The improving effects of transglutaminase (TGase) were investigated on the frozen dough system and its breadmaking quality. Rheological properties and microstructure of fresh and frozen doughs were measured using a Rapid Visco‐Analyser (RVA), dynamic rheometer, and scanning electron microscopy (SEM). The frozen doughs with three storage periods (1, 3, and 5 weeks at –18°C) were studied at three levels (0.5, 1.0, and 1.5%) of TGase. As the amount of TGase increased, hot pasting peak viscosity and final viscosity from the RVA decreased, but breakdown value increased. The TGase content showed a positive correlation with both storage modulus G′ (elastic modulus) and the loss modulus G″ (viscous modulus): G′ was higher than G″ at any given frequency. The SEM micrographs showed that TGase strengthened the gluten network of fresh, unfrozen dough. After five weeks of frozen storage at –18°C, the gluten structure in the control dough appeared less continuous, more disrupted, and separated from the starch granules, while the dough containing 0.5% TGase showed less fractured gluten network. Addition of TGase increased specific volume of bread significantly (P < 0.05) with softer bread texture. Even after the five weeks of frozen storage, bread volume from dough with 1.5% TGase was similar to that of the fresh control bread (P < 0.05). The improving effects of TGase on frozen dough were likely the result of the ability of TGase to polymerize proteins to stabilize the gluten structure embedded by starch granules in frozen doughs. 相似文献

This study was designed to isolate bifidobacteria from human intestines that efficiently converts monolinolein, a monoglyceride form of linoleic acid, into conjugated linoleic acid (CLA), as well as to optimize culture conditions for improving CLA production during milk fermentation. Among 150 screened neonatal bifidobacteria, Bifidobacterium breve LMC 520 showed the highest CLA-producing ability and was tested with different types of fat substrates at various concentrations to determine the optimal conditions for CLA production. Monolinolein was tested as a substrate for CLA production. The incubation time optimized for CLA production was 24 h, and CLA production was proportionally increased with monolinolein concentration. The incubation of LMC 520 with commercial starter strains caused minimal reduction in CLA production. Our results demonstrate that the CLA-producing ability of B. breve LMC 520 could offer beneficial effects when utilized as a starter culture for the development of functional dairy products. 相似文献

Soil vapor extraction (SVE) is one of the most effective remediation technologies for soil and groundwater contamination. Soil particles can be mobilized by air perturbation during SVE, resulting in the differentiation of porous media, which has not been well addressed. This paper developed a numerical method to study the flow pattern and quantify the change of porous media for the first time. Based on the mass equilibrium and Darcy’s law, a two-phase water–air flow model was constructed with integration of saturation, relative permeability, and capillary pressure during SVE. Relationship between porosity and saturation was deduced and coupled with the two-phase flow model for quantifying change of porous media in real time. Results reveal that both porosity and permeability increase sharply in the early stage of SVE then gradually to a quasi-steady state. These increases in vadose zone tapered off with distance from the SVE screen and the steady period occurred later as well. The influence radius of a single SVE well and the change degree in porosity and permeability of media were proportional to the extraction vacuum and the driving coefficient C, which is more sensitive than extraction vacuum according to the simulation results. Knowledge from this modeling exercise provides a useful tool to estimate the change of remediated zone and assess the environmental risk of remedial activities at real-world contamination sites. 相似文献

Ballast seawater is considered globally as a major vector for invasions of non-indigenous organisms. Several technologies have been tested for their ability to remove organisms from ballast water. In the present study, we constructed a novel pulsed high-voltage discharge (PHVD) system that could operate in either high current mode with several hundred amperes or shockwave generating mode with relatively lower current in seawater. In laboratory-scale experiments, the PHVD system with shockwave-generating mode was found to be more effective in killing zooplankton (1.9- to 4.0-fold) and phytoplankton (3.3-fold) than high current mode at discharge with 300–500 pulses at 7.1 kV. Further experiments were carried out at different voltages and pulse-numbers to examine effects of the shockwave-generating PHVD system on viabilities of one zooplankton larva, two phytoplankton species, and an indicator bacterium suspended in seawater in a static chamber. For zooplankton, live cells were not detected at discharge with 400 pulses at 13 kV. For phytoplankton, the initial live cells of a dinoflagellate was decreased by 77?±?0.5%, and the initial chl a concentration of a diatom was decreased by 76?±?6% at discharge with 700 pulses at 13 kV. For an indicator bacterium Escherichia coli, live cells were not detected at discharges with 200 or 700 pulses at 13 kV. Measurements of ATP content of organisms showed congruent results with those obtained by the above methods, suggesting it may be a rapid method for evaluating treatment efficiency. Though further scale-up studies are necessary, these results suggest that the PHVD system have a high potential for applying to ballast seawater treatment. 相似文献

