Dendritic cells (DCs) and macrophages are critical to innate and adaptive immunity to the intestinal bacterial microbiota. Here, we identify a myeloid-derived mucosal DC in mice, which populates the entire lamina propria of the small intestine. Lamina propria DCs were found to depend on the chemokine receptor CX3CR1 to form transepithelial dendrites, which enable the cells to directly sample luminal antigens. CX3CR1 was also found to control the clearance of entero-invasive pathogens by DCs. Thus, CX3CR1-dependent processes, which control host interactions of specialized DCs with commensal and pathogenic bacteria, may regulate immunological tolerance and inflammation. 相似文献

Both dauer formation (a stage of developmental arrest) and adult life-span in Caenorhabditis elegans are negatively regulated by insulin-like signaling, but little is known about cellular pathways that mediate these processes. Autophagy, through the sequestration and delivery of cargo to the lysosomes, is the major route for degrading long-lived proteins and cytoplasmic organelles in eukaryotic cells. Using nematodes with a loss-of-function mutation in the insulin-like signaling pathway, we show that bec-1, the C. elegans ortholog of the yeast and mammalian autophagy gene APG6/VPS30/beclin1, is essential for normal dauer morphogenesis and life-span extension. Dauer formation is associated with increased autophagy and also requires C. elegans orthologs of the yeast autophagy genes APG1, APG7, APG8, and AUT10. Thus, autophagy is a cellular pathway essential for dauer development and life-span extension in C. elegans. 相似文献

A two-year experiment was conducted at the Beltsville Agricultural Research Center (BARC), MD, and Virginia Polytechnic Institute and State University Kentland Agricultural Research Farm (KARF), VA, to evaluate potato (Solanum tuberosum L.) production of 32- to 57-mm-size-range tubers (referred to hereafter as creamers) in a conservation-tillage, cover-cropping strategy. The experiments used a split-plot design in which the main-plots were cover crop treatments and the sub-plots were different potato selections. Main plot treatments included rye (Secale cereale L.), crimson clover (Trifolium incarnatum L.), Austrian winter pea (Pisum sativum L.), rape (Brassica napus L.), oat (Avena sativa L.), rye/crimson clover mixture, rape/crimson clover mixture, bare soil/raised beds, and bare soil/flat beds (control). Potato selections tested were B1145-2, B1491-5, and B1492-12 in 2000 and B1145-2, B1102-3, and B0811-4 in 2001. Yields in the conservation-tillage treatments were equal to or better than those in the bare soil/flat bed control with few exceptions. Large-sized tubers (<57 mm) in almost all cases remained below 6% of total marketable yield even when the tubers were harvested late. Delayed harvest did not reduce creamer-sized yields nor did it increase yield of large-sized tubers. Economic analysis shows that net returns from some conservation-tillage treatments are equal to or higher than the conventional-tillage strategy and confirms the viability of the conservation-tillage, cover-cropping strategy. Furthermore, the conservation-tillage strategy in many cases allows timely planting using machinery in the wet soils of Maryland and Virginia during the narrow window of spring potato planting time, whereas the conventional tillage strategy does not offer this advantage. 相似文献

Wild sablefish, Anoplopoma fimbria, are a valuable commercial species whose populations are declining. Fortunately, sablefish are excellent species for commercial aquaculture. Sablefish raised under high‐density conditions in the marine environment require the use of efficacious vaccines to control disease. Sablefish impacted by disease in net pens may have poor flesh quality and high mortality during grow‐out. As a result, disease can cause financial hardship for sablefish aquaculture operators. The efficacy of a multivalent vaccine preparation for sablefish, administered either by intraperitoneal (i.p.) injection or by immersion, against atypical and typical Aeromonas salmonicida, the causative agents of atypical and typical furunculosis, respectively, was examined. A. salmonicida can affect sablefish at any age and size. Consequently, an efficacious vaccine that can be appropriately and optimally administered to all life stages is desirable. Sablefish vaccinated by immersion at ~1.5 or ~4.5 g with a whole‐cell multivalent vaccine were not protected against either typical or atypical A. salmonicida. Factors that may have contributed to the ineffectiveness of the immersion vaccine are discussed. By contrast, the relative per cent survival (RPS) or potency of the whole‐cell multivalent vaccine injected i.p. in juvenile sablefish at ~50 g against typical and atypical A. salmonicida was 94.3% and 81.7% respectively. The high RPS values indicated that the vaccine successfully initiated an immune response in sablefish upon a second encounter with the pathogen. 相似文献

A 2-yr-old female forest buffalo (Syncerus caffer nanus) presented with acute trauma to the right horn following a fight with a conspecific resulting in fracture and avulsion of the frontal bone, cornual process, horn, and juxtaposed skin. The animal was sedated for wound care and debridement followed by placement of an antibacterial-impregnated polymethylmethacrylate (PMMA) dressing. The dressing was noted to be intact on examination at day 7 post-injury, but had become dislodged and had to be replaced at day 11 postinjury. The replacement dressing ultimately fell off 68 days post-injury, revealing a healed circular scar. The use of PMMA in this case provided optimal wound care. In the 6 yr since the injury, the animal has grown a nearly normal-appearing horn, suggesting that the noncornual skin and associated skull structures may retain the ability to differentiate into a near-normal appearing horn because the entire horn apparatus in this case was avulsed. 相似文献

