ABSTRACT A 3-year experiment was conducted in field microplots infested with Fusarium oxysporum f. sp. ciceris race 5 at Córdoba, Spain, in order to assess efficacy of an integrated management strategy for Fusarium wilt of chickpea that combined the choice of sowing date, use of partially resistant chickpea genotypes, and seed and soil treatments with biocontrol agents Bacillus megaterium RGAF 51, B. subtilis GB03, nonpathogenic F. oxysporum Fo 90105, and Pseudomonas fluorescens RG 26. Advancing the sowing date from early spring to winter significantly delayed disease onset, reduced the final disease intensity (amount of disease in a microplot that combines disease incidence and severity, expressed as a percentage of the maximum possible amount of disease in that microplot), and increased chickpea seed yield. A significant linear relationship was found between disease development over time and weather variables at the experimental site, with epidemics developing earlier and faster as mean temperature increased and accumulated rainfall decreased. Under conditions highly conducive for Fusarium wilt development, the degree of disease control depended primarily on choice of sowing date, and to a lesser extent on level of resistance of chickpea genotypes to F. oxysporum f. sp. ciceris race 5, and the biocontrol treatments. The main effects of sowing date, partially resistant genotypes, and biocontrol agents were a reduction in the rate of epidemic development over time, a reduction of disease intensity, and an increase in chickpea seedling emergence, respectively. Chickpea seed yield was influenced by all three factors in the study. The increase in chickpea seed yield was the most consistent effect of the biocontrol agents. However, that effect was primarily influenced by sowing date, which also determined disease development. Effectiveness of biocontrol treatments in disease management was lowest in January sowings, which were least favorable for Fusarium wilt. Sowing in February, which was moderately favorable for wilt development, resulted in the greatest increase in seed yield by the biocontrol agents. In March sowings, which were most conducive for the disease, the biocontrol agents delayed disease onset and increased seedling emergence. B. subtilis GB03 and P. fluorescens RG 26, applied either alone or each in combination with nonpathogenic F. oxysporum Fo 90105, were the most effective treatments at suppressing Fusarium wilt, or delaying disease onset and increasing seed yield, respectively. The importance of integrating existing control practices, partially effective by themselves, with other control measures to achieve appropriate management of Fusarium wilt and increase of seed yield in chickpea in Mediterranean-type environments is demonstrated by the results of this study.  相似文献

We have recently described the isolation and molecular characteristics of two recombinant avian leukosis subgroup J viruses (ALV J) with an avian leukosis virus subgroup A envelope (r5701A and r6803A). In the present study, we examined the role of the subgroup A envelope in the pathogenesis of these recombinant viruses. Chickens of line 151(5) x 7(1) were inoculated at 1 day of age with r5701A, r6803A, Rous-associated virus type 1 (RAV-1), or strain ADOL-Hcl of ALV-J. At 2, 4, 10, 18, and 32 wk postinoculation (PI), chickens were tested for avian leukosis virus (ALV)-induced viremia, shedding, and neutralizing antibodies. All except one chicken inoculated with the recombinant viruses (98%) developed neutralizing antibodies by 10 wk PI compared with only 16% and 46% of the ADOL-Hcl and RAV-1-inoculated birds, respectively. ALV-induced tumors and mortality in the two groups inoculated with recombinant viruses were different. The incidence of tumors in groups inoculated with r5701A or RAV-1 was 100% compared with only 9% in the groups inoculated with r6803A or ADOL-Hcl. The data suggest that differences in pathogenicity between the two recombinant viruses might be due to differences in the sequence of the 3' untranslated region (presence or absence of the E element), and, therefore, not only the envelope but also other elements of the viral genome play an important role in the pathogenesis of ALV.  相似文献

An acidic peroxidase (pI approximately 2.5) was purified from turnip roots (TAP), and its thermal properties were evaluated. TAP is a monomeric protein having a molecular weight (MW) of 49 kDa and a carbohydrate content accounting for 18% of the MW. The yield of pure TAP was relatively high ( approximately 2 mg/kg of fresh roots), with a specific activity of 1810 2,2'-azinobis(3-ethylbenzthiazoline-6-sulfonic acid) (ABTS) units/mg at pH 6. The activity increased 4-fold at the optimum pH (4.0) to 7250 ABTS units/mg, higher than that of most peroxidases. TAP was heat stable; heat treatment of 25 min at 60 degrees C resulted in 90% initial activity retention, whereas an activity of 20% was retained after 25 min of heating at 80 degrees C. TAP regained 85% of its original activity within 90 min of incubation at 25 degrees C, following heat treatment at 70 degrees C for 25 min. Thermal inactivation caused noticeable changes in the heme environment as evaluated by circular dichroism and visible spectrophotometry. TAP was rapidly denatured by heating in the presence of 1.0 mM ethylene glycol bis(beta-aminoethyl ether) N,N,N',N'-tetraacetic acid, but the Soret band and activity were fully recovered by adding an excess of Ca(2+). This is further evidence that Ca(2+) plays an important role in the stability of TAP. The high specific activity of TAP, together with its relatively high thermal stability, has high potential for applications in which a thermally stable enzyme is required.  相似文献

Since xylanases can differ widely in their bleaching efficiency, the performance of one new and two commercial xylanases was evaluated in an eucalyptus kraft pulp following XD (X: xylanase; D: chlorine dioxide) and XP (P: hydrogen peroxide) sequences. The new xylanase did not show a significant bleach boosting effect but increased the hexenuronic acid (HexA) removal by 10% after the D stage. The two commercial xylanases behaved in a different way, being one of them (XC) the most effective in increasing delignification (9%) and brightness (3%ISO). Its effectiveness was related to its greater action on releasing the xylan polymer, thus producing also a strong decrease in the HexA contents during the enzymatic stage (15%). All xylanases produced morphological changes in the fibre surfaces, but only with XC cracks and holes that improved the diffusion of reactives were observed. Finally, the best bleaching results were obtained with the XD sequence and therefore, a complete bleaching sequence XDEopD1 (Eop: alkaline extraction with oxygen and peroxide) was carried out with the best enzyme.  相似文献

This paper reports an attempt to functionally and chemically characterize commercial ingredients from Vitis vinifera L. grape skins, grape pomace, and leaves, which are used in the formulation of dietary antioxidant supplements. The antioxidant capacity of these ingredients was assessed for the first time by the oxygen radical absorbance capacity (ORAC) methodology. Ingredients from grape skins and pomace (n = 17) showed ORAC values from 1.38 to 21.4 mumol Trolox equivalents/mg whereas ingredients from leaves (n = 4) showed ORAC values from 1.52 to 2.55 mumol Trolox equivalents/mg. The high-performance liquid chromatography-diode array detection/electrospray ionization-mass sprectrometry analysis of anthocyanins and flavonols revealed the authenticity of the ingredients as derived from V. vinifera L. and confirmed large differences in their phenolic content and distribution. A progressive decline in both antioxidant capacity and total anthocyanin content of a grape skin ingredient (43 and 40% decrease, respectively) was observed over a 60 day storage period (45 degrees C and 75% relative humidity), demonstrating its poor stability under these conditions.  相似文献
