Many different fungi grow indoors as mold in the presence of moisture. Some of these fungi can cause allergic or toxic reactions, while a few may cause infections in susceptible individuals. A comprehensive treatment of this complex topic would take volumes. Therefore many groups search for alternative management strategies of these fungi. It has been tested if the growth of nine fungi (Alternaria alternata, Botrytis cinerea, Cladosporium herbarum, Fusarium culmorum, F. solani, F. verticillioides, Mucor sp., Penicillium sp. and Trichoderma sp.) which are common in interior buildings could be inhibited by mixing lignin into wall plaster. In maltextract agar lignin concentrations above 10 % inhibited mould growth except for Penicillium sp. However, none of the mould species showed and degradation of lignin. Further investigations on a lignin-plaster-agar, which contained only plaster as nutrient and had a high pH at 8, showed no inhibition on the tested fungi (Alternaria alternata, Fusarium verticillioides, Mucor sp., Trichoderma sp. and Penicillium sp.) at concentrations up to 5 %. Higher lignin concentrations lead to insufficient quality of building materials, e. g. cracking. Fusarium oxysporum and Fusarium proliferatum are important causal agents of crown and root rot of asparagus. In order to detect differences in pathogenicity and aggressiveness, two F. proliferatum and five F. oxysporum single spore isolates from asparagus spears from plantings in Austria and Germany, 55 pure cultures of F. oxysporum from asparagus roots from a planting in Hesse, Germany, and a single F. oxysporum isolate from an asparagus shoot collected in Austria were evaluated in a 28-day quick test on Hoagland??s agar in glass culture tubes. Plantlets were inoculated with spore suspensions from each respective isolate after 14 days of growth under sterile, controlled conditions in a growth chamber. A severity scale was used to assess symptoms on roots two weeks after inoculation. The effects of the single-spore isolates on root and shoot fresh weights of the plantlets were also determined. The pathogenicity of the majority of the F. proliferatum and F. oxysporum isolates included in this study was confirmed. Inoculation with pure and single-spore cultures resulted in elevated disease severity in comparison to non-inoculated controls. In particular, the two F. proliferatum isolates were found to be more aggressive than the F. oxysporum isolates. Moreover, all single spore isolates caused a reduction in fresh weight of roots and shoots in comparison to the controls. With respect to differences among asparagus cultivars, ??Ramos??, was found to be more susceptible than ??Ravel??. Overall, the quick test method was found to be capable of evaluating the pathogenicity and aggressiveness of the tested F. oxysporum and F. proliferatum isolates towards asparagus within 28 days. 相似文献

Zusammenfassung Erstmalig wurden mit den vorliegenden Untersuchungen Spargelstangen zur Haupterntezeit auf endophytischen Pilzbefall untersucht. Sie zeigen, dass im Ernteprodukt zwar Fusarium proliferatum als potenzieller Mykotoxinbildner zu finden ist. Eine mögliche natürliche Kontamination mit Fumonisinen bestätigte sich nicht. Von den mit F. proliferatum infizierten Stangen wies nur eine Stange mit grau-rosa-orange farbenen Gewebeveränderungen an der Basis sichtbare Symptome auf. Allgemeine Rückschlüsse auf eine mögliche Gefährdung oder Nichtgefährdung des Verbrauchers beim Verzehr von mit F. proliferatum kontaminierten, symptomlosen Stangen können aus der Analyse nicht gezogen werden. Powdery mildew fungi are parasitized by strains of the genetically distinct Ampelomyces quisqualis. To investigate whether differences in the phylogeny and other cultural, morphological and physiological characteristics
of these different strains are related to differences in their geographic origins or the host species from which they were
isolated, several A. quisqualis strains isolated from different species of Erysiphaceae collected in different countries and possessing different ITS rDNA
sequences were selected and characterized. The results revealed some significant variation among the selected strains, which
provides evidence for the existence of different physiological forms within the A. quisqualis species. Two groups that display differential growth on artificial media were identified. These groups also differ in the
morphology of their mycelium, but not in the morphology of their pycnidia and conidia. Temperature greatly affected the in vitro growth of the A. quisqualis strains and growth rate was closely correlated to colony color. Differences in the conidial germination of distinct strains
were observed during the recognition phase of the parasitic relationship. The germination of each of the investigated strains
was greatly stimulated by all of the examined powdery mildew species and not only by the conidia of their original hosts.
An Italian strain isolated from grapevine in the Trentino Alto-Adige region was identified as the strain that germinates the
most quickly in the presence of powdery mildew conidia. Phylogenetic analysis revealed that these A. quisqualis strains can be classified into five different genetic groups, which generally correlate with the fungal host of origin and

