A study of the aquatic fate of the triethylamine salt of triclopyr (3,5,6‐trichloro‐2‐pyridinyloxyacetic acid) was conducted in three bays of Lake Minnetonka, Minnesota. Triclopyr is under review by the US Environmental Protection Agency as a selective aquatic herbicide. The primary purpose of this study was to determine dissipation rates of the parent active ingredient, triclopyr, and its major metabolites, 3,5,6‐trichloropyridinol (TCP) and 3,5,6‐trichloro‐2‐methoxypyridine (TMP) in selected matrices including water, sediment, plants, finfish and shellfish. Two 6.5‐ha plots dominated by the weedy species Eurasian watermilfoil (Myriophyllum spicatum L) were treated with triclopyr‐triethylammonum at a rate of 2.5 mg AE liter−1 (2.5 ppm) on 21–23 June 1994. A third 6.5‐ha plot was established as an untreated reference. Water and sediment samples were collected from within the plots and at selected locations up to 1600 m outside of the plots through six weeks post‐treatment for chemical residue analysis. In addition, residue samples were collected from the target and non‐target plants and other non‐target matrices, including game and rough fish, clams and crayfish. All test animals were sequestered in cages located in the center of each plot and samples were collected through four weeks post‐treatment. Half‐lives for dissipation of triclopyr and TCP in water ranged from 3.7 to 4.7 days and from 4.2 to 7.9 days, respectively, with trace amounts of TMP found. Peak triclopyr sediment values ranged from 257 to 335 ng gram−1, with a mean half‐life of 5.4 days, while peak TCP sediment levels ranged from 27 to 65 ng gram−1 (mean half−life = 11.0 days). Trace levels of TMP were detected at one treatment site at one sampling event. Triclopyr and TCP accumulated and cleared from animal tissues proportionately to concentrations in the water (triclopyr dissipation half‐lives <11 days, TCP < 14 days). TMP levels were two to three times higher than those of the other compounds, particularly in visceral tissue. In all cases, residues of these compounds were higher in the inedible portions of the animals, and were usually higher in bottom‐feeding fish species. © 2000 Society of Chemical Industry  相似文献

Orchard and vineyard producers conduct preplant site evaluations to help prevent planting permanent tree and vine crops on lands where the crop will not perform to its highest potential or attain its full life expectancy. Physical soil characteristics within specific soil profiles and spatially throughout an orchard influence decisions on land preparation, irrigation system selection, horticultural choices, and nutrient management. Producers depend on soil surveys to help them understand the soil characteristics of the land and may be interested in technology that provides additional information. Electromagnetic induction (EM38) and four-probe soil resistance sensors (VERIS) are being used in combination with global positioning systems to map spatial variability of soils using apparent soil electrical conductivity (ECa). The hypothesis evaluated in this study is whether rapid, in situ, and relatively low-cost methods of measuring ECa (EM38 and VERIS) can effectively identify and map physical soil variability in non-saline soils. The supposition is that in non-saline soils, ECa levels will relate well to soil texture and water-holding capacity and can be used to map physical soil variability. In turn, the information can be used to guide decisions on preplant tillage, irrigation system design, water and nutritional management, and other horticultural considerations. Two sites in the Sacramento Valley were mapped each with EM38 and VERIS methods. Site-specific management zones were identified by each provider on ECa maps for each site, and then soil samples were collected by University of California researchers to verify these zones. Results showed that on non-saline soils, ECa measured with both EM38 and VERIS correlate with physical soil properties such as gravel, sand, silt, and clay content but the relationship between conductivity and these physical soil properties varied from moderately strong to weak. The strength of the correlation may be affected by several factors including how dominant soil texture is on conductivity relative to other soil properties and on methods of equipment operation, data analysis and interpretation. Overall, the commercial providers of ECa surveys in this study delivered reasonable levels of accuracy that were consistent with results reported in previous studies. At one site, an ECa map developed with VERIS provided more detail on physical soil variability to supplement published soil surveys and aided in the planning and development of a walnut orchard. At a second site, almond yield appeared to correlate well with distinctly different soil zones identified with EM38 mapping.  相似文献

