Twelve feeder pigs were used to determine the effects of dietary protein on colonic microstructure and function. Scanning electron microscopy indicated marked differences in mucosal structure of pigs fed 12% vs 20% crude protein. Quantitative assessment of the morphologic features of the colon, using light microscopy, indicated significant differences in the mucosa of pigs fed 12% vs 20% crude protein and significant differences between the proximal and distal portions of the colon. Morphologic changes were associated with differences in the colonic transport of fluids and electrolytes. Dietary protein principally affected the transport of sodium, bicarbonate, and acetate. The transport of total volatile fatty acids and acetate flux were significantly different between the proximal and distal portions of the colon.  相似文献

The element sulfur has an outstanding role in the crop protection chemistry because it is used in its elemental form as a multisite fungicide, but is also part of agrochemicals in the form of aromatic or aliphatic sulfur-containing rings or sulfur-based functional groups. This review gives an exhaustive overview over the latter category. Several fundamental agrochemical compound classes are named after a sulfur-based functionality, such as the dithiocarbamate fungicides and sulfonylurea herbicides. Altogether, 16 different sulfur-based functional groups are presented with their typical synthesis approaches and most important representatives in crop protection. © 2023 Society of Chemical Industry.  相似文献

Novel types of mitochondrial respiration inhibitors at complex I, with emphasis on acaricidal activity, have been designed and prepared. The synthetic approach to these 4‐pyrimidinylphenyl ethyl ethers and amines with a specific ketal or oxime function in the phenyl side chain is outlined. Bioassays demonstrate their high potential against important spider mites, like Tetranychus urticae and Panonychus ulmi. Cytochromes P450 (P450s) metabolize a wide range of endogenous compounds and xenobiotics, such as pollutants, environmental compounds, and drug molecules. The microsomal, membrane-associated, P450 isoforms CYP3A4, CYP2D6, CYP2C9, CYP2C19, CYP2E1, and CYP1A2 are responsible for the oxidative metabolism of more than 90% of marketed drugs. Cytochrome P450 3A4 (CYP3A4) metabolizes more drug molecules than all other isoforms combined. Here we report three crystal structures of CYP3A4: unliganded, bound to the inhibitor metyrapone, and bound to the substrate progesterone. The structures revealed a surprisingly small active site, with little conformational change associated with the binding of either compound. An unexpected peripheral binding site is identified, located above a phenylalanine cluster, which may be involved in the initial recognition of substrates or allosteric effectors.  相似文献

The modern Indian summer monsoon (ISM) is characterized by exceptionally strong interhemispheric transport, indicating the importance of both Northern and Southern Hemisphere processes driving monsoon variability. Here, we present a high-resolution continental record from southwestern China that demonstrates the importance of interhemispheric forcing in driving ISM variability at the glacial-interglacial time scale as well. Interglacial ISM maxima are dominated by an enhanced Indian low associated with global ice volume minima. In contrast, the glacial ISM reaches a minimum, and actually begins to increase, before global ice volume reaches a maximum. We attribute this early strengthening to an increased cross-equatorial pressure gradient derived from Southern Hemisphere high-latitude cooling. This mechanism explains much of the nonorbital scale variance in the Pleistocene ISM record.  相似文献

Arsenic mobility, bioavailability, and toxicity in soil‐water systems are strongly affected by species distribution. In spite of species‐selective analytical tools, results may be biased by postsampling oxidation or reduction of compounds. The scope of this study was to delineate a speciation method for inorganic arsenic integrated with pore‐water sampling to prevent problems related to postsampling species transformation. Experiments were performed with exchange cartridges packed with quaternary amine (nitrate‐form) to separate anionic arsenate from uncharged arsenite. Standard solutions with varied As(III) : As(V) ratios as well as groundwater and soil‐solution samples were studied. Analyses were performed by ICP‐MS using either extraction cartridges directly coupled to the instrument or samples obtained by offline speciation tests. Results showed that As(III) passes the packed bed with the rinsing solution, while As(V) is retained on the column and can be released by elution with 0.25 M NH4NO3. Recoveries between 98% and 116% were insensitive to pH variation (pH 3–8) and competing anions. Groundwater samples from a contaminated site yielded recoveries between 92% and 125% when the sum of As(III) and As(V) was compared to total As. Integration of the speciation scheme with pore‐water sampling was tested in a soil‐column experiment. Soil material from a fen site with elevated geogenic As contents was subjected to a varying moisture regime. Reducing conditions during prolonged saturation caused a marked increase of As(III) concentrations. Conversely, when the soil was drained, total As levels decreased and were dominated by As(V). Overall, extraction results with standard solutions and natural samples indicate reliable performance of the method in experimental investigations and field studies.  相似文献

Ammonia volatilization from slurry is undesirable because of environmental N eutrophication and loss of fertilizer value. The dry matter content of slurry, the application technique and the weather conditions are the main factors influencing NH3 losses from landspread slurry. In a field of winter wheat a two factor plot experiment was conducted to study single and combined effects of slurry separation and application techniques, including broadcast and banded application, as well as incorporation by injection and the flexible harrow. Ammonia volatilization from all treatments could be measured simultaneously, and at ambient climatic conditions by an indirect, open measurement technique. The experiment was repeated four times. Due to varying weather conditions and treatment effects, cumulative NH3 volatilization from the slurry during the first 48 hours ranged from 4 to 90% of total ammoniacal nitrogen (TAN). Both separation and incorporation significantly decreased NH3 losses, but only the combination of dry matter reduction and injection or harrowing reduced NH3 volatilization to about 30% of TAN in all weather conditions. Banding alone did not efficiently conserve slurry N, but even enhanced NH3 volatilization in wet conditions.  相似文献

For the detection of prohibited meat and bone meal (MBM) in animal feed, monoclonal antibodies (MAbs) were raised against heat-stable h-caldesmon purified from bovine intestinal smooth muscle. The obtained hybridoma cells were screened against extracts of the bovine MBM and heat-treated smooth muscle, and MAb 5E12 was identified as having the best performance. Antibody 5E12 did not react with animal feed, milk product, plant proteins, and other ingredients used for commercial animal feed except for the gelatin. This antibody diluted to 100-fold was able to detect MBM mixed in animal feed at 0.05% in an ELISA, and it showed strong affinity toward bovine smooth muscle autoclaved at 130 degrees C. Therefore, this antibody can be used in the ELISA system for field testing of the presence of MBM in animal feed.  相似文献

Adult mallard ducks were administered steel pellets to determine the rate of excretion from the gastrointestinal tract. In separate studies the ducks were administered 5 number 6 lead pellets. Birds were examined for clinical signs and sacrificed at given intervals over a 20 day period to assess changes in tissue structure and concentrations of lead with time. The above studies were conducted in 2 groups of ducks, fed a low or a high fiber diet. The rate of steel pellet excretion on birds on the low fiber diet decreased with an increase in pellet size. Pellet excretion was greatly reduced in birds fed the high fiber diet. Administration of lead shot resulted in the development of green diarrhea, anorexia and weakness. It also produced high concentrations of lead in the blood, kidney, liver and bone with lower concentrations in skeletal muscle. The major lesions were destruction of the mitotically active proventricular epithelium and medullary osteocytes, destruction of pectoral muscle cells and the presence of intranuclear inclusion bodies in the proximal tubular epithelium of the kidneys. Birds on the high fiber diet demonstrated more severe clinical signs and higher concentrations of lead in the tissues.  相似文献