The volatile components of different meats (legs with claws, body, and carapace) of a popularly consumed edible crab in Asia, Charybdis feriatus, were investigated. Samples were extracted by simultaneous steam distillation-solvent extraction and analyzed by gas chromatography/mass spectrometry. Among 177 compounds detected, 130 were positively identified. Seventy-six compounds were previously reported in other crab species. A greater number of naphthalenes were detected in this crab compared with other crabs in the literature. Aromatic compounds, alcohols, and sulfur-containing compounds were the three predominant groups with >15 components. Carapace tissue contained a greater number of volatile components in each group, except for sulfur-containing compounds. Most of the common components in the leg meat and the body meat were found at similar levels (p > 0.05). Carapace tissue generally had the highest quantity of common components among the meats. The higher levels of volatile components present in the carapace tissue might account for its stronger flavor compared with the other meats. Furthermore, the higher number of aldehydes and lower number of sulfur-containing compounds detected in the carapace meat might contribute to its unique flavor. 相似文献

Selected types of commercial breads obtained from local markets, including white sandwich, Irish oatmeal, soft rye, hearty rye, sour dough, home-like white, and onion-basil, were analyzed for volatiles. Using a purge and trap instrument, volatiles were purged directly from fresh crumb and crust samples of each bread type, collected on a trap (Tenax-TA), and transferred to a gas chromatograph. Separated components were detected and identified using mass and infrared spectroscopic detectors. Many components were present in all of the bread samples, with relative amounts varying among bread types and crust and crumb samples of a given bread type. Alcohols were generally the most abundant, followed in approximate order by aldehydes, esters, ketones, acids, various aromatics, terpenes, and hydrocarbons. Flavor additives, such as limonene, carvone, and other related compounds, were found mostly in rye and onion-basil breads. Composition of volatiles from sour dough bread differed greatly from the other breads, especially in increased levels of aldehydes, acids, and certain esters. Brachiaria decumbens is an extremely productive tropical grass due to its aggressive growth habit and its adaptation to a varied range of soil types and environments. As a result of the vast availability, treated B. decumbens demonstrates as a promising local material that could be utilised as an improved diet for sheep and goats. Despite the fact that the grass significantly increases weight gains in grazing farm animals, there were many reports of general ill-thrift and sporadic outbreaks of photosensitivity in livestock due to the toxic compound of steroidal saponin found in B. decumbens. Ensiling and haymaking were found to be effective in removing toxin and undesirable compounds in the grass. Biological treatments using urea, activated charcoal, polyethylene glycol, and effective microorganisms were found to be useful in anti-nutritional factor deactivation and improving the nutritive values of feedstuffs. Besides, oral administration of phenobarbitone showed some degree of protection in sheep that fed on B. decumbens pasture. Viral clearance during hepatitis B virus (HBV) infection has been thought to reflect the destruction of infected hepatocytes by CD8(+) T lymphocytes. However, in this study, HBV DNA was shown to largely disappear from the liver and the blood of acutely infected chimpanzees long before the peak of T cell infiltration and most of the liver disease. These results demonstrate that noncytopathic antiviral mechanisms contribute to viral clearance during acute viral hepatitis by purging HBV replicative intermediates from the cytoplasm and covalently closed circular viral DNA from the nucleus of infected cells. 相似文献

Acid rain at a rural site in Chongwon (KHN), central Korea has been under observation since 1990. To substantiate the 10-year observations, rain samples from six other sites were also collected and analyzed. Results show that acid rain occurred frequently at all sites. At KHN, seasonal and annual variability of pH values had a 10-year VWM value of 4.67 and an arithmetic mean value of 5.09. In general, a high value of pH occurred during springtime when the occurrence of yellow sand (dust) was prevalent in East Asia; during summer, heavy rain usually contained neutral pH values. During other seasons, the degree of acidic precipitation usually coincided with certain air flows. Most frequently, air flows and cyclones coming from south and central China often resulted in precipitation with low pH values observed in Korea. Using daily satellite observations from 1996, we have also been monitoring the long-range and large-scale transport of air pollutants (LSTAP) over East Asia. Many samples of LSTAP were obtained when they were crossing the Yellow Sea and moving over the Korean Peninsula and to SW Japan. The width of the Yellow Sea is ca. 350~700 km and air pollutants take 1~2 days to cross it. Cases of acid rain in Korea clearly correlate wth the LSTAP from source regions in China. 相似文献