Bacterial coldwater disease, caused by Flavobacterium psychrophilum, remains one of the most significant bacterial diseases of salmonids worldwide. A previously developed and reported live‐attenuated immersion vaccine (F. psychrophilum; B.17‐ILM) has been shown to confer significant protection to salmonids. To further characterize this vaccine, a series of experiments were carried out to determine the cross‐protective efficacy of this B.17‐ILM vaccine against 9 F. psychrophilum isolates (representing seven sequence types/three clonal complexes as determined by multilocus sequence typing) in comparison with a wild‐type virulent strain, CSF‐259‐93. To assess protection, 28‐day experimental challenges of rainbow trout (Oncorhynchus mykiss) fry were conducted following immersion vaccinations with the B.17‐ILM vaccine. F. psychrophilum strains used in challenge trials were isolated from several fish species across the globe; however, all were found to be virulent in rainbow trout. The B.17‐ILM vaccine provided significant protection against all strains, with relative percent survival values ranging from 51% to 72%. All vaccinated fish developed an adaptive immune response (as measured by F. psychrophilum‐specific antibodies) that increased out to the time of challenge (8 weeks postimmunization). Previous studies have confirmed that antibody plays an important role in protection against F. psychrophilum challenge; therefore, specific antibodies to the B.17‐ILM vaccine strain appear to contribute to the cross‐protection observed to heterologous strain. The ability of such antibodies to bind to similar antigenic regions for all strains was confirmed by western blot analyses. Results presented here support the practical application of this live‐attenuated vaccine, and suggest that it will be efficacious even in aquaculture operations affected by diverse strains of F. psychrophilum.  相似文献

Wild caught post-pueruli, year one and year two post settlement juvenile western rock lobster, Panulirus cygnus, were held at ambient temperatures (15.6 °C to 23.1 °C; mean 19.0 ± 0.07 °C) or at 23 °C, and fed the same ration of a formulated pellet diet either once per night, or 3 times per night, over 12 months, to determine whether elevated temperatures and multiple feeds per night would stimulate growth through increased metabolism and feed utilisation without significant negative impacts on survival. Survival of post-pueruli (mean 63%) did not differ between ambient and 23 °C. Survival of year 1 and 2 juveniles was higher at ambient temperatures (p < 0.01 ambient: year 1 juveniles, 68%; year 2 juveniles, 88%; 23 °C: 57% and 74%, respectively). Feeding frequency did not affect survival of post-pueruli and year 2 juveniles (mean 63%, 81% respectively), but survival was 9% higher for year 1 juveniles fed three times per night (58% versus 67%; p < 0.01). All lobsters grew faster at 23 °C than at ambient temperatures (p < 0.05), with the growth of post-pueruli almost doubled at 23 °C (weight gain at 23 °C versus ambient: post-pueruli, 18 438 % versus 9 915 %; year 1 juveniles 259% versus 165%; year 2 juveniles 23% versus 21%). Feed frequency did not influence the growth of year 1 and 2 juveniles. However, there was an interaction effect of temperature and feed frequency on post-pueruli where weight and carapace length were significantly higher at ambient temperatures when post-pueruli were fed three times a day, whereas at 23 °C weight and carapace length were significantly greater when fed once per day (p < 0.05). Feed intake (g pellet dry matter lobster− 1 day− 1) of pellet was higher at 23 °C for all lobsters (p < 0.05), but was the same between lobsters fed 3 times per night versus once per night. This study has shown that increasing temperatures to 23 °C significantly improved the growth of P. cygnus post-pueruli without any adverse effects on survival. The faster growth rates exhibited by year 1 and 2 juveniles at 23 °C may potentially offset their lower survival by significantly reducing culture period. There is no benefit of feeding P. cygnus multiple times at night in terms of growth and survival. The implications for P. cygnus culture are that temperatures should be maintained close to 23 °C during the entire growout period, with due care taken to minimise mortalities through adequate provision of food and shelter. Feeding P. cygnus once daily to excess just prior to dusk to co-incide with nocturnal feeding behaviour is recommended.  相似文献

