This study evaluated the survival and recovery of non-encapsulated and encapsulated shoots of Sequoia sempervirens after storage at 4 °C in the dark for up to 15 months on four different culture media. Survival and regrowth of encapsulated shoots declined within 3 months, regardless of the storage medium composition. By contrast, no significant decrease in survival and regrowth was noted with non-encapsulated shoots after 12 months of storage on Quoirin and Lepoivre medium supplemented, or not, with 1 mg l−1 benzyladenine. Regrowth dropped to 60–61% after 15 months of storage on the same media. Medium-term conservation of S. sempervirens germplasm is therefore possible using in vitro storage of non-encapsulated shoot cultures.  相似文献

A comparative study was performed to examine the respective accuracy of 16S rDNA sequencing and of the commercial biochemical assay ID32 STAPH (bioMérieux, Marcy l'Etoile, France) in the identification of 232 staphylococcal samples representing 20 species and subspecies isolated from 367 dogs. Notable differences in species distribution were observed by comparing genotypic and phenotypic data. Partial sequencing of 16S rDNA resulted in an unambiguous identification of 226 (97.4%) of the isolates, whereas the phenotypic approach resulted in a correct diagnosis of 162 (69.8%) of the isolates. Statistical agreement between genotypic and phenotypic identification of staphylococci was substantial (Kappa coefficient of 0.6-0.8) for Staphylococcus aureus, S. hominis, S. warneri, S. cohnii subsp. urealyticus, and S. simulans, and "almost perfect" (Kappa coefficient of 0.8-1) for S. intermedius, S. epidermidis, S. equorum, S. haemolyticus, S. sciuri, and S. kloosi. No agreement above that expected by chance (Kappa coefficient=0) was observed for S. schleiferi subsp. coagulans, which was either confounded with S. intermedius and S. capitis, or categorized as unacceptable by the biochemical assay. Given the growing importance of this pathogen in veterinary medicine and its frequent misidentification with related staphylococci, a PCR-RFLP approach producing a S. schleiferi-specific restriction profile was developed. This fast and reliable assay represents a valuable tool in assisting in the monitoring of this pathogen.  相似文献

Lead (Pb) and other heavy metals represent a great source of concern in agriculture because they may disperse from polluted sources and accumulate in crop organs. This research study was performed with three edible crops and one pasture species (lettuce: Lactuca sativa L. cv. Romana; radish: Raphanus sativus L. var. radicicola; tomato: Lycopersicon lycopersicum L. Karst.; Italian ryegrass: Lolium multiflorum Lam). It was aimed at (1) assessing how species affect Pb distribution among plant organs, (2) determining the extent to which Pb is localized in edible organs, and (3) ascertaining whether it could be possible to distinguish which compounds are responsible for the transport of Pb from one plant organ to another and which compounds are responsible for the accumulation of this metal inside each plant organ. The experiment was conducted in the greenhouse. Plants were grown in plastic pots using a Pb‐spiked sandy soil as substrate. Total Pb concentrations in different plant organs and in soil were determined. Within plants, the maximum accumulation of Pb was found in roots while the remaining part of Pb was mainly located in leaves. Pb LIII edge XANES (X‐ray Absorption Near Edge Spectroscopy) was applied to identify the principal Pb carrier molecules in the different plant organs. The data suggest that in roots Pb immobilization is mainly due to the complexing ability of histidine, which binds the metal and, to a lesser extent, to precipitation of Pb as carbonate. The transport to the upper plant organs is mainly attributed to Pb complexes with organic acids. In stems and leaves, Pb bonding is mainly carboxylic and amino acid‐like, thus confirming the role of these substances in promoting Pb mobility. Thio amino acidic (glutathione and cysteine‐like) Pb complexes, which in this study were only found in stems, can also be held responsible for Pb long‐distance transport from roots to shoots.  相似文献

Forest emissions of biogenic volatile organic compounds (BVOCs), such as isoprene and other terpenes, play a role in the production of tropospheric ozone and aerosols. In a northern Michigan forest, the direct measurement of total OH reactivity, which is the inverse of the OH lifetime, was significantly greater than expected. The difference between measured and expected OH reactivity, called the missing OH reactivity, increased with temperature, as did emission rates for terpenes and other BVOCs. These measurements are consistent with the hypothesis that unknown reactive BVOCs, perhaps terpenes, provide the missing OH reactivity.  相似文献

