chromosome numbers of BC1F1 plants ranged from 35 to 67; plants with 49 or more chromosomes occurred at frequencies of 0.09 to 0.21 among progeny of A. squarrosa and T. urartu and 0.29 in progeny of T. aestivum/T. monococcum crosses involving PI 355520. These results are consistent with those of previous studies, demonstrating the potential of direct Hexaploid/diploid crosses for rapidly introgressing useful genes into Hexaploid wheat with minimum disturbance of the background genotype. 相似文献

Lacking a tradition of settling immigrants and the appropriate infrastructure to integrate them, small‐ and medium‐sized cities often face the challenge of attracting and retaining immigrants. Using a mixed methods approach, this study compares the experiences of immigrant and non‐immigrant entrepreneurs in a mid‐sized Canadian city, Kelowna, British Columbia. A survey reveals different experiences between these two groups, with immigrants facing more challenges. In the absence of institutionally complete communities or strong ethnic economies, immigrants do not rely extensively on their own community resources, an element considered instrumental for immigrant business development in large cities. Compared to non‐immigrants, immigrant entrepreneurs have a more optimistic outlook on doing business in Kelowna; this is encouraging for a city trying hard to attract immigrant investment. Key informants recommended transforming the city into a more welcoming community, establishing appropriate support infrastructure, and removing potential institutional offsets. This paper adds new theoretical insights to the literature on immigrant entrepreneurship; all socio‐cultural, political‐institutional, and economic‐structural considerations are embedded in geography. BR-105 and BR-106 are important tropical maize populations, which were submitted to a high-intensity reciprocal recurrent
selection, generating the IG-3 and IG-4synthetics. Using 30-microsatellite loci,we measured and compared the genetic diversity
of these populations and their synthetics. The populations did not differ significantly regarding the amount of genetic diversity.
As a consequence of selection, genetic variability losses, in terms of mean number of alleles per locus,proportion of polymorphic
loci, and gene diversity did occur and were greater in the synthetic IG-3 than IG-4. In the synthetics, the number of loci
in adherence to Hardy-Weinberg proportions was superior to that observed in the populations. The Wright's mean fixation index
was higher than the mean value expected for outcrossing species (5%) indicating as light excess of homozygotic individuals
in both populations. The genetic distances confirmed the favourable effects of one cycle of recurrent selection, as the synthetics
became more isolated in comparison to the original populations.

The antioxidizing potency of phenol compounds contained in olive oil mill wastewater (OOMWW) has been elucidated. Commercially available phenol standards at varying concentrations and the Rancimat oxidation test have been used. Refined purified olive oil was utilized as an oxidation lipid substrate. Synthetic antioxidants, such as 2,3-tert-butyl-4-hydroxyanisole (BHA), 3,5-di-tert-butyl-4-hydroxytoluene (BHT), l-ascorbic acid, and gallates (commonly used as food preservatives), and other known chemicals endowed with antioxidizing properties have been employed as reference compounds. The OOMWW phenol compounds have been classified into different groups depending on their antioxidizing potency. This was significantly affected by the tested concentrations of the standards. Mixtures of phenol standards and other antioxidants (l-proline, chlorophyll a, chlorophyll b, and alpha-, gamma-, and delta-tocopherol) have also been tested. Many phenol compounds present in OOMWW showed antioxidizing potency higher compared to that of the less safe synthetic antioxidants and could therefore replace these in the industrial preservation of food items. They could also be used in combination with other natural antioxidants (e.g., tocopherols). The concept of Cumulative Erosion Potential (CEP) proposed by De Ploey, Kirkby and Ahnert in 1991 (Earth Surf. Process. Landforms, 16: 399–409) provides a simple climatic index of erosion potential based on annual rainfalls and their distribution, which is assumed to be exponential (or gamma with shape parameter 1). A modified CEP (CSEP) is presented here, extending the original in three ways. First, the index is disaggregated by month to include the effects of seasonality. Second, a mixed gamma(1) distribution is used to provide a better fit to the whole of the daily rainfall distribution. Third, the index is calculated both for a bare soil and for a natural vegetation cover, giving an additional index to the potential for managing erosion through landuse change. For the natural vegetation, the index is based on an estimate of equilibrium soil organic matter which is associated with differences in soil moisture storage capacities and derived from monthly rainfall and temperature values. The index thus remains a measure of climatic potential only, based on widely available monthly data on temperature, rainfall and number of rain days. It may be compared with the patterns shown by the European Soil Erosion Map and by the CORINE classification, which are dominated by a combination of climate and lithology. 相似文献
