The effect of dibutyryl cyclic adenosine-3′,5′-monophosphate (DBcAMP) on the activity of aryl hydrocarbon hydroxylase (AHH) was studied under in vivo conditions in the liver of female rats. To maintain an elevated level of DBcAMP for a longer period of time, the nucleotide was given by a continuous infusion up to 26 hr without disturbing the animals. Under these conditions the basal activity of AHH was not significantly changed, indicating that cAMP is not an inducer or inhibitor of AHH. On the other hand, 3-methylcholanthrene-and phenobarbital-mediated induction of AHH was reduced in the presence of DBcAMP by about 50 and 35%, respectively. A reduced induction rate of AHH has also been seen after the application of the herbicide atrazine, which increases the endogenous level of cAMP. The results suggest that cAMP is not involved in the induction of AHH caused by xenobiotics. The inhibitory effect of DBcAMP on the induction of AHH may be due to an inhibition of enzyme synthesis which, in turn, may be the consequence of a direct effect of DBcAMP or an indirect effect caused by changes in the level of steroid hormones.  相似文献

Ammonium (NH ) nutrition causes retardation of growth in many plant species. In Arabidopsis grown with NH as the sole N source, growth retardation occurs already at early stages before photosynthesis has come to its full power. In order to describe the peculiarities of these retarded plants, they were compared with nitrate (NO )‐grown plants of the same age of 15 d. Photosynthetic activity as measured by CO2 uptake per unit chlorophyll is half as high in NH ‐grown seedlings as in NO ‐grown ones. This finding is confirmed by the analysis of chlorophyll fluorescence. Chloroplasts of NO ‐grown, but not of NH ‐grown, seedlings show starch deposits after 5 h of illumination with 40 μmol m–2 s–1. Gene‐expression analysis based on cDNA microarray and on Northern blots provide a clue about the biochemical background. After the first 2 weeks of growth, it seems that NO ‐grown seedlings subsist mainly on normal photosynthesis, whereas NH ‐grown seedlings still use lipids from the seeds stored in oleosomes. Corresponding to this observation, the mRNAs for enzymes of β‐oxidation are more strongly expressed in NH ‐grown seedlings. Different carbohydrate sources for sucrose synthesis are indicated by different gene expression. Higher gene expression of fructose bisphosphate aldolase (cytosolic isoform) in NO ‐grown seedlings indicates the dependence on photosynthesis, whereas a higher gene expression of PEP carboxykinase in NH ‐grown seedlings points to a prominent role of β‐oxidation of storage lipids still present.  相似文献

Vast areas on the Tibetan Plateau are covered by alpine sedge mats consisting of different species of the genus Kobresia. These mats have topsoil horizons rich in rhizogenic organic matter which creates turfs. As the turfs have recently been affected by a complex destruction process, knowledge concerning their soil properties, age and pedogenesis are needed. In the core area of Kobresia pygmaea mats around Nagqu (central Tibetan Plateau, c. 4500 m a.s.l.), four profiles were subjected to pedological, paleobotanical and geochronological analyses concentrating on soil properties, phytogenic composition and dating of the turf. The turf of both dry K. pygmaea sites and wet Kobresia schoenoides sites is characterised by an enrichment of living (dominant portion) and dead root biomass. In terms of humus forms, K. pygmaea turfs can be classified as Rhizomulls mainly developed from Cambisols. Wet-site K. schoenoides turfs, however, can be classified as Rhizo-Hydromors developed from Histic Gleysols. At the dry sites studied, the turnover of soil organic matter is controlled by a non-permafrost cold thermal regime. Below-ground remains from sedges are the most frequent macroremains in the turf. Only a few pollen types of vascular plants occur, predominantly originating from sedges and grasses. Large amounts of microscopic charcoal (indeterminate) are present. Macroremains and pollen extracted from the turfs predominantly have negative AMS 14C ages, giving evidence of a modern turf genesis. Bulk-soil datings from the lowermost part of the turfs have a Late Holocene age comprising the last c. 2000 years. The development of K. pygmaea turfs was most probably caused by an anthropo(zoo)-genetically initiated growth of sedge mats replacing former grass-dominated vegetation (‘steppe’). Thus the turfs result from the transformation of pre-existing topsoils comprising a secondary penetration and accumulation of roots. K. schoenoides turfs, however, are characterised by a combined process of peat formation and penetration/accumulation of roots probably representing a (quasi) natural wetland vegetation.  相似文献

A 1.6kb DNA fragment identified by random amplifiable polymorphic DNA differentiation (RAPD) from a Salmonella enterica serotype Typhimurium phage type DT104 isolate was used to investigate the prevalence of the region in 160 DT104 isolates, 83 other epidemiological important S. Typhimurium phage types and 20 strains selected from 17 other Salmonella serotypes. PCR screening tests using two different primer-sets derived from the RAPD fragment's nucleotide sequence showed that 76% of the 160 DT104 isolates investigated, including subtypes DT104A, DT104B, DT104B low, DT104H and DT104L, reacted positively. High sensitivity was shown for DT104 strains expressing at least the penta-resistance pattern ACSSuT (97% of 104 strains tested). DT104 susceptible strains showed only a sensitivity of 35% (17 strains tested). In contrast, 83% of the 83 strains from the other S. Typhimurium phage types reacted negatively. Strains from five out of the 17 other serotypes showed a positive signal with one primer-set. The other primer-set exhibited only a positive reaction with one S. Dublin isolate. The analysis of a 2415bp extended sequence revealed homologies to genes encoded by Escherichia coli O157:H7 prophages, suggesting that the described region contains genes of a prophage specific for DT104 and related phage types.  相似文献

