Because of its role in human disease, there are increasing global requirements for reliable diagnostic and control methods for Trichinella in food animals to ensure meat safety and to facilitate trade. Consequently, there is a need for standardization of methods, programs, and best practices used in the control of Trichinella and trichinellosis. This review article describes the biology and epidemiology of Trichinella, and describes recommended test methods as well as modified and optimized procedures that are used in meat inspection programs. The use of ELISA for monitoring animals for infection in various porcine and equine pre- and post-slaughter programs, including farm or herd certification programs is also discussed. A brief review of the effectiveness of meat processing methods, such as freezing, cooking and preserving is provided. The importance of proper quality assurance and its application in all aspects of a Trichinella diagnostic system is emphasized. It includes the use of international quality standards, test validation and standardization, critical control points, laboratory accreditation, certification of analysts and proficiency testing. Also described, are the roles and locations of international and regional reference laboratories for trichinellosis where expert advice and support on research and diagnostics are available.  相似文献

Endometritis is a postpartum uterine disease of cows that interrupts reproductive cycles resulting in suboptimal fertility, reduced performance and profitability of the dairy herd. The objective of the study was to estimate the perceived and observed prevalence of endometritis among zero-grazed dairy cows in smallholder farms in Rwanda. A snowball sampling method was applied in cross-sectional survey to obtain data from 370 farms on 466 cows within their 21–60 days postpartum (dpp). The survey, conducted from September 2018 to March 2019, simultaneously examined cows using the Metricheck Device (MED) to determine the presence and type of vaginal mucus (VMC) based on a score scale of 0 to 3. Cows scoring VMC ≥ 1 were recorded as clinical endometritis (CLE)–positive. Cytotape (CYT) was used to determine the percentages of polymorphonuclear cells (PMN) in endometrial cytology sample. Cows with ≥ 5% PMN were recorded subclinical endometritis (SCLE)–positive, whereas cows with VMC-0 and < 5% PMN were considered healthy cows. At cow-level, endometritis prevalence was 70.2% with 67.2% CLE and 31.8% SCLE while at the herd-level, prevalence was 71.1% with 68.1% CLE and 34.4% SCLE. The differences between the diagnostic performance of the MED and CYT were significant (p < 0.001). Perceived prevalence by farmers was much lower (3.2%) and without agreement with the observed prevalence (kappa = − 0.02, p > 0.05). The highly observed prevalence and farmer underestimation of endometritis prevalence indicate knowledge gaps about endometritis. The extension service therefore needs to increase awareness and education among smallholder farmers about detection and management of endometritis.

Catadromy among freshwater eels is increasingly recognised as being facultative, with some individuals carrying out growth exclusively in brackish or coastal marine waters, or switching between brackish or marine waters and freshwater habitats. In an attempt to reconstruct habitat use of yellow‐stage American eels in a large river‐lake ecosystem, trace element line scans were obtained, using LA‐ICP‐MS, from the otoliths of 110 eels sampled at various locations throughout the St. Lawrence River–Lake Ontario (SLRLO) system. Elemental profiles for strontium (Sr), barium (Ba), manganese (Mn) and magnesium (Mg) enabled us to distinguish three chemical signatures that appear to represent three distinct habitats within the SLRLO. Of these, one was shown to likely correspond to the brackish estuary (high strontium values). The other two signatures, characterised by low strontium but variable concentrations of barium and manganese, may correspond to habitats within the main‐stem St. Lawrence River and one or more of its tributaries. Most (78%) of the switches among habitats occurred within the first four years after recruitment suggesting an increasing likelihood for eels to maintain residence in one habitat as they grow older. This suggests that tributaries may provide important habitats for American eels during the first several years after recruiting to the SLRLO. In addition, our results suggest that a small proportion American eels in the SLRLO can undertake movements on the order of at least 200 km during the early growth stage. This information has important implications for the management and conservation of this species in the system.  相似文献

European sea bass, Dicentrarchus labrax L., groups reared under different conditions during larval stages (mesocosm and intensive rearing) were monitored during on‐growing in sea cages until marketable size (350–400 g). Four caged groups were followed for a period of 17 months each at a pilot scale farm, where vertical distribution behaviour was monitored. This was performed while fish were under calm conditions, during feeding and when stress events occurred. Also during two specific periods: (i) spawning and (ii) high water temperature. Clear differences in the behavioural pattern of swimming depth, displacement and used space between the groups were observed. The individuals from the mesocosm rearing were more sensitive to human presence, showing stronger reactions (speed of displacement and vertical distribution). Most pronounced differences were observed during the ‘extreme’ warm period and the reproductive season. Within the seasons, European sea bass responded during feeding and stress showing a tendency to move deeper compared to calm conditions. The increased displacements were longer during feeding and stress.  相似文献

ABSTRACT:   This study is concerned with the effect of hyrdated diets on growth and feed performance of Dentex dentex . Three levels of dietary water were tested: 7%, 20% and 40%. For the 7% level a commercial feed was used and the 20% and 40% water levels were obtained after adding an appropriate amount of deionized water to the commercial feed and mixing the pellets well. The diets were offered to fish by self-feeders, by which fish were allowed continuous access to diets after activating the self-feeders; by belt feeders, by which the diets were fed continuously and in excess over a 12 h interval; and by hand to apparent satiation two times a day. After 151 days of feeding, the group fed on the 20%-water diet showed significantly ( P  < 0.05) higher growth rate than the 7 and 40% groups, while no significant differences were observed in feed conversion rate or daily feed consumption. The means of feed delivery did not influence growth rate. Self-feeder, belt and hand feeding all gave comparable growth. Fish fed the more condensed 7%-water diet had smaller stomachs than fish fed the larger volume 40%-water diet. In the stomach, all groups of fish hydrated their digesta within 2 h after feeding to a moisture content of around 60%. Apparently no difference in digestive process was observed between groups. It was concluded that the addition of water in diet at the level of 20% has a beneficial effect on dentex growth.  相似文献

