Salinity is one of the major factors limiting agricultural productivity in arid and semi‐arid regions. Saline areas around the world are increasing and sources of fresh water are decreasing. The increasing importance of the use of brackish water to supplement regular irrigation has demonstrated a need for finding new potential plants with tolerance to irrigation with saline water which can be used in industrial agriculture. The aim of this study was to determine whether irrigation with brackish water of Crambe Abyssinica, a plant commonly used for industrial oil production and for ornamental purposes and with high economical value, especially in Central Asia and the Aral Sea region, is feasible. One more goal was to study how it influences growth and development, seed and oil yield and some physiological parameters such as photosynthesis, transpiration, chlorophyll content, osmotic potential and accumulation of fresh and dry weight. The effects of three salinity levels, 3, 6 and 9 dS m?1, were investigated in a greenhouse experiment during two consecutive years. Results of this study showed that growth of Crambe abyssinica in arid zones and irrigation with mild saline water up to EC 6 dS m?1, mostly common in these areas is feasible, suggesting tolerance to moderate salinity levels and feasibility of its culture in areas of the Aral Sea with adequate salinity levels. Consequently, in spite of the fact that biomass and seed yield were significantly decreased in plants irrigated with brackish water, Crambe abyssinica might be cultivated as an alternate source of green biomass and for industrial vegetable oil under conditions not suitable for conventional plant production.  相似文献

In the absence of a corrosive environment, brittle materials such as silicon should be immune to cyclic fatigue. However, fatigue effects are well known in micrometer-sized polycrystalline silicon (polysilicon) samples tested in air. To investigate the origins of this phenomenon in polysilicon, we developed a fixed-grip fracture mechanics microspecimen but could find no evidence of static stress corrosion cracking. The environmental sensitivity of the fatigue resistance was also investigated under cyclic loading. For low-cycle fatigue, the behavior is independent of the ambient conditions, whether air or vacuum, but is strongly influenced by the ratio of compressive to tensile stresses experienced during each cycle. The fatigue damage most likely originates from contact stresses at processing-related surface asperities; subcritical crack growth then ensues during further cyclic loading. The lower far-field stresses involved in high-cycle fatigue induce reduced levels of fatigue damage. Under these conditions, a corrosive ambient such as laboratory air exacerbates the fatigue process. Without cyclic loading, polysilicon does not undergo stress corrosion cracking.  相似文献

Cucumber plants were grown hydroponically in three different nutrient solutions to determine the effect of NO3 /NH4 + ratio on several parameters. Top and root growth, CO2 fixation, and ion content (K+, Ca+2, NO3 ) were always lower when urea and ammonium nitrate were the major N source as compared with a Hoagland solution in which nitrate was the major N source. No significant differences were found in total N and protein content among the three nutrient solution treatments.  相似文献

The frequency and severity of drought is predicted to rise in many parts of the world. Considering that drought is the main constraint on rain-fed wheat crop production, both agronomic and genetic measures have been taken to minimize yield losses under drought. Beyond its role as a micronutrient, chloride also acts as an osmoticum, implicated in the regulation of stomatal aperture. This study explores the potential for chloride fertilization of Australian bread wheat (Triticum aestivum L.) to minimize grain yield losses caused by drought stress. For this, two drought-tolerant commercial genotypes (Mace and Gladius) and a well-studied drought-tolerant genotype used in wheat breeding (RAC875) were treated with ammonium chloride, potassium chloride, or ammonium bicarbonate, the latter two treatments served as controls for chloride and ammonium, respectively. Plants were grown under either a watered or water-restricted (drought) regime. The genotype RAC875 was found to accumulate leaf chloride at a significantly higher level than the other genotypes under optimal growth conditions. Under drought conditions, top-dressing RAC875 plants with ammonium chloride resulted in up to a 2.5-fold increase in grain number and this effect was not seen when plants were top-dressed with either of the control fertilizers. The ammonium chloride treatment also minimized losses of grain yield in RAC875 plants grown under drought. Treatment effects were accompanied by an increase in stomatal conductance. These results collectively suggest that the compound fertilizer ammonium chloride can improve drought tolerance of wheat.  相似文献

PCR is a highly accurate technique for confirming the presence of Mycobacterium avium subsp. paratuberculosis (Map) in broth culture. In this study, a simple, efficient, and low-cost method of harvesting DNA from Map cultured in liquid medium was developed. The proposed protocol (Universidad Austral de Chile [UACH]) was evaluated by comparing its performance to that of two traditional techniques (a QIAamp DNA Stool Mini Kit and cethyltrimethylammonium bromide [CTAB] method). The results were statistically assessed by agreement analysis for which differences in the number of cycles to positive (CP) were compared by Student''s t-test for paired samples and regression analysis. Twelve out of 104 fecal pools cultured were positive. The final PCR results for 11 samples analyzed with the QIAamp and UACH methods or ones examined with the QIAamp and CTAB methods were in agreement. Complete (100%) agreement was observed between data from the CTAB and UACH methods. CP values for the UACH and CTAB techniques were not significantly different, while the UACH method yielded significantly lower CP values compared to the QIAamp kit. The proposed extraction method combines reliability and efficiency with simplicity and lower cost.  相似文献

Abstract

AIMS: To determine within-farm prevalence, longitudinal pattern of exposure measured by serology, antibody titre longevity and point prevalence of shedding in urine of Leptospira borgpetersenii serovar Hardjo and L. interrogans serovar Pomona in naturally infected sheep on a sample of commercial farms in New Zealand.

METHODS: On eight commercial sheep farms, between September 2011 and January 2014, blood samples were collected from 115–217 ewe lambs on each farm, at intervals of 2–11 months. They were analysed by microscopic agglutination test (MAT) for antibodies to L. borgpetersenii serovar Hardjo and L. interrogans serovar Pomona, using a titre cut-point of 48. Urine from 98 animals was tested by quantitative PCR (qPCR). The half-life of antibodies was estimated in 185 sheep for serovar Hardjo and 21 for Pomona, and the seroprevalence and mean titre of animals lost to follow-up was compared with those remaining in the study.

RESULTS: Within-flock seroprevalence for serovar Hardjo reached a maximum at 17–22 months of age, ranging from 79 to 100%. Seroprevalence for serovar Pomona rose above 10% on three farms and increased to 21–54% by 4–14 months. Seroconversions occurred mainly from late autumn to early summer at 7–15 months of age. Seroprevalences ranging from 3 to 76% for serovar Hardjo and 0.5 to 15% for serovar Pomona were observed up to 3 months of age, likely due to maternally derived immunity. The half-life of antibody in response to infection was estimated to be 6.7 (95% CI=5.8–7.9) months for serovar Hardjo and 6.3 (95% CI=4.8–9.0) months for Pomona. The prevalence of sheep with urine positive for leptospires on qPCR on each farm ranged from 11 to 88%. All but one of the qPCR-positive animals were seropositive for serovar Hardjo. On two farms where Pomona exposure was observed, animals that were lost to follow-up had a higher geometric mean titre for serovar Pomona than those remaining in the study.

CONCLUSIONS: This study demonstrated seasonal exposure from autumn to early summer in young sheep, a wide range of within-flock serological and shedding prevalence, and gives an estimation of the half-life of MAT titres in sheep. More extensive data are needed to fully understand the epidemiology of leptospirosis in sheep flocks across New Zealand and, along with economic analysis, to justify and design cost-effective and efficient control measures to protect human and animal health.  相似文献
