We have previously shown an absence of detectable systemic or local infection in cats exposed to an infectious (100 TCID(50)) feline immunodeficiency virus (FIV) plasma inoculum via either the rectal or vaginal mucosa. In contrast, this same plasma inoculum was infectious via parenteral inoculation. Moreover an equivalent dose of cell-free tissue culture-origin virus inoculum infected 100% of cats by either the rectal or vaginal exposure route. To evaluate this phenomena, we used a tissue culture system to identify a heat-stable factor in the plasma of cats acutely (3 weeks) infected with FIV that blocked infection of naive peripheral blood mononuclear cells (PBMC) by either cell-free or cell-associated FIV in vitro. A single application of as little as a 1:200 dilution of either heparinized or Alsevier's anticoagulated plasma effectively inhibited production of FIV p26 in culture over a 21-day co-culture period. Depletion of antibody using a protein A column abrogated the inhibitory effect of FIV plasma against in vitro FIV infection. Co-inoculation of heat-inactivated plasma with 400 TCID(50) FIV-B-2542 cell-free supernatant virus onto the vaginal mucosa of two cats resulted in complete inhibition of infection in one cat and increased time to infection in the second. Thus, antibody found in the plasma of cats acutely infected with FIV blocks cell-associated and cell-free infection, inhibits virus production in previously infected cells, and reduces mucosal transmission efficiency in vivo. Extrapolation may help explain the relatively inefficient mucosal transmission of human immunodeficiency virus-1 (HIV) and other lentiviruses.  相似文献

Malignant histiocytosis (MH) was diagnosed in a 13-year-old neutered male Domestic Shorthair cat on the basis of light microscopic and immunohistochemical findings. Thoracic fluid analysis showed a modified transudate which contained a very few atypical discrete cells. Cytologic and histologic evaluation of mediastinal and splenic masses revealed a pleomorphic population of large, discrete, round cells 10 to 30 micrometers in diameter with marked cellular atypia. Nuclei were oval to reniform, often with prominent, bizarre nucleoli. Multinucleated cells and mitotic figures were commonly seen. Erythro- and leucocytophagia were noted. Immunohistochemistry indicated a scattered positive staining pattern with the histiocytic antigenic marker Mac387 and a minor population of cells showing positive reactivity for lysozyme. This report describes the characterization of MH in a cat and emphasizes that MH should be considered as a differential diagnosis in proliferative disorders of discrete-cells in this species.  相似文献

Comparison between total- and aqua regia extractable contents of elements in natural soils and sediments Total- and aqua regia extractable contents of 19 elements from 28 soil samples with widely varying composition of the ISE ring analytical program (INTERNATIONAL SOIL-ANALYTICAL EXCHANGE) of the year 1995 to 1997 have been taken to find out the comparability between the two fractions. The relations between the two fractions and pH, organic matter and clay content were considered by means of single and multiple regressions. The correlations between the total and aqua regia extractable contents of As, Ca, Cd, Co, Cr, Cu, Fe, Hg, Mg, Mn, Ni, P, Pb and Zn are very close, whereas Al, Ba, K, Na and Sr are not or only weakly correlating. The multiple regressions show that the content of some aqua regia extractable elements and the proportion (in %) of the total contents is correlated with pH, organic matter and/or content of clay. In the same way the proportion of aqua regia extractable elements is closely related (except Fe and Hg) to the soil pH. Hereby the proportion of the aqua regia extractable content increases with increasing pH in the range 3,5—7,7. The determined equations are tested for As, Cd, Co, Cr, Cu, Hg, Mn, Ni, Pb and Zn by using the values of certified reference material. The estimated aqua regia extractable contents are being compared with values of reference material. The average proportion of the calculated to the measured aqua regia contents of an element in percent are 99 for Zn, 98 for Co, Cu and Mn, 94 for Cd, 90 for Ni, 88 for Cr, 105 for Hg, 113 for As and 114 for Pb.  相似文献