Manipulating the N release from high-N crop residues by simultaneous mixing of these residues with organic biological waste (OBW) materials seems to be a possible method to reduce NO3 leaching. The aim of this study was to examine whether the incorporation of OBW materials together with a high-N crop residue (celery) had also an effect on N2O emission from horticultural soil under short-term and optimised laboratory conditions. A sandy loam soil and celery residues were mixed with different OBW materials and brought into PVC tubes at 80% water-filled pore space and 15°C. Every 2.5 h, a gas sample was taken and analysed by gas chromatography for its N2O concentration. The soil amended with only celery residues had a cumulative N2O emission of 9.6 mg N kg–1 soil in 50 h. When the celery residues were mixed with an OBW material, the N2O emission was each time lower than the emission from the celery-only treatment (between 3.8 and 5.9 mg N kg–1 soil during maximum 77 h), except with paper sludge (17.2 mg N kg–1 soil in 100 h). The higher N2O emission from the paper sludge treatment was probably due to its unusually low C:N ratio. Straw, green waste compost 1 (GWC1) and 2 (GWC2), saw dust, and tannic acid reduced the N2O emission of the celery treatment by 40 to 60%. Although the N2O reduction potential can be expected to be lower and with differing dynamics under field conditions, this study indicates that apart from reducing NO3 leaching, OBW application may at the same time reduce N2O emissions after incorporation of high-N crop residues.  相似文献

MyD88 is a key downstream adapter for most Toll-like receptors (TLRs) and interleukin-1 receptors (IL-1Rs). MyD88 deficiency in mice leads to susceptibility to a broad range of pathogens in experimental settings of infection. We describe a distinct situation in a natural setting of human infection. Nine children with autosomal recessive MyD88 deficiency suffered from life-threatening, often recurrent pyogenic bacterial infections, including invasive pneumococcal disease. However, these patients were otherwise healthy, with normal resistance to other microbes. Their clinical status improved with age, but not due to any cellular leakiness in MyD88 deficiency. The MyD88-dependent TLRs and IL-1Rs are therefore essential for protective immunity to a small number of pyogenic bacteria, but redundant for host defense to most natural infections.  相似文献

Soil is considered as one of the most biodiverse environments on Earth; yet, the taxonomy, occurrence, and role of its different microbial populations are largely unknown. Here, two sterilized soils (from England and Italy) were inoculated with a subsample of their initial microbial communities and/or those from the other soil to study their microbial community evolution. This approach compared two driving factors (original community and soil physico-chemical characteristics) for microbial community definition. After 2 months of incubation and based on metagenomic datasets, the two inoculated communities (from an English grassland and an Italian forest) possessed similar functional and taxonomical structures when inoculated in the same sterile soil. For example, the newly colonized Italian soil was dominated by Actinobacteria related organisms (>66 % of the detected community) with a functional distribution independent of the inoculated soil origin. In addition, some of the organisms that dominated the different inoculated communities after 2 months were similar for a given sterile soil whether they came from the English grassland or the Italian forest, and they had not been detected in the original microbial community from either soil. Thus, similar microorganisms with low representation from the two distinct communities emerged in each sterilized soil, thus increasing the microbial diversity recovered from the microbial community of the donor soil. So far, these observations support the idea that different temperate soil microbial communities have different evenness due to environmental physico-chemical variations, yet have similar community composition (richness), and thus develop similarly when colonizing the same habitat.  相似文献

A study was conducted to estimate the seroprevalence of Brucella ovis infection in rams in the Estrie and Bas-Saint-Laurent regions (Quebec). Rams sera (n = 258) were serologically evaluated from 224 rams in 30 commercial flocks and from 34 rams at 2 slaughterhouses by using an enzyme linked immunosorbent assay. Epididymides and testes were examined by palpation on farms and microscopically for culled rams. No ram was seropositive to Brucella ovis or had lesions suggestive of brucellosis from the farm or slaughterhouse surveys.  相似文献

A pathogen was transmitted from apricot trees showing symptoms of viral infection to GF305 peach seedlings which reacted by stunting, shortened internodes and chlorotic mottling. The agent was transmitted to cherry, apricot, peach and plum by grafting and to several herbaceous hosts by mechanical inoculation. Isometric nepovirus-like particles of 30–31 nm diameter extracted from infected Chenopodium quinoa sedimented as two peaks in sucrose gradients. These particles contained two single stranded RNAs of approximately 5.9 and 7.9 kb, and a single coat protein subunit of 53.7 kDa. No cross-reactions were observed with a number of nepoviruses infecting fruit trees. Inoculation of purified particles to herbaceous or woody hosts reproduced the same symptoms caused by the original isolate. Sequencing of a 2.2 kbp cDNA clone covering the 3 end of the small genomic RNA identified an open reading frame encoding a 317 aa N-truncated protein exhibiting significant similarities with the coat protein of nepoviruses. The 1257 nt long 3 non-coding region showed up to about 65% homology to the equivalent region of members of the subgroup C of nepoviruses. The properties of this pathogen do not match those of any previously described nepovirus. It should therefore be considered as a new member of the subgroup C of nepoviruses, for which the name of Apricot latent ringspot virus (ALRSV) is proposed.The nucleotide sequence reported in this work has been deposited in the EMBL databank under the accession number AJ278875.  相似文献
