Plants can take up intact amino acids, even in competition with soil microbes, yet we lack detailed information on which amino acids dominate the soil and whether amino acid composition varies seasonally. This study tested the hypotheses that 1) the pool of amino acid N is generally larger than inorganic N; 2) temporal changes in the concentration of amino acid N is related to changes in the size of the microbial N pool; and 3) amino acid N is dominated by simple, neutral amino acids during warm months, whereas during cold months the amino acid N is dominated by more complex aromatic and basic amino acids. Approximately every month for two years we collected soil from a temperate, sub-alpine grassland in the Snowy Mountains of Australia. We quantified exchangeable pools of amino acids, nitrate and ammonium in 1 M KCl extracts. Microbial N was quantified by chloroform fumigation. Averaged across the 21 monthly samples, nitrate was 13% of the quantified pool of soluble non-protein N, ammonium was 34% and amino acid N was 53%. These data are consistent with our hypothesis that the pool of amino acid N is larger than inorganic N. There was substantial variation between months in concentrations of amino acids and inorganic N, but no clear temporal pattern. Microbial N did not vary between months, and thus changes in amino acid N were unrelated to microbial N. Principal components analysis indicated multivariate groupings of the different pools of N that were broadly indicative of function and/or biosynthetic relationships. Thus PCA identified a grouping of aromatic amino acids (Phe and Try) with amino acids derived from oxaloacetate (Asp, Ala, Val, Leu, Ile), and a second group comprising microbial N, nitrate and glycine. The pool of exchangeable amino acid N was dominated by Arg (26% of amino N) Val (20%) Gln (18%), Try (8%) and Asn (8%). Contrary to our hypothesis, the composition of the amino acid pool did not vary in a consistent way between months, and there was no evidence simple amino acids were relatively more abundant in warm months and complex amino acids in cool months.  相似文献

Changes in the protein content and profile of postharvest tomato fruit treated with the hormetic dose (3.7 kJ m?2) of ultraviolet light C (UV-C) at the mature green stage was investigated. In UV-C treated fruits, the total protein content increased until 10 d after treatment and decreased thereafter during a 30 d storage period; whereas in control fruit, protein content decreased constantly throughout the storage period. Using polyacrylamide gel electrophoresis (PAGE) it was shown that UV-C treatment affected the protein profile of tomato fruit in several manners: (1) UV-C repressed the expression of some proteins presumably associated with ripening; (2) it enhanced the expression of several constitutive proteins, of which one was an acidic β-1,3-glucanase, three acidic chitinases and three basic chitinases; and (3) it induced the synthesis of at least 5 new proteins of which four were basic proteins. Among the proteins induced by UV-C, three (a basic β-1,3-glucanase and two acidic chitinases) were apparently pathogenesis-related proteins as they were also induced by inoculation with Botrytis cinerea. The molecular mass (MM) of five of the UV-C induced proteins was determined using SDS-PAGE. Their molecular masses were 45, 39.4, 34.6, 10 and 8.9 kDa. The UV-C induced β-1,3-glucanase had a MM of 33.1 kDa. The MM of two constitutive chitinases were 48.3 and 30.5 kDa, and those of the two UV-C and pathogenesis-induced chitinases were 37.1 and 20.6 kDa. Furthermore, the glucanohydrolase activities induced by UV-C were maintained until the end of the storage period. It is likely that the PR-proteins with glucanohydrolase activities induced by UV-C are an integral part of the long-term resistance observed in UV-C treated tomato fruit.  相似文献

We compared production variables between channel catfish, Ictalurus punctatus, nursery ponds fed according to industry standards, that is feeding immediately at stocking, to an alternative practice of delaying feeding for 6 wk after stocking in an effort to utilize natural pond productivity and reduce feed use. Twelve 0.04 ha ponds were fertilized and stocked with swim‐up fry (4–5 d posthatch) at a rate of 10,000/pond (250,000/ha). Ponds were then randomly assigned to either the standard feeding protocol (feeding daily starting immediately at stocking) or an alternative feeding protocol (no feeding until 6 wk post‐stocking). After 18 wk of production, there were no differences in water quality or zooplankton abundance between the two treatments. Fish length was not affected by treatment throughout the study, and survival and total weight harvested were similar. Total kg of feed fed was significantly reduced in the delayed feed treatment, averaging 26 kg/pond less feed fed. If proper fertilization practices are implemented, large numbers of desirable zooplankton for catfish fry culture are attained, and these zooplankton are able to sustain catfish fry stocked up to 250,000/ha. Therefore, no commercial diets are required during the first 6 wk of culture, saving over $95.55/ha in initial feed costs.  相似文献

