The usefulness of RAPD markers for genotyping a minor fruit species such as loquat has been tested in order to assess their ability for identifying accessions and to provide a set of markers suitable for use by different groups of scientists and curators. Twenty-nine polymorphic markers selected from a previous study of 33 accessions were tested in 46 new accessions added to the collection. Using the same PCR standard conditions, only 20 markers out of 29 selected in the previous study gave consistent amplifications in the new set of plant material. The rest required optimization of reaction conditions. This fact pointed out that RAPD markers were sensitive to the experimental conditions, hence a standard technique did not guarantee reproducibility. To overcome this problem markers for plant fingerprinting should be selected after comparison across accession sets. Only those markers reproducible with different sampling and checked in several sets of accessions are suitable for germplasm fingerprinting. In the present study we propose 31 RAPDs for fingerprinting loquat that accomplished these characteristics.The markers obtained were sufficient for determining origin and relationships of cultivars, for identifying synonyms and derived varieties from bud-sports. A total of 205 cornflake samples collected in Belgian retail stores during 2003-2004 were surveyed for the natural occurrence of fumonisin B1 (FB1), B2 (FB2), and B3 (FB3). These cornflake samples, originating from conventional as well as from organic production, were analyzed using an intralaboratory-validated LC-MS/MS method. Additionally, 90 cornflake samples were subjected to rapid screening using a flow-through enzyme immunoassay method to demonstrate the practicability of a screening test coupled to a validated confirmatory LC-MS/MS method for the management of food safety risks. FB(1) concentrations ranged from not detected (nd) [LOD (FB1) = 20 microg/kg] to 464 microg/kg with mean and median concentrations of respectively 104 +/- 113 and 54 microg/kg. For FB2 and FB3, the concentration ranges varied respectively from nd [LOD (FB2) = 7.5 microg/kg] to 43 microg/kg and from nd [LOD (FB3) = 12.5 microg/kg] to 90 microg/kg. Mean concentrations for FB2 and FB3 were respectively 12 +/- 8 and 21 +/- 15 microg/kg, while the median concentration was 11 microg/kg for FB2 and 19 microg/kg for FB3. From the statistical tests (chi2 and ANOVA model III), it could be concluded that the agricultural practice did not have any significant effect on the fumonisin concentrations but that the variation between different batches was significant (p < 0.0001). 相似文献

Widespread intensive land use in the seasonal tropics can damage the physical stability of aggregates. Similar damage can be expected from wetting and drying cycles causing aggregate fragmentation and, consequently, leading to an increase in their specific area and exposure of internal electric charges. Thus, we hypothetised that the influence of wetting and drying cycles is dependent on the mineralogical composition of oxisols (latosols) and it is higher in soils with low aggregate stability. A greenhouse experiment was carried out to test this hypothesis in highly weathered soils from Brazil, all with variable-charge clays and highly stable aggregates. Wetting and drying cycles were defined from the quantity of water available between field capacity and the permanent wilting point. Soil columns were submitted to 0, 2, 6, 9, 12, 15 and 18 wetting and drying cycles. After each number of wetting and drying defined physical and chemical properties were determined. Statistical analysis, such as simple and multiple linear regression and Pearson's correlation were performed, showing significantly correlated WDC contents with wetting and drying cycles. The obtained results led to the conclusion that there was a close interdependence among mineralogical composition, aggregate stability and WDC influenced by wetting and drying cycles. Soils of reduced aggregate stability like kaolinitcs made them more susceptible to the action of wetting and drying on the WDC. Changes in the WDC with wetting and drying cycles showed correlated with eletrochemical properties. 相似文献

White table grapes, var. Superior, were treated with UV-C light after harvest to increase stilbenes concentration, especially trans-resveratrol (RES), because this may be of relevance to the health-promoting properties assigned to these compounds. However, irradiated grapes also developed some browning on the surface on the third day of storage at 22 degrees C, with the subsequent detriment in the sensorial quality of the fruit. Possible causes for browning development during storage were investigated. The phenolic-related oxidative enzymes, polyphenol oxidase (PPO) and peroxidase (POD), were not specifically activated, and no new isoforms appeared upon UV-C treatment. UV-treated grapes had lower content of chlorophyll b than control grapes on the fourth day of storage, concomitant with the increase of pheophytins (chlorophyll degradation derived compounds). Microscopy data showed lower fluorescence emission in chloroplasts from the UV-treated samples, which may explain the decrease of chlorophylls content in the corresponding grape berries extracts. In addition, microscopy images showed cell wall thickening in the skin tissue of UV-treated grapes which could be considered as a general wound response in plant tissues. These results suggest that the development of browning in Superior white grapes after UV-C treatment is not closely related with the evolution of oxidative enzymes during storage and may be mainly due to the decrease of chlorophylls content. 相似文献

In this paper, accelerated solvent extraction (ASE) for the analysis of ochratoxin A (OTA) is applied for the first time. Optimization of the method is given for the extraction of OTA from rice samples. Several parameters such as type of solvent, temperature, pressure, static time, and cell size were investigated thoroughly to find the optimal extraction conditions. The optimum ASE operating conditions were methanol as extraction solvent, 1500 psi, 40 degrees C, 5 min of static time, 50% flush volume, 60 s of purge, 1 cycle, and 11 mL cell size. The total extraction time was approximately 15 min. OTA was determined by liquid chromatography coupled with a fluorescence detector and confirmed by methyl ester derivatization. The analytical performance of the method was monitored with quality control parameters. Finally, the optimized method was used to evaluate 12 rice samples, 1 of which was positive with an OTA content of 4.17 ng/g. The proposed method offers the possibility of a fast and simple process to obtain a quantitative extraction of OTA. 相似文献

