Purpose

Discharge of untreated domestic and industrial waste in many European rivers resulted in low oxygen concentrations and contamination with trace metals, often concentrated in sediments. Under these anoxic conditions, the formation of insoluble metal sulfides is known to reduce metal availability. Nowadays, implementation of waste water treatment plants results in increasing surface water oxygen concentrations. Under these conditions, sediments can be turned from a trace metal sink into a trace metal source.

Materials and methods

In an ex situ experiment with metal contaminated sediment, we investigated the effect of surface water aeration on sediment metal sulfide (acid volatile sulfides (AVS)) concentrations and sediment metal release to the surface water. These results were compared with long-term field data, where surface water oxygen and metal concentrations, before and after the implementation of a waste water treatment plant, were compared.

Results and discussion

Aeration of surface water in the experimental setup resulted in a decrease of sediment AVS concentrations due to sulfide oxidation. Metals, known to precipitate with these sulfides, became more mobile and increasing dissolved metal (arsenic (As), cadmium (Cd), copper (Cu)) concentrations in the surface water were observed. Contrary to As, Cd, or Cu, manganese (Mn) surface water concentrations decreased in the aerated treatment. Mn ions will precipitate and accumulate in the sediment as Mn oxides under the oxic conditions. Field data, however, demonstrated a decrease of all total metal surface water concentrations with increasing oxygen concentrations following the implementation of the waste water treatment plant.

Conclusions

The gradual decrease in surface water metal concentrations in the river before the treatment started and the removal of metals in the waste water treatment process could not be countered by an increase in metal flux from the sediment as observed in the experiment.  相似文献

The aim of this study was to quantify the effects of compaction on water flow patterns at the soil profile scale. Control and trafficked plots were established in field trials at two sites. The trafficked treatment was created by four passes track‐by‐track with a three‐axle dumper with a maximum wheel load of 5.8 Mg. One year later, dye‐tracing experiments were performed and several soil mechanical, physical and hydraulic properties were measured to help explain the dye patterns. Penetration resistance was measured to 50 cm depth, with saturated hydraulic conductivity (Ks), bulk density, and macroporosity and mesoporosity being measured on undisturbed soil cores sampled from three depths (10, 30 and 50 cm). Significant effects of the traffic treatment on the structural pore space were found at 30 cm depth for large mesopores (0.3–0.06 mm diameter), but not small mesopores (0.06–0.03 mm) or macroporosity (pores > 0.3 mm). At one of the sites, ponding was observed during the dye‐tracing experiments, especially in the trafficked plots, because of the presence of a compacted layer at plough depth characterized by a larger bulk density and smaller structural porosity and Ks values. Ponding did not induce any preferential transport of the dye solution into the subsoil at this site. In contrast, despite the presence of a compacted layer at 25–30 cm depth, a better developed structural porosity in the subsoil was noted at the other site which allowed preferential flow to reach to at least 1 m depth in both treatments.  相似文献

Further NIRS calibrations were developed for the accurate and fast prediction of the total contents of methionine, cystine, lysine, threonine, tryptophan, and other essential amino acids, protein, and moisture in the most important cereals and brans or middlings for animal feed production. More than 1100 samples of global origin collected over five years were analyzed for amino acids following the Official Methods of the United States and European Union. Detailed data and graphics are given to characterize the obtained calibration equations. NIRS was validated with 98 independent samples for wheat and 78 samples for corn and compared to amino acid predictions using linear crude protein regression equations. With a few exceptions, validation showed that 70-98% of the amino acid variance in the samples could be explained using NIRS. Especially for lysine and methionine, the most limiting amino acids for farm animals, NIRS can predict contents in cereals much better than crude protein regressions. Through low cost and high speed of analysis NIRS enables the amino acid analysis of many samples in order to improve the accuracy of feed formulation and obtain better quality and lower production costs.  相似文献