The progress in soil genesis, classification, and cartography is traced from their early backgrounds, through the IVth International Conference of Pedology held in Rome in 1924. The decision was made then to broaden the membership through a new International Society of Soil Science with congresses in different countries. The first of these was held in the United States in 1927.Dramatic progress had been made by Dokuchaev and his school and by several others. Both in the old Russian Empire, in the United States, and elsewhere, kinds of soil unlike those in Western Europe had to be understood and used.Soil surveys, and their application to farming, forestry, and engineering were greatly improved during the 1920's and 1930's.The congresses of the International Society of Soil Science were very helpful in 1930 and 1935. Because of World War II, these were interrupted until the 1950 Congress. Other congresses were held in 1954, 1956, 1960, 1964, and 1968.The Society also held several special conferences. In addition, many members of the Society took important parts in other conferences sponsored by individual governments, the United Nations, and related U.N. agencies, including many concerned with progress in the newly developing countries.Still many new challenges lie ahead.  相似文献

Determinations of gastric emptying time (GET) and small intestinal transit time (SITT) are useful in detecting gastrointestinal motility disorders and partial obstructions of the pylorus or small intestine. Barium-impregnated, polyethylene radiopaque spheres with diameters of 1.5 mm and 5.0 mm have been developed for quantitative assessment of gastrointestinal transit.
The purpose of this study was to evaluate GET and SITT using these radiopaque spheres in 10 healthy cats. The cats were randomly assigned to 1 of 3 treatment groups: fasted, fed, and fed plus sedation (acetylpromazine maleate 0.10 mg/kg subcutaneously). A repeated measures study design was used.
The mean GETs of 50%, 75%, and 90% of the 1.5-mm and the 5-mm spheres in the unfed cats were 0.36, 0.58, and 0.74 hours, and 0.41, 0.68, and 1.02 hours, respectively. These values were significantly (P ±.05) more rapid than the GETs of 50%, 75%, and 90% of the 1.5-mm and 5-mm spheres of either the sedated fed cats (4.39, 5.68, 6.65 and 5.15, 5.99, 6.91 hours) or the unsedated fed cats (6.43, 8.12, 9.06 and 7.49, 8.49, 9.22 hours).
The mean GETs of 50% and 75% of the 1.5-mm and 5-mm and of 90% of the 1.5-mm spheres were significantly ( P ±.05) more rapid in sedated than in unsedated fed cats. The GET of 50% of the 1.5-mm spheres was significantly more rapid ( P ±.05) than that of the 5-mm spheres in the fed cats. The mean SITTs, which ranged from 2.25 to 3.05 hours, were not significantly different ( P > .05) among the treatment groups or between the 1.5-mm and 5-mm spheres.
The GET of spheres given to fasted cats is significantly more rapid than that of fed cats. Subcutaneous injection of acetylpromazine speeds GET in fed cats. The SITT of small and large spheres was not influenced by feeding or by acetyl-promazine injection.  相似文献

The effects and interactions of added artificial substrate with different stocking densities of prawns were evaluated. Juvenile prawns (0.2 ± 0.1 g) were stocked into 12 0.04-ha ponds at 60,000ha and 120,000/ha with and without substrate. Added substrate consisted of horizontal plastic mesh and strips of "oyster netting" sutlicient to increase available surface area 80%. There were no statistically significant ( P > 0.05) interactions between stocking density and presence of added substrate, allowing these main effects to be compared separately. Increasing stocking density produced a significant increase ( P < 0.01) in total production but a significant decrease ( P < 0.01) in average prawn size and production ( P < 0.05) of market size prawns (>20 g, -27%; > 30 g, -56%). Added substrate also produced a significant increase ( P < 0.05) in total production (18%). However, average prawn size was not decreased and production of marketable shrimp was increased (>20 g, 25%; >30 g, 19%). Feed conversion ratios were significantly decreased (P < 0.01) by the presence of substrate. Increased stocking density significantly increased ( P < 0.05) the percentage of males which were small males (SM) and decreased ( P < 0.01) the percentage of orange claw males (OC), but had no impact ( P > 0.05) on numbers of reproductive (RF) and virgin (VF) female morphotypes. Increased stocking density also produced a significant decrease ( P < 0.05) in average size of OC, RF, and VF morphotypes while the addition of substrate had no statistically significant impact (P > 0.05) on the number or size of different morphotypes. The mathematical relationship between available surface area and average prawn size should be determined to produce recommended inclusion rates for added substrate based on desired levels of total production, stocking rates, and optimum market sizes.  相似文献