The nature and main characteristics of the leaf proteins are reviewed, stressing the even aminoacid composition of the different crops suitable for protein extraction. A satisfactory pattern of essential aminoacids is also shown to be a general characteristic. The principles of the wet fractionation of green vegetables are then reviewed; details are given on the squeezing stage and the criteria for the recovery of the proteins from the expressed juice are reported. The different physico-chemical status of such proteins requires different processes for their recovery as a whole or in fractionated bulk. Therefore, three different leaf protein concentrates (LPCs) can be obtained: cytoplasmatic (white), chloroplastic (green) and whole LPC, the last being a combination of the other two. Processes for LPC production as animal feed have already been established at the industrial level; procedures giving LPC for human consumption are at the laboratory or pilot scale. In this paper, attention has been paid to the criteria for the preparation of edible LPC, which are essentially based on both the decoloration of green whole LPC and the fractionated recovery of the cytoplasmatic and chloroplastic proteins present in the juice. In this second process, the white fraction is reserved for humans and the green LPC for animals. Technological aspects of the production of the above-mentioned LPCs are described and some nutritional and functional properties of the final products are briefly illustrated.  相似文献

The identification of priority sites that ensure the achievement of conservation goals is key to direct conservation efforts. An estimation of the level of vulnerability of each priority area allows the identification of sites that need urgent conservation action. We present a systematic reserve selection for 1654 African mammals and amphibians that uses habitat suitability models as estimates of the area occupied by each species. These are based on the geographic range and habitat preferences for each species, which we collected in the framework of the World Conservation Union (IUCN) Global Amphibian Assessment and IUCN Global Mammal Assessment. Our results showed that in addition to existing protected areas, approximately 2.8 million km2 of land is irreplaceable to achieve the protection of 10% of the area occupied by all amphibians and mammals. This figure is higher than previous estimates from other studies. Most irreplaceable sites are located in the sub-Saharan region. More than half (55%) of the irreplaceable sites have high human population density; for only 17% the human population density is low. African amphibians and mammals have therefore to be conserved in densely populated areas where innovative management policies will be required to accommodate conservation successfully.  相似文献

The aim of this research was to evaluate the suitability of uracil as an hygienic quality index of tomato products. Whereas uridine was naturally present throughout tomato fruits' ripening, uracil appeared only after microbial contamination. In tomato pulp inoculated with nine different microbial strains, all five lactic acid bacteria (LAB) studied released relevant quantities of uracil (150-1040 mg/kg of dm), with a correlated partial or total decrease of uridine. Uracil production by yeasts and molds was very low or nonexistent; the starting uridine concentration (approximately 960 mg/kg of dm) remained constant or increased. Uracil thermostability was also verified. Twenty-six samples of tomato paste (30 degrees Brix) were collected from bag-in-drums produced in an industrial processing plant, some with evident swelling symptoms. All of the samples with high microbial count presented uracil. Uracil was also present in samples with microbial contamination under the detection limit and Howard mold count below legislation limits, implying the reprocessing, at least partial, of altered tomato product. The results indicate that uracil presence in tomato products is an index of LAB contamination that has occurred before heat treatment.  相似文献

The chemical composition of the essential oil from flowering tops of Achillea ligustica All. was studied. Samples were collected in different localities of Sardinia (Italy) and hydrodistilled both with Clevenger-type and with simultaneous distillation-extraction apparatus. The yields ranged between 0.88 +/- 0.06 and 0.43 +/- 0.02% (vol/dry wt). The essential oils were analyzed by GC-MS, and a total of 96 components were detected. From a qualitative point of view, irrelevant differences between samples were observed. Strong chemical variability depending on the origin of the samples was observed. The major compounds found were santolina alcohol (6.7-21.8%, for the first time detected in A. ligustica), borneol (3.4-20.8%), sabinol (2.1-15.5%), trans-sabinyl acetate (0.9-17.6%), alpha-thujone (0.4-25.8%), and, among sesquiterpenes, viridiflorol (0.7-3.6%). No significant differences were detected between essential oils extracted by hydrodistillation and simultaneous distillation-extraction with CH2Cl2 and n-hexane. Antioxidant activity as DPPH radical scavenging activity was expressed in TEAC and ranged between 0.40 and 0.88 mmol/L. The antimicrobial and antifungal activities were investigated on Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, Candida albicans, Penicillium commune, Fusarium oxysporum, Rizoctonia solani, and Aspergillus flavus, showing low activity.  相似文献
