Plant parasitic nematodes of the genus Heterodera show a high degree of sexual dimorphism, which is reflected by different nutritional demands and differences in the structure of the induced specific syncytial feeding site in the plant. The determination of the sex of the nematode Heterodera schachtii and other related species was repeatedly reported to be dependent on trophic factors, which are provided by the induced syncytia. The structural differences of syncytia induced by H. schachtii in roots of Arabidopsis thaliana were analysed at the anatomical and ultrastructural level. Syncytia of males were induced in the root pericycle. The developing syncytium then expanded into procambial or cambial cells of the vascular cylinder. Differentiated vascular elements were not included. The expansion of the syncytium triggered the proliferation of cambial and peridermal tissues, in a manner similar to secondary growth, and the formation of additional xylem and phloem elements. In comparison to syncytia associated with females, syncytia associated with males were less hypertrophied and were composed of more cells. Distinct cell wall openings were mostly found between the few strongly hypertrophied syncytial elements at the actual feeding site in the pericycle. The ultrastructure was very similar to female-associated syncytia but showed conspicuous differences in the structure and localization of cell wall ingrowths. These ingrowths were rare and weakly developed and occurred not only at the interface with xylem elements but also at the internal and external walls of the syncytia. After feeding had ceased at the end of the third developmental stage the syncytia degenerated.  相似文献

Pathological tissues from a variety of plants turn red when treated with the histochemical reagent phloroglucinol (PG)–HCl. This induced PG–HCl reacting material has been termed both wound gum and, more recently, induced lignin or lignin-like material. We are exploring alternative approaches to better identify this induced material. In this report, we describe methods for purifying the PG–HCl reactive material from solvent-washed tissue preparations and measuring the amount of p -coumaryl aldehyde released from the purified material by boiling in water. Acorn squash fruit wall tissue was sliced, sprayed with pectinase to elicit the formation of PG–HCl reactive material and incubated up to 72 h at 27°C. Sampled tissue was washed extensively with water and organic solvents. Insoluble residues were treated with cell wall degrading enzymes to remove cell wall materials, and dimethyl sulfoxide was used to dissolve starch. Yields of residual material increased from 7.5% in the time zero samples to 29.7% after 72 h. Refluxing the purified material in water released <6 μ g p -coumaryl aldehyde g−1 from preparations of tissue at time zero and 8100 μ g p -coumaryl aldehyde g−1 from samples incubated for 48 h. These results suggest that the function of the material is to sequester phytoalexins, but are not consistent with the material being lignin. Published by Elsevier Science Ltd.  相似文献

A reducing solution of 2-mercaptoethanol and its oxidized form 2-hydroxyethyl disulfide, whose variable concentrations set variable disulfide reduction potentials, was applied to progressively reduce the disulfide bonds of proteins extracted from doughs made from Meneba and Robin Hood flour. Several dough proteins had disulfide bonds stronger than those of other dough proteins. A SDS-sedimentation method was applied to monitor the baking of dough into bread. Dough proteins susceptible to heat (baking) were studied by SDS-fractionation, extraction with reducing alcoholic solution, SDS-PAGE, and N-terminal protein sequencing. High or low molecular weight glutenins, α, β, and γ-gliadins, α-amylase inhibitor, and α-amylase trypsin inhibitor were identified among the dough proteins modified by heat (as shown by reduced solubility in aqueous-SDS solution). The heat-induced modification of the gliadins and glutenins might contribute to the coagulation of dough proteins, while the heat-induced modification of the amylase or trypsin inhibitors might contribute to the regulation of endogenous or exogenous amylolytic or proteolytic activities in dough or bread.  相似文献