Protein microarrays provide a powerful tool for the study of protein function. However, they are not widely used, in part because of the challenges in producing proteins to spot on the arrays. We generated protein microarrays by printing complementary DNAs onto glass slides and then translating target proteins with mammalian reticulocyte lysate. Epitope tags fused to the proteins allowed them to be immobilized in situ. This obviated the need to purify proteins, avoided protein stability problems during storage, and captured sufficient protein for functional studies. We used the technology to map pairwise interactions among 29 human DNA replication initiation proteins, recapitulate the regulation of Cdt1 binding to select replication proteins, and map its geminin-binding domain.  相似文献

Alcohol dependence is a major public health challenge in need of new treatments. As alcoholism evolves, stress systems in the brain play an increasing role in motivating continued alcohol use and relapse. We investigated the role of the neurokinin 1 receptor (NK1R), a mediator of behavioral stress responses, in alcohol dependence and treatment. In preclinical studies, mice genetically deficient in NK1R showed a marked decrease in voluntary alcohol consumption and had an increased sensitivity to the sedative effects of alcohol. In a randomized controlled experimental study, we treated recently detoxified alcoholic inpatients with an NK1R antagonist (LY686017; n = 25) or placebo (n = 25). LY686017 suppressed spontaneous alcohol cravings, improved overall well-being, blunted cravings induced by a challenge procedure, and attenuated concomitant cortisol responses. Brain functional magnetic resonance imaging responses to affective stimuli likewise suggested beneficial LY686017 effects. Thus, as assessed by these surrogate markers of efficacy, NK1R antagonism warrants further investigation as a treatment in alcoholism.  相似文献

Two unusual pyridones, trichodin A (1) and trichodin B (2), together with the known compound, pyridoxatin (3), were extracted from mycelia and culture broth of the marine fungus, Trichoderma sp. strain MF106 isolated from the Greenland Seas. The structures of the new compounds were characterized as an intramolecular cyclization of a pyridine basic backbone with a phenyl group. The structure and relative configuration of the new compounds were established by spectroscopic means. The new compound 1 and the known compound 3 showed antibiotic activities against the clinically relevant microorganism, Staphylococcus epidermidis, with IC50 values of 24 μM and 4 μM, respectively.  相似文献

Microbial studies of the Mediterranean sponge Tethya aurantium led to the isolation of the fungus Bartalinia robillardoides strain LF550. The strain produced a number of secondary metabolites belonging to the chloroazaphilones. This is the first report on the isolation of chloroazaphilones of a fungal strain belonging to the genus Bartalinia. Besides some known compounds (helicusin A (1) and deacetylsclerotiorin (2)), three new chloroazaphilones (helicusin E (3); isochromophilone X (4) and isochromophilone XI (5)) and one new pentaketide (bartanolide (6)) were isolated. The structure elucidations were based on spectroscopic analyses. All isolated compounds revealed different biological activity spectra against a test panel of four bacteria: three fungi; two tumor cell lines and two enzymes.  相似文献

Our primary objective was to determine the relationships between Fasciola-specific antibody levels in bulk-tank milk and measures of productivity to estimate economic losses that are associated with Fasciola infections. A bulk-tank milk sample was collected in March 2004 from 1105 dairy herds in Flanders and the antibody levels against Fasciola hepatica (ODRf) and Ostertagia ostertagi (ODRo) were determined. The association of ODRf with four production parameters (milk yield, milk-protein %, milk-fat % and inter-calving interval) was assessed by multivariable linear-regression models. Production data were available for 463 out of the 1105 herds sampled. An increase in ODRf from the 25% quantile (0.428) to the 75% quantile (1.064) was associated with a decrease in the annual average milk yield of 0.7kg/(cowday) (P=0.002), with a decrease in the average milk-fat % of 0.06% (P<0.001) and with an increase of the mean inter-calving interval of 4.7 days (P=0.03). No significant relationship was found with the average milk-protein %. When the relationships of ODRf and ODRo with milk yield were tested simultaneously, we saw an additive rather than synergistic effect of concurrent infections.  相似文献