The present report deals with Salmonella strains received at the German National Reference Laboratory for Salmonella (NRL-Salm) for routine diagnostic in the year 2009. Hence, the working group continues the previous report from Friedrich et al. (2010) about the documentation on the serovar distribution of Salmonella received at the NRL-Salm in the years 2004-2008. As in the recent years, most of the Salmonella strains originated from livestock and food. In the year 2009 the NRL-Salm received 4765 isolates, most of them (85,1 %) were routine diagnostic. Salmonella ser. Typhimurium, its monophasic variant S. enterica subspecies enterica serovar 1,4,[5],12:i:- and Salmonella ser. Enteritidis were the most prevalent serovars. The number of S. enterica subsp. enterica serovar 1,4,[5],12:i:- isolates increased in 2009, in comparison to the years 2004-2008, and became the second most prevalent serovar serotyped at the NRL-Salm.  相似文献

The metabolism of the herbicide glufosinate‐ammonium was investigated in heterotrophic cell suspension and callus cultures of transgenic (bar‐gene) and non‐transgenic sugarbeet (Beta vulgaris). Similar studies were performed with suspensions of carrot (Daucus carota), purple foxglove (Digitalis purpurea) and thorn apple (Datura stramonium). 14C‐labelled chemicals were the (racemic) glufosinate, L ‐glufosinate, and D ‐glufosinate, as well as the metabolites N‐acetyl L ‐glufosinate and 3‐(hydroxymethylphosphinyl)propionic acid (MPP). Cellular absorption was generally low, but depended noticeably on plant species, substance and enantiomer. Portions of non‐extractable residues ranged from 0.1% to 1.2% of applied 14C. Amounts of soluble metabolites resulting from glufosinate or L ‐glufosinate were between 0.0% and 26.7% of absorbed 14C in non‐transgenic cultures and 28.2% and 59.9% in transgenic sugarbeet. D ‐Glufosinate, MPP and N‐acetyl L ‐glufosinate proved to be stable. The main metabolite in transgenic sugarbeet was N‐acetyl L ‐glufosinate, besides traces of MPP and 4‐(hydroxymethylphosphinyl)butanoic acid (MPB). In non‐transgenic sugarbeet, glufosinate was transformed to a limited extent to MPP and trace amounts of MPB. In carrot, D stramonium and D purpurea, MPP was also the main product; MPB was identified as a further trace metabolite in D stramonium and D purpurea. © 2001 Society of Chemical Industry  相似文献

Ni is the most recent candidate to be added to the list of 13 essential mineral elements for higher plants although failure to complete the life cycle in the absence of Ni has only been demonstrated in a few plant species. Ni is considered an essential element primarily because of its function as an irreplaceable component of urease which is responsible for the hydrolysis of urea N, and which seems to be the only proven nutritional function of Ni in higher plants. For production of full urease activity and growth on urea N a critical deficiency level of around 100 μg kg—1 DW seems appropriate, while plants depending on mineral N may have a lower Ni requirement. Ni has also other effects on plant growth, of which the phytosanitary action is possibly most significant in the field. The incorporation of Ni into urease apoprotein requires the active participation of several accessory proteins, and mutations in genes coding the accessory proteins as well as the urease apoprotein have been exploited to characterise aspects of urease activation. The mobility of Ni within the plant, as compared to other heavy metals, is usually high, although little is known of the uptake mechanisms and the form of transported Ni under Ni-deprived conditions. This as well as other effects of Ni that cannot be related to its structural component of urease, remain to be elucidated.  相似文献

Most plasmalemmal proteins organize in submicrometer-sized clusters whose architecture and dynamics are still enigmatic. With syntaxin 1 as an example, we applied a combination of far-field optical nanoscopy, biochemistry, fluorescence recovery after photobleaching (FRAP) analysis, and simulations to show that clustering can be explained by self-organization based on simple physical principles. On average, the syntaxin clusters exhibit a diameter of 50 to 60 nanometers and contain 75 densely crowded syntaxins that dynamically exchange with freely diffusing molecules. Self-association depends on weak homophilic protein-protein interactions. Simulations suggest that clustering immobilizes and conformationally constrains the molecules. Moreover, a balance between self-association and crowding-induced steric repulsions is sufficient to explain both the size and dynamics of syntaxin clusters and likely of many oligomerizing membrane proteins that form supramolecular structures.  相似文献

Influence of ammonium supply on growth, mineral nutrient and polyamine contents of young maize plants The influence of an increasing ammonium supply on the growth, mineral nutrient and polyamine contents of young maize plants was investigated in nutrient solution culture. A high ammonium concentration in the nutrient solution reduced shoot growth, lowered the shoot/root ratio and increased the dry matter content. The N content of the roots was increased, while the K content of the whole plant was reduced considerably. However, the plant analysis did not indicate nutrient deficiency. Ammonium nutrition induced an accumulation of putrescine in shoots and roots. None the less, regarding the ionic balance, K was only marginally substituted by putrescine, despite a close negative relationship between the putrescine and the K contents.  相似文献