A 14-year-old female bottlenose dolphin was diagnosed with mycobacterial panniculitis based on punch biopsy specimens. The necropsy revealed numerous pyogranulomas in the blubber, as well as marked acute multifocal necrosuppurative pneumonia and lymphadenitis. In addition, the animal had marked scoliosis, which had first been noted clinically in the dolphin at about 1 mo of age. The animal had been treated with low-dose dexamethasone for approximately the last 19 mo to reduce perceived discomfort from spondyloarthritis and with the progestational agent altrogenest for approximately 8 yr to prevent pregnancy. Acid-fast positive bacilli were detected in the dermis but not in lung or lymph nodes. Mycobacterium chelonae was isolated from pooled skin, lung, and peripheral lymph-node tissue. Mycobacterial infection may be considered as a differential diagnosis in bottlenose dolphins with generalized cutaneous inflammation, particularly if chronic steroid and progesterone treatments were administered, both of which may have an immunomodulatory effect. 相似文献

Nutritional programming represents the mechanism through which broodstock feeding strategies have lifelong effects on the offspring generation. The objective of this study was to determine whether supplementing rainbow trout, Oncorhynchus mykiss broodstock diets with choline and methionine affects offspring growth performance. Experimental diets were produced by top-dressing a commercial broodstock diet with (a) 12,000 ppm methionine, (b) 7,400 ppm choline, (c) 12,000 ppm methionine and 7,400 ppm choline, or (d) water (control). Six rainbow trout families were fed experimental diets beginning 18 months post-hatch through spawning; the offspring consumed a commercial diet. The broodstock diet did not affect maternal body weight, body condition, egg size, or egg yield (p > .05). Eggs from choline-treated broodstock contained 10% more choline than control eggs (p < .05). Offspring from choline-treated broodstock were smaller than controls at 146 days post-hatch (dph) (p < .05). At the final harvest (439 dph), offspring from the control broodstock weighed 12–18% less than offspring from broodstock consuming the supplemented diets (p < .05). However, there was a significant diet-by-family interaction on offspring growth (p < .05); supplemented diets improved performance in three of the six broodstock families. These findings indicate that supplementing broodstock diets with methionine and choline causes a nutritional programming effect that benefits growth in offspring. 相似文献

Cooled stallion semen has a short viable life, which ranges with acceptable motility and viability from 24 up to 48 hours. The purpose of this study was to compare the effects of storage pH, the ability of three different zwitterionic buffers, and cholesterol-loaded cyclodextrins (CLC) to preserve the motility and integrity of stallion sperm cooled to 5°C for 48 hours. Fourteen ejaculates were collected and split to receive CLC or not (control group). After incubation, each sample was split into six subsamples and diluted in KMT extender containing 4-(2-hydroxyethyl)piperazine-1-ethanesulfonic acid (HEPES), N,N-bis(2-hydroxyethyl)-2-aminoethanesulfonic acid (BES), or 2-(N-morpholino)ethanesulfonic acid (MES) buffers, and the final pH was adjusted to either 7.0 or 6.6, totalizing 12 experimental groups as a function of CLC, buffer, and pH variables (2 × 3 × 2 factorial). The motility parameters and integrity of plasma and acrosome membranes (live cell index) were determined using computer-automated semen analysis and epifluorescence microscopy at 3, 6, 24, and 48 hours of cooling period. According to results, pH was not a significant source of variation for motility and live sperm over different cooling periods. However, samples diluted in BES exhibited higher progressive motility within 3 hours and higher percentages of total motile cells after 48 hours of incubation at 5°C (P < .05). After 24 hours of storage, CLC-treated sperm samples presented higher motility than control group (P < .05), and after 48 hours of incubation, CLC-treated sperm exhibited higher percentages of live, motile, and progressively motile sperms (P < .05). We inferred that equine semen diluted in KMT containing BES as buffer and CLC treatment improve the equine sperm survival during storage at 5°C for 48 hours. 相似文献

An Allee effect occurs when at low densities there is a positive relationship between density and fitness leading to a higher probability of extinction. One circumstance in which it may arise is when isolated individuals are less effective in modifying their environment in a favourable way than groups of individuals. In this paper we use Porcellio scaber to test the hypothesis that the fitness correlate of growth rate will vary as a function of density not just due to the negative effects of increasing interference competition whilst foraging at higher densities, but also positively at lower densities due to isolated individuals spending more time and energy searching for other individuals with which to aggregate whilst sheltering.Below densities of 70 m–2 relative growth rates increased as a function of density, above 70 m–2 growth rate decreased with increasing density. Time spent walking and searching increased at lower densities. These behavioural observations, together with the original observations on water loss and oxygen consumption made by Allee [1], are used to interpret the curvilinear growth rate response to density as an example of a component Allee effect, which is discussed in relation to the importance of aggregation as an adaptation to the terrestrial environment in the Oniscidea. 相似文献