The dynamics of soil water was investigated in a Mediterranean vineyard during the 2003-2007 period in order to identify the inter-seasonal modification of water storage due to intercropping. The intercrop was a mixture of tall fescue and rye grass. Soil water content was measured on 3 m soil profiles (plus one 5 m profile) as was runoff from local stands.Great variations in soil refilling were observed over the years, for both the bare soil and intercrop treatments. The complete refilling occurred once and it was associated with a rise of the water table up to less than 2 m from the soil surface in the low part of the field. During the other years, the wetting front was identified between 1 m and 2.3 m at spring in the bare soil treatment, deeper in the lowest part of the field. The available soil water content at bud-break was influenced by the topography. A higher infiltration was observed in the intercropped treatment. Runoff kinetics was compared to rainfall kinetics in a selection of three rain events differing by duration and intensity. The curve number method was used to perform the analysis of runoff at a 1-day time-step rather than the quarter of an hour time-step of the registrations. With this method, the time limitation of water balance studies for Mediterranean vineyards would be partially removed.  相似文献

Context

Animal population dynamics are shaped by their movement decisions in response to spatial and temporal resource availability across landscapes. The sporadic availability and diversity of resources can create highly dynamic systems. This is especially true in agro-ecological landscapes where the dynamic interplay of insect movement and heterogeneous landscapes hampers prediction of their spatio-temporal dynamics and population size.

Objectives

We therefore systematically looked at population-level consequences of different movement strategies in temporally-dynamic resource landscapes for an insect species whose movement strategy is slightly understood: the Queensland Fruit Fly (Bactrocera tryoni)

Methods

We developed a spatially-explicit model to predict changes in population dynamics and sizes in response to varying resources across a landscape. We simulated the temporal dynamics of fruit trees as the main resource using empirical fruiting dates. Movement strategies were derived from general principles and varied in directedness of movement and movement trigger.

Results

We showed that temporal continuity in resource availability was the main contributing factor for large and persistent populations. This explicitly included presence of continuous low-density resources such as fruit trees in urban areas. Analysing trapping data from SE Australia supported this finding. We also found strong effects of movement strategies, with directed movement supporting higher population densities.

Conclusions

These results give insight into structuring processes of spatial population dynamics of Queensland Fruit Fly in realistic and complex food production landscapes, but can also be extended to other systems. Such mechanistic understanding will help to improve forecasting of spatio-temporal hotspots and bottlenecks and will, in the end, enable more targeted population management.

Compounds released by plant roots during growth can make up a high proportion of below-ground plant (BGP) carbon and nitrogen, and therefore influence soil organic matter turnover and plant nutrient availability by stimulating the soil microorganisms. The present study was conducted to examine the amount and fate of C (CdfR) and N rhizodeposits (NdfR), in this study defined as root-derived C or N present in the soil after removal of roots and root fragments, released during reproductive growth. BGP biomass of peas (Pisum sativum L.) and oats (Avena sativa L.) was successfully labelled in situ with a 13C-glucose-15N-urea mixture under field conditions using a stem feeding method. Pea plants were labelled at the beginning of flowering and harvested 36 and 52 days after labelling at pod filling (PP) and maturity (PM), respectively. Oat plants were labelled at grain filling and harvested 42 days after labelling at maturity (OM). CdfR was 24.2% (PP), 29.6% (PM) and 30.8% (OM) of total recovered plant C. NdfR was 32.1% (PP), 36.4% (PM) and 30.0% (OM) of total plant N. Due to higher N assimilation, amounts of NdfR were four times higher in peas in comparison with oats. The results for NdfR in peas were higher than results from other studies. The C-to-N ratio of rhizodeposits was lower under peas (17.3) than under oats (41.9) at maturity. At maturity, microbial CdfR at 0-30 cm soil depth was 37% of the microbial biomass C in peas and 59% in oats. Microbial NdfR was 15% of microbial N in peas and 5% in oats. Furthermore, inorganic NdfR was 34% in peas and 9% in oats at 0-30 cm at maturity. These results show that rhizodeposits of peas provide a more easily available substrate to soil microorganisms, which are incorporated to a greater extent and turned over faster in comparison with oats. Beside the higher amounts of N released from pea roots, this process contributes to the higher N-availability for subsequent crops.  相似文献
