Hessian fly [Mayetiola destructor (Say)] is one of the major insect pests of wheat (Triticum aestivum L.) worldwide. Hessian fly resistance gene H9 was previously reported to condition resistance to Hessian fly biotype L that is prevalent in many wheat‐growing areas of eastern USA and an RAPD marker, OPO051000, linked to H9 in wheat was developed using wheat near‐isogenic lines (NILs). However, marker‐assisted selection (MAS) with RAPD markers is not always feasible. One of the objectives in this study was to convert an RAPD marker linked to the gene H9 into a sequence characterized amplified region (SCAR) marker to facilitate MAS and to map H9 in the wheat genome. The RAPD fragment from OPO051000 was cloned, sequenced, and converted into a SCAR marker SOPO05909, whose linkage relationship with H9 was subsequently confirmed in two F2 populations segregating for H9. Linkage analysis identified one sequence tagged site (STS) marker, STS‐Pm3, and the eight microsatellite markers Xbarc263, Xcfa2153, Xpsp2999, Xgwm136, Xgdm33, Xcnl76, Xcnl117 and Xwmc24 near the H9 locus on the distal region of the short arm of chromosome 1A, contrary to the previously reported location of H9 on chromosome 5A. Locus Xbarc263 was 1.2 cM distal to H9, which itself was 1.7 cM proximal to loci Xcfa2153, Xpsp2999 and Xgwm136. The loci Xgwm136, Xcfa2153 and SOPO05909 were shown to be specific to H9 and not diagnostic to several other Hessian fly resistance genes, and therefore should be useful for pyramiding H9 with other Hessian fly resistance genes in a single genotype. 相似文献

在工程实践中,直墙拱应用较多。由于拱脚位移的存在,合理假设拱脚约束为转角约束、竖向约束和水平弹性约束,应用力法推导出了在弯矩、剪力和轴力共同作用下直墙拱沿弧长的弯矩公式。研究了竖向均布荷载、竖向三角形荷载和竖向集中力荷载作用下,直墙拱沿弧长的弯矩分布及反弯点形成的规律;发现在3种荷载作用下,使得拱脚弯矩为零时的圆心角依次减小。取弹簧支座刚度为等效的下端固支、上端允许水平位移的直墙的抗推刚度,得出了随着直墙高度增加,圆弧拱拱顶弯矩增大而拱脚弯矩减小的变化规律。在直墙拱的设计中,建议选取合适的直墙高度和使得圆弧拱拱脚弯矩为零的圆心角大小,从而有利于提高结构抗弯承载力及拱脚抗剪承载力。 相似文献

Summary The banana cultivars are originated from the intra- and inter-specific hybridization of two wild diploid species, Musa acuminata Colla and Musa balbisiana Colla, contributing the A and B genomes, respectively. They are classified into genomic groups by scoring morphological features.
Molecular markers provide a quick and reliable system of genome characterization and manipulation in breeding lines. In the
present study a PCR based molecular marker specific for B genomes is been reported. The IRAP primer, designed based on the
LTR sequence of banana Ty3-gypsy-like retroelement (Musa acuminata Monkey retrotransposon, AF 143332), was used to identify the B genome in the banana cultivars. Further a primer pair designed
from B specific bands of Musa balbisiana `Pisang Gala' was used to classify AAB and ABB cultivars in the collection. Among the 36 cultivars tested with this primer,
the B specific band was absent in the AA and AAA cultivars (except in one AAA and AAB cultivar) but present in all other AB,
AAB and ABB cultivars. Among the triploid AAB/ABB, the PCR products with B specific primers showed restriction pattern polymorphism
with AluI. In ABB genomes the band intensity was high whereas low intensity band observed in AAB genomes. Four cultivars reported
to have the ABB genome showed a pattern similar to AAB, and one cultivar reported to have AAA genome showed a pattern similar
to ABB genome, suggesting missampling or misidentification. The primers used in this study are useful to identify the presence
of B genome in banana cultivars, and band intensity may be a preliminary indicator of ploidy level of the B genome but needs
further studies with competitive PCR for clarification.

农村居民点整治应满足农户生产生活的需求,提高农村居民点的生产生活可达性。该文从生产生活可达性切入,以农村居民点生产生活可达性作为整治分区和采取何种整治模式的依据,从生产可达性和生活可达性两方面出发构建生产生活可达性评价指标体系,采用空间可达性测度方法和指标综合评判法计算得到农村居民点(斑块)生产生活可达性,将其分为I、II、III、IV 4个等级,分别对应发展区、优化区、引导区和拆迁区。根据整治分区并结合实际情况,划分了城镇化发展模式、城乡统筹发展模式、综合优化模式、辐射引导模式、增减挂钩模式和生态搬迁模式等6种整治模式,并对整治思路和具体做法做了探讨。该研究尝试将生产生活可达性评价结果应用于农村居民点整治,对有序整治农村居民点和改善提高农村人居环境有指导意义。 相似文献

A novel aporphine alkaloid, magnofficine (1), has been isolated from the root barks of the Chinese medicinal plant Magnolia officinalis, along with seven known compounds. 利用8种不同类型的农药对美国白蛾3龄幼虫进行了致死效果试验。结果表明,4.5%高效氯氰菊酯乳油、25%高氯.辛硫磷乳油对美国白蛾致死速度最快,在36h的致死率均达100%。48%毒死蜱乳油、5.7%高氯.氟铃脲乳油在48h的致死率也达到了100%,3.2%阿维菌素乳油2000倍液在72h的致死效果达到了97.5%,0.2%甲氨基阿维菌素苯甲酸盐乳油1000倍液在72h的致死效果达到了90%。而5%氟铃脲乳油和5%灭幼脲乳油在72h对美国白蛾的致死率较低,仅分别达到27.6%、42.2%。 相似文献