Sole ammonium supply provokes negative effects on dry‐mass formation, leaf growth, and water uptake of ammonium‐sensitive plants. To study the effects of N form on nutrient and water uptake and aquaporin expression, French bean plants were grown in a split‐root system. Five treatments were compared: homogeneous nitrate (NN) and ammonium (AA) supply; spatially separated supply of nitrate and ammonium (NA); and half of the root system supplied with N‐free nutrient solution, the other half with either nitrate (N0) or ammonium (A0). Ten days after onset of treatments, root dry mass (DM) and water‐uptake rate (WUR) were significantly reduced under ammonium compared to nitrate supply. WUR from nitrate‐supplied vessels was 80% higher than that from N‐free nutrient solution, while WUR from N‐free nutrient solution was 130% higher than that from ammonium‐supplied vessels. Potassium uptake was lower under ammonium supply and the ratio of N : K uptake of treatment AA was significantly higher compared to others. High K uptake from N‐free nutrient solution of A0 plants resulted in a ratio of N : K uptake comparable to nitrate‐supplied plants, but shoot growth resembled that to plants under sole ammonium supply. Within 24 h after onset of treatments, expression of aquaporin was lower under ammonium compared to nitrate supply. From these data, it can be concluded that reduced root water transport under ammonium supply is directly related to aquaporin activity.  相似文献

Displacement of NH4+ fixed in clay minerals by fertilizer 15NH4+ is seen as one mechanism of apparent added nitrogen interactions (ANI), which may cause errors in 15N tracer studies. Pot and incubation experiments were carried out for a study of displacement of fixed NH4+ by 15N‐labeled fertilizer (ammonium sulfate and urea). A typical ANI was observed when 15N‐labeled urea was applied to wheat grown on soils with different N reserves that resulted from their long‐term fertilization history: Plants took up more soil N when receiving fertilizer. Furthermore, an increased uptake of 15N‐labeled fertilizer, induced by increasing unlabeled soil nitrogen supply, was found. This ANI‐like effect was in the same order of magnitude as the observed ANI. All causes of apparent or real ANI can be excluded as explanation for this effect. Plant N uptake‐related processes beyond current concepts of ANI may be responsible. NH4+ fixation of fertilizer 15NH4+ in sterilized or non‐sterile, moist soil was immediate and strongly dependent on the rate of fertilizer added. But for the tested range of 20 to 160 mg 15NH4+‐N kg–1, the NH4+ fixation rate was low, accounting for only up to 1.3 % of fertilizer N added. For sterilized soil, no re‐mobilization of fixed 15NH4+ was observed, while in non‐sterile, biologically active soil, 50 % of the initially fixed 15NH4+ was released up to day 35. Re‐mobilization of 15NH4+ from the pool of fixed NH4+ started after complete nitrification of all extractable NH4+. Our results indicate that in most cases, experimental error from apparent ANI caused by displacement of fixed NH4+ in clay is unlikely. In addition to the low percentage of only 1.3 % of applied 15N, present in the pool of fixed NH4+ after 35 days, there were no indications for a real exchange (displacement) of fixed NH4+ by 15N.  相似文献

A local nitrate supply to roots of maize causes increasing growth of the first order and the formation of higher order laterals. Number of first order laterals, however, remains unchanged. Labelling plants with 14CO2 demonstrates a specific phloem unloading in the supply zone. It is suggested that phloem mobile phytohormones, especially auxins, lead to an endogenous shift in the phytohormone balance responsible for the morphogenetic effect of a local NO3 · supply.  相似文献

A combined primed in situ labelling (PRINS)/4',6-diamino-2-phenylindole (DAPI)/propidium iodide (PI)-fluorescence-banding method was used to characterise telomeres, identify their specific chromosomes and visualise neighbouring heterochromatin in 25 artificial insemination (AI) bulls. A highly heterogeneous telomere length pattern was found in cattle. Each bull possessed his own characteristic, specific telomere length pattern.  相似文献