Human patients with atopic dermatitis (AD) commonly exhibit IgE reactivity to cutaneous self-antigens. The presence of serum IgE autoantibodies appears to correlate with disease severity, and it is suspected to reflect or contribute to tissue damage. The objective of this study was to determine whether IgE autoantibodies specific for cutaneous antigens could be detected in the serum of dogs with AD. Serum was collected from 19 dogs with untreated moderate to severe AD and four specific-pathogen free (SPF) dogs. Indirect immunofluorescence was performed using normal canine skin collected at four different locations (concave ear, nose, medial thigh and lateral thorax), while Western immunoblotting was done using normal canine ear pinna epidermal and dermal extracts and reducing conditions. In both methods, IgE was detected using a monoclonal antibody specific for heat stable epitopes of canine IgE. At 1:10 dilution, specific IgE autoantibodies against cutaneous autoantigens were not detected, with either method, in AD and SPF canine sera. Either IgE autoreactivity is not associated with moderate to severe AD in dogs, or the methods employed herein were not sensitive enough to permit IgE autoantibody detection.  相似文献

Phoma stem canker is a damaging disease of oilseed rape (Brassica napus) that causes annual yield losses to UK oilseed rape growers worth approximately £100 million, despite the use of fungicides. In the UK, oilseed rape is sown in August/September and harvested in the following July. The disease epidemics are initiated by ascospores released from Leptosphaeria spp. pseudothecia (ascocarps) on stem stubble in the autumn/winter. Control of this disease is reliant on the use of cultivars with “field resistance” and azole fungicides. This study investigated the effects of cultivar resistance and application of the fungicide prothioconazole on the severity of stem canker before harvest and the subsequent production of pseudothecia on the infected stubble under natural conditions in the 2017/2018, 2018/2019, and 2019/2020 cropping seasons. The application of prothioconazole and cultivar resistance decreased the severity of phoma stem canker before harvest, and the subsequent production of Leptosphaeria spp. pseudothecia on stubble in terms of pseudothecial density. Results showed that stems with less severe stem cankers produced fewer mature pseudothecia of Leptosphaeria spp. on the infected stubble. This investigation suggests that the most sustainable and effective integrated control strategy for phoma stem canker in seasons with low quantities of inoculum is to use cultivars with medium or good field resistance and apply only one spray of prothioconazole when required.  相似文献

A phytotoxic protein that evokes the typical symptoms of Verticillium wilt disease in seedlings of Gossypium hirsutum L. (Upland cotton) was isolated from culture filtrates of Verticillium dahliae. The protein was purified by ammonium sulfate precipitation, Sephadex-G100 fractionation, and native PAGE. The 18.5 kDa protein, designated VD18.5, appears to be a single subunit protein with an isoelectric point between 3 and 5. VD18.5 induces symptoms of leaf dehydration, chlorosis, necrosis and stem discoloration in seedlings of the disease susceptible cotton cultivar Siokra 1–4. The LD50 of VD18.5 on protoplasts of Siokra 1–4 was 18 μg mL−1. VD18.5 had no noticeable effect on Pima S-7, which is a disease resistant cultivar. Phytotoxic activity was partially destroyed at high temperature and was abolished by digestion with proteinase K. Mass spectrometry fingerprinting and protein sequence data from VD18.5 yielded no significant matches when submitted to the Mascot search engine and NCBI non-redundant protein databases, respectively. These results suggest that VD18.5 is a novel protein that may be involved in the development of some of the symptoms associated with Verticillium wilt disease in the cotton plant.  相似文献
