Rainbow trout acclimated to soft water were submitted to an incremental velocity trial, and exhibited a 14% decrease in critical swimming speed (U crit ∼ 1.37 ± 0.055 vs. 1.54 ± 0.044 m s−1) compared to fish kept in hard water. After a standardized swimming protocol, soft-water-acclimated fish had higher blood lactate concentrations (6.5 ± 0.66 and 6.0 ± 0.64 mmol L−1 (soft water) vs. 5.0 ± 0.46 and 3.9 ± 0.32 mmol L−1 (hard water)), revealing a greater use of anaerobic metabolism for the same exercise. Cardiovascular parameters were investigated while fish were swimming at increasing water velocities, revealing that soft-water-acclimated fish had lower increases in heart rate (105% vs. 118% of pre-exercise values), due to higher heart rates observed during acclimation and during the first 10 min of the swimming trial. This was also reflected by the plateau in heart rate and stroke volume observed during the swimming protocol, which can be attributed to increased cardiovascular function in response to soft-water acclimation. These results are in accord with previously reported increases in blood-to-water diffusion distance, due to proliferation of chloride cells at the gills in response to soft-water conditions, and underscore the costs and limitations of soft-water acclimation. A previous proteomic study examining the plasma acute‐phase response of rainbow trout to sterile inflammation highlighted an unidentified 9.5‐kDa spot using 2D‐PAGE, which was dramatically increased. The 15 amino acid sequence obtained from this protein spot allowed rapid amplification of cDNA ends PCR to generate a 443‐bp nucleotide sequence that was 98.6% similar to type‐4 ice‐structuring protein LS‐12 from Atlantic salmon Salmo salar Linnaeus. Quantitative reverse translation PCR and an ELISA were used to measure gene expression and plasma concentrations of LS‐12 following experimental intraperitoneal injection of rainbow trout with either 106 or 108 colony‐forming units (CFU) of Flavobacterium psychrophilum. There was no significant change in the plasma concentration of LS‐12 up to 15 days post‐infection in any group. Hepatic LS‐12 gene expression was significantly reduced at 3 and 6 days (p < 0.001) post‐infection in fish injected with 108 CFU of F. psychrophilum relative to control fish, while branchial or head kidney expression was unchanged. Infected fish had significantly increased hepatic gene expression of serum amyloid A, confirming an acute‐phase response. Under the conditions used, LS‐12 is not a positive acute‐phase protein in rainbow trout.  相似文献

Pre‐surgical investigation of digital flexor tendon sheath pathology remains challenging with current standard imaging techniques. The aim of this prospective, anatomical, pilot study was to describe the anatomy of the equine hind limb digital flexor tendon sheath using a combination of computed tomography (CT) and computed tomographic contrast tenography in clinically normal cadaver limbs. Ten pairs of hind limbs with no external abnormalities were examined from the level of the tarsometatarsal joint distally. Limbs initially underwent non‐contrast CT examination using 120 kVp, 300 mAs, and 1.5 mm slice thickness. Sixty millilitres of ioversol iodinated contrast media and saline (final concentration 100 mg/ml) were injected using a basilar sesamoidean approach. The computed tomographic contrast tenography examination was then repeated, before dissection of the specimens to compare gross and imaging findings. The combined CT and computed tomographic contrast tenography examinations provided excellent anatomical detail of intra‐thecal structures. The borders of the superficial and deep digital flexor tendons, and the manica flexoria were consistently identifiable in all limbs. Detailed anatomy including that of the mesotenons, two of which are previously undescribed, and the plantar annular ligament were also consistently identifiable. Dissection of all 10 pairs of limbs revealed there to be no pathology, in accordance with the imaging findings. In conclusion, the combination of CT and computed tomographic contrast tenography may be useful adjunctive diagnostic techniques to define digital flexor tendon sheath pathology prior to surgical exploration in horses.  相似文献
