Immunization and culture of rainbow trout organ sections in vitro

Splenic and anterior kidney sections or whole organs were excised from large (1 kg) or small (200 g) rainbow trout (Salmo gairdneri) and placed in sterile 60 mm plastic plates containing 10 ml of Eagle's minimal essential medium (EMEM) supplemented with normal or fetal calf serum for in vitro culture. The organ samples were immunized in vitro by direct injection or by mixing in the medium Yersinia ruckeri O-antigen or dinitrophenyl-Ficoll. The medium was changed once during the 10-day incubation at 15 C. The passive hemolytic plaque assay demonstrated antibody production from the plaque-forming cells (PFC); passive hemagglutination was used to measure antibody titers in the media. High numbers of PFC occurred in cultures of either kidney or spleen, demonstrating that these organs can function independently for antibody production. Splenic sections from large fish produced more PFC than comparable whole organs from small fish. EMEM supplemented with 2% normal calf serum was a satisfactory culture medium. 2-hydroxyethyl-mercaptan an ingredient used in mammalian cell culture, inhibited antibody production in trout cells. These techniques are being used in the culture of organs and cells to elucidate pathways and sequences of antigen uptake and delivery of the immunopoietic tissues in trout.     

Concentration of plasma testosterone in adult roosters of a low-weight strain

The levels of blood-plasma testosterone were studied in the adult light bodied strain cocks. Blood was sampled (1 ml) from the wing vein by means of a heparinized syringe six times within a 24-hour interval. The plasma obtained by centrifugation was kept in a refrigerator at a temperature of --18 degrees C. Testosterone was determined by the radioimmunological method. The testosterone levels were significantly higher (P less than 0.001), 18.8 nmol/l on the average, in the blood samples taken during the dark period of the day (10 o'clock p.m., 2 and 6 o'clock a.m.) than in the samples taken in the light part of the day (8 and 10 o'clock a.m., 2 o'clock p.m.) when the average concentration of the studied hormone was 10.7 nmol/l of plasma. the lowest levels of testosterone were recorded in the morning and the highest values were recorded late in the evening (10 o'clock p.m.). The differences in the levels of the hormone between sampling terms indicate that the testosterone level in adult animals shows diurnal variances. It was found by the study of the concentration of testosterone in the blood plasma of the same individuals within short intervals of time (30-60 min) that testosterone secretion in adult cocks had a pulsatory manner. The evaluation of hormone levels should include not only the diurnal variances but also the pulsatory manner of testosterone secretion.     

SELECTED SURGICAL DISEASE STATES WHERE MICROBIOLOGICAL EVALUATION IS ESSENTIAL

Infection caused by bacterial organisms constitutes an important problem in surgical practice. Bacteremia due to both Gram positive and Cram negative bacteria is often complicated by shock. Frequent antibiotic susceptibility tests and bacterial culture of blood, urine, body fluids, draining wounds or other sources of infection is an essential adjunct to the proper management of surgical infections which may lead to septic shock. In combination with current medical and surgical principles of septic shock therapy, two things may help to reverse the high mortality rate, making the diagnosis early enough and supporting the patient until the shock can be reversed.     

Alteration of cellular immune responses by nutrition and weaning in calves

The effects of two levels of nutrition (400 g and 1000 g air dry matter milk substitute powder per day) and three ages of weaning (rive, nine and 13 weeks) on cellular immune responses were determined in 32 calves. The lower level of nutrition was found to increase skin sensitivity responses to keyhole limpet haemocyanin (KLH) and decrease lymphocyte blastogenesis test (LBT) responses to ConA and pokewood mitogen (P<0·05). Weaning at five weeks old resulted in increased KLH skin responses at nine weeks old compared with unweaned calves and decreased LBT responses to ConA and phyto-haemagglutinin at 10 weeks old compared with calves weaned at nine weeks old (P<0·05). Weaning at five weeks old also increased peripheral blood concentrations of BoCD2+ and BoCD8+ lymphocytes (P<0·05). The results show that the choice of husbandry conditions alters cellular immune responses in young calves and suggest that early weaning effects are essentially nutritional.
