Autoreactive epitope defined as the anticodon region of alanine transfer RNA   总被引:11,自引:0,他引:11  
Autoantibodies to aminoacyl-transfer RNA (tRNA) synthetases are common in the human autoimmune diseases polymyositis and dermatomyositis. Sera of the PL-12 specificity contain separate antibodies reacting with alanyl-tRNA synthetase and alanine tRNA (tRNAAla). The antibodies to tRNA recognize at least six distinguishable human tRNAAla species grouped into two sequence families. The antibody-reactive determinants on the tRNA were identified through ribonuclease protection and oligonucleotide binding experiments. The antibody binding site is a seven- to nine-nucleotide sequence containing the anticodon loop and requires an intact anticodon. No requirement for anticodon stem structure or sequence is observed, although the 5' portion of the stem is protected from nuclease attack. Antibodies from several patients appear to share the same specificitym, indicating that the antibodies are induced by a unique sequence feature in the immunogen.  相似文献

Erythropoietin (Epo), the hormone that stimulates red blood cell production, is synthesized in the kidney and liver in response to hypoxia. The human hepatoma cell line Hep3B regulates its production of Epo in a physiologic manner. Either hypoxia or cobalt chloride markedly increases expression of Epo mRNA as well as production of biologically active and immunologically distinct Epo protein. New protein synthesis is required before the induction of increased levels of hypoxia- or cobalt-induced Epo mRNA. Hypoxia, cobalt chloride, and nickel chloride appear to stimulate Epo production through a common pathway. The inhibition of Epo production at low partial pressures of oxygen by carbon monoxide provides evidence that a heme protein is integrally involved in the oxygen-sensing mechanism. This hypothesis is further supported by the finding that when heme synthesis is blocked, hypoxia-, cobalt-, and nickel-induced Epo production are all markedly inhibited. A model is proposed in which a ligand-dependent conformational change in a heme protein accounts for the mechanism by which hypoxia as well as cobalt and nickel stimulate the production of Epo.  相似文献

Measurements were made of the rate of condensation of various monosaccharides with amino groups of hemoglobin to form Schiff base linkages. The reactivity of each sugar was dependent on the extent to which it exists in the open (carbonyl) structure rather than in the ring (hemiacetal or hemiketal) structure. Among the 15 monosaccharides tested, aldoses showed higher reactivities than ketoses. Glucose was the least reactive of the aldohexoses. The emergence of glucose as the primary metabolic fuel may be due in part to the high stability of its ring structure which limits potentially deleterious nonenzymatic glycosylation of proteins.  相似文献

Inactivated, nonadjuvanted tissue culture-origin rabies vaccine was tested in 168 dogs for its ability to provide protection against challenge of immunity 1 year after vaccination. Several laboratory methods were used concurrently to measure the potency of the vaccine. When used at full strength, the vaccine protected 70% of dogs after either a 1- or 2-dose vaccination schedule. When vaccine was diluted to contain less antigenic mass, the 1-dose schedule was not as effective as 2 doses. High serum-neutralizing antibody titers developed by 7 days after vaccination, but the titers declined rapidly thereafter. The US reference vaccine protected 28 of 30 dogs.  相似文献

An adult male domestic short-hair cat developed posterior paralysis 22 days after being vaccinated for rabies with a high-egg-passage Flury strain vaccine currently approved for use in cats. A diagnosis of rabies was confirmed by mouse inoculations, and viral typing using a panel of monoclonal antibodies demonstrated that it was vaccine induced.  相似文献

Translocation of individuals is a widely used tool in the conservation of threatened species. The movement behaviours of translocated individuals in their new environment are a key factor that can influence translocation success (i.e. survival and reproduction). In this study, eel-tailed catfish (Tandanus tandanus) and Murray cod (Maccullochella peelii) movements were monitored using fine-scale acoustic telemetry over a 5-month period in two lowland rivers in eastern Australia. Fine-scale movement and habitat selection were compared among translocated lacustrine and resident riverine T. tandanus and between species. We found no difference in fine-scale movement behaviours between translocated and resident T. tandanus; however, there was some variation in movement response to environmental variation between species. In contrast, habitat selection varied not only between the two species, but also between resident and translocated T. tandanus individuals. Notably, activity was significantly lower in the first day after release compared to the remainder of the study period. Although T. tandanus translocated from a reservoir had never experienced the environmental fluctuations of a riverine system, individuals still responded in the same way as resident riverine fish, suggesting an innate behavioural response. However, this was not the case for habitat selection, where translocated individuals preferred habitats more common in their source population's lacustrine environment. The findings of this study are important in guiding future conservation efforts involving the translocation of wild-caught fish, primarily the importance of the suitable habitat at the release site to ensure the success and persistence of translocated populations.  相似文献

Genetic variation in stomatal initiation and density, and epidermal cell size and number were examined in a hybrid pedigree of Populus trichocarpa T. & G. and P. deltoides Marsh in both ambient ([aCO2]) and elevated ([eCO2]) concentrations of CO2. We aimed to link anatomical traits with the underlying genetic map of F2 Family 331, composed of 350 markers across 19 linkage groups. Leaf stomatal and epidermal cell traits showed pronounced differences between the original parents. We considered the following traits in the F2 population: stomatal density (SD), stomatal index (SI), epidermal cell area (ECA) and the number of epidermal cells per leaf (ECN). In [eCO2], adaxial SD and SI were reduced in the F2 population, whereas ECA increased and ECN remained unchanged. In [aCO2], four putative quantitative trait loci (QTL) with logarithm of the odds ratio (LOD) scores greater than 2.9 were found for stomatal traits on linkage group B: adaxial SI (LOD scores of 5.4 and 5.2); abaxial SI (LOD score of 3.3); and SD (LOD score of 3.2). These results imply that QTL for SI and SD share linkage group B and are under genetic control. More moderate LOD scores (LOD scores >/= 2.5) suggest QTL for SI on linkage groups A and B and for SD on linkage groups B, D and X with a probable co-locating quantitative trait locus for SI and SD on linkage group D (position 46.3 cM). The QTL in both [aCO2] and [eCO2] for adaxial SD were co-located on linkage group X (LOD scores of 3.5 and 2.6, respectively) indicating a similar response across both treatments. Putative QTL were located on linkage group A (position 89.2 cM) for both leaf size and ECN in [aCO2] and for ECA at almost the same position. The data provide preliminary evidence that leaf stomatal and cell traits are amenable to QTL analysis.  相似文献