Primers for the PCR amplification of homologous genes encoding polyketide coronafacic acid and coronafacic ligase in the cells
of Pectobacterium atrosepticum SCRI1043 (BX950851) were developed to study the presence of these genes in the genome of Pectobacterium sp. and Dickeya sp. Coronafacic ligase catalyses the formation of coronatine from polyketide coronafacic acid and coronamic acid. Coronatine
is a toxin produced by Pseudomonas syringae and is one of the major virulence factors in this bacterium. This study using several strains of P. atrosepticum, P. carotovorum subsp. carotovorum and Dickeya sp. isolated in different countries, indicated that all strains of P. atrosepticum possess genes coding coronafacic acid (cfa gene cluster) and coronafacic ligase (cfl). However, these genes were present only in the genome of five out of 50 tested P. carotovorum subsp. carotovorum strains and two out of 34 strains of Dickeya sp. tested. The PCR products homologous to the sequence of cfa7 and cfl gene fragments were sequenced in order to check the level of homology between genes of P. atrosepticum, P. carotovorum subsp. carotovorum and Dickeya sp. The sequences of the gene fragments amplified from all P. atrosepticum strains were almost identical (100% and 99.97%, respectively). The homology of the sequences obtained for P. atrosepticum and sequences of five P. carotovorum subsp. carotovorum and two Dickeya sp. was lower, between 89.69% to 95.00% for the cfl gene fragment, and about 94% for the cfa7 gene fragment. 相似文献

An experiment was carried out in a climate chamber to analyse if Bt-maize may cause particular changes in soils with different levels of microbial biomass and activity due to long-term management history. Among the soils selected, the ones managed organically for 30 years exhibited twice the microbial biomass and 2.6 times the dehydrogenase activity (DHA) of the soil from a field with long-term conventional maize monoculture. Soils were cultivated twice in a row with Bt-maize, its near-isogenic line and a conventional breeding line. We tested the hypotheses that (a) soil microbial biomass and activity are affected by the cultivation of Bt-maize and that (b) the influence of Bt-maize depends on the level of soil microbial biomass and activity. Shoot and root yield and shoot C-content of Bt-maize were higher than the ones of the near-isogenic line. DHA under Bt-maize was 6 % higher, and the metabolic quotient for CO2 (qCO2) was 9 % lower than under its near-isogenic line, giving some support to hypothesis (a). No significant interactions of the soils and the varieties used were found in this study, thus hypothesis (b) was not confirmed, and soils with different microbial biomass and activity appear to react in a similar way to the cultivation of Bt-maize. 相似文献

Activated CD8+ T cells detect virally infected cells and tumor cells by recognition of major histocompatibility complex class I-bound peptides derived from degraded, endogenously produced proteins. In contrast, CD8+ T cell activation often occurs through interaction with specialized antigen-presenting cells displaying peptides acquired from an exogenous cellular source, a process termed cross-priming. Here, we observed a marked inefficiency in exogenous presentation of epitopes derived from signal sequences in mouse models. These data indicate that certain virus- and tumor-associated antigens may not be detected by CD8+ T cells because of impaired cross-priming. Such differences in the ability to cross-present antigens should form important considerations in vaccine design. 相似文献

Glucosinolates, present in Brassica vegetables, are thought to contribute to human health prevention because of their enzymatically induced breakdown products, primarily isothiocyanates (ITCs). ITCs are reactive substances that readily react with nucleophilic (food) compounds. The reactivity of allyl-ITC and 4-(methylsulfinyl)butyl-ITC (sulforaphane) toward thiol and amino groups of cysteine and lysine derivatives was studied in buffered model systems as well as broccoli sprouts. The thiol group is the preferred reaction site, and it was demonstrated that even endogenously released sulforaphane is able to react very fast with cysteine in broccoli sprouts. Amino groups reacted slower and only under basic conditions. However, great differences in the reactivity between the different amino compounds were revealed. The aliphatic allylamine reacted very fast with allyl-ITC, forming N,N'-diallylthiourea, a compound identified as a main thermal degradation product of allyl-ITC. 相似文献