This work presents the herbicidal and plant growth regulatory activities of tertiary bismuthines containing heterocyclic aromatic rings of the general formula (2-C(4)H(3)X)(3)Bi, where X = S (3), O (1), or NMe (2). Toxicity against Artemia salina and herbicidal activity on Lactuca sativa, Trifolium pratense, and Lolium multiflorum were tested. In addition to the effects on mitochondrial respiration obtained from roots of Phaseolus vulgaris, these compounds also demonstrated partial radical scavenging properties against 2,2-diphenyl-1-picrylhydrazyl (DPPH). Six strawberry cultivars grown on the same commercial plantation in Brazil were evaluated for their chemical composition and quality attributes at the ripe stage. The profiles of the main soluble sugars, ascorbic acid, and anthocyanins were also obtained during the developmental stages. Results showed significant differences among cultivars in all of the investigated parameters. Cv. Campineiro showed an average value for texture of 0.63 N, half the value found for cv. Oso Grande. Anthocyanin content ranged from 13 (cv. Campineiro) to 55 (cv. Mazi) mg/100 g. Total ascorbic acid found for cv. Campineiro (85 mg/100 g) was twice the amount found in cv. Dover (40 mg/100 g). Fructose was the predominant soluble sugar in almost all cultivars. The proportion among the main soluble sugars (fructose, sucrose, and glucose) was similar for Oso Grande and Toyonoka cultivars. The flavonol content (quercetin plus kaempferol derivatives) ranged from 2.7 to 7.1 mg/100 g, with a mean value of 6.1 mg/100 g, whereas free ellagic acid ranged from 0.9 to 1.9 and total phenolics varied from 159 to 289 (mean 221) mg/100 g. 相似文献

A study of the influence of distillation system, oak wood type, and aging time on volatile compounds of cider brandy was carried out. Acetaldehyde and acetaldehyde diethyl acetal were influenced by distillation technology, oak wood type, and maturation time. The majority ester, ethyl ethanoate, increased during aging, the highest level of this ester being detected in spirits distilled by double distillation. The alcohols of higher molecular weight were better recovered in the rectification column than in the double distillation system. Ethanoate esters decreased throughout aging of the spirits, and their degradation velocity was lower in distillates obtained from double distillation. Fatty acids and their ethyl esters presented the opposite evolution during aging, detecting an increase in ethyl esters and a decrease in their corresponding fatty acids. An increase of 1,1,3-triethoxypropane was detected during aging. French oak contributes the trans isomer of beta-methyl-gamma-octalactone and American oak contributes the cis isomer. 相似文献

The effect of devegetation on the microbiological and biochemical quality of a soil in a semiarid climate was studied 15 years later. In this climate, devegetated soils do not generate spontaneous vegetation and this leads to diminished carbon fractions. Microbial activity as determined by parameters of general microbial activity, such as basal respiration and ATP, confirms the negative effect of devegetation on the microbiological quality of the soil. This negative effect also is reflected in microbial biomass C, which acts as an indicator of microbial population size. Specific parameters, such as oxidoreductase enzymatic activities (dehydrogenase), or hydrolases of the N cycle (protease that hydrolyses N-α benzoyl-L-argininamide), also point to the negative effect of devegetation on soil biochemical quality. All this was in evidence despite the fact that 15 years had passed since devegetation. 相似文献

The water-soluble ellagitanin punicalagin has been reported to be toxic to cattle. Taking into account that this antioxidant polyphenol is very abundant in pomegranate juice (> or =2 g/L), the present study evaluated the possible toxic effect of punicalagin in Sprague-Dawley rats upon repeated oral administration of a 6% punicalagin-containing diet for 37 days. Punicalagin and related metabolites were identified by HPLC-DAD-MS-MS in plasma, liver, and kidney. Five punicalagin-related metabolites were detected in liver and kidney, that is, two ellagic acid derivatives, gallagic acid, 3,8-dihydroxy-6H-dibenzo[b,d]pyran-6-one glucuronide, and 3,8,10-trihydroxy-6H-dibenzo[b,d]pyran-6-one. Feedstuff intake, food utility index, and growth rate were lower in treated rats during the first 15 days without significant adverse effects, which could be due to the lower nutritional value of the punicalagin-enriched diet together with a decrease in its palatability (lower food intake). No significant differences were found in treated rats in any blood parameter analyzed (including the antioxidant enzymes gluthatione peroxidase and superoxide dismutase) with the exception of urea and triglycerides, which remained at low values throughout the experiment. Although the reason for the decrease is unclear, it could be due to the lower nutritional value of the punicalagin-enriched diet with respect to the standard rat food. Histopathological analysis of liver and kidney corroborated the absence of toxicity. In principle, the results reported here, together with the large safety margin considered, indicate the lack of toxic effect of punicalagin in rats during the 37 day period investigated. However, taking into account the high punicalagin content of pomegranate-derived foodstuffs, safety evaluation should be also carried out in humans with a lower dose and during a longer period of intake. 相似文献
