The coreceptor cytotoxic T lymphocyte-associated antigen 4 (CTLA-4) is pivotal in regulating the threshold of signals during T cell activation, although the underlying mechanism is still not fully understood. Using in vitro migration assays and in vivo two-photon laser scanning microscopy, we showed that CTLA-4 increases T cell motility and overrides the T cell receptor (TCR)-induced stop signal required for stable conjugate formation between T cells and antigen-presenting cells. This event led to reduced contact periods between T cells and antigen-presenting cells that in turn decreased cytokine production and proliferation. These results suggest a fundamentally different model of reverse stop signaling, by which CTLA-4 modulates the threshold for T cell activation and protects against autoimmunity.  相似文献

Understanding the fundamental patterns and determinants of human cooperation and the maintenance of social order in human societies is a challenge across disciplines. The existing empirical evidence for the higher levels of cooperation when altruistic punishment is present versus when it is absent systematically ignores the institutional competition inherent in human societies. Whether punishment would be deliberately adopted and would similarly enhance cooperation when directly competing with nonpunishment institutions is highly controversial in light of recent findings on the detrimental effects of punishment. We show experimentally that a sanctioning institution is the undisputed winner in a competition with a sanction-free institution. Despite initial aversion, the entire population migrates successively to the sanctioning institution and strongly cooperates, whereas the sanction-free society becomes fully depopulated. The findings demonstrate the competitive advantage of sanctioning institutions and exemplify the emergence and manifestation of social order driven by institutional selection.  相似文献

ObjectiveTo test the hypothesis that subarachnoid administration of buprenorphine and lidocaine provides more intense and longer lasting perioperative analgesia with less side effects than xylazine and lidocaine in goats.Study designRandomized, blinded, controlled study.Study animals Ten healthy female goats randomly assigned to two groups of five animals each.MethodsAfter sedation with acepromazine (0.1 mg kg?1) intravenously (IV), lidocaine 2% (0.1 mL kg?1) combined with either xylazine (0.05 mg kg?1; Group X) or buprenorphine (0.005 mg kg?1; Group B) were injected intrathecally at the lumbo-sacral junction prior to stifle surgery. Electrocardiogram, heart rate, direct systolic, mean, and diastolic arterial blood pressures, rectal temperature and arterial blood gases were recorded as were post-operative sedation and pain scores using a visual analogue and numeric rating scale, respectively. Data were analyzed with one-way anova for repeated measures, one-way anova, Friedman's and Kruskal–Wallis tests as necessary (p< 0.05).ResultsSurgery was successfully performed under both analgesia protocols. Total pain and sedation scores were significantly lower in the B as compared with X group from 3–24 hours and 30–120 minutes, respectively after subarachnoid drug administration (SDA). Heart rate and arterial blood pressures decreased post SDA and were consistently lower in X versus B (p< 0.05). In B arterial blood gas parameters did not change post SDA, but in group X PaCO2 increased slightly within 15 minutes of SDA and remained elevated for at least 3 hours (p< 0.05).ConclusionIn these goats intrathecal administration of buprenorphine and lidocaine produced more profound and longer lasting analgesia with less sedation and hemodynamic and respiratory impairment than xylazine with lidocaine.Clinical relevanceIn these goats undergoing hind limb surgery, subarachnoid buprenorphine/lidocaine offered more intense and longer lasting analgesia than a xylazine/lidocaine combination, with less sedation and impairment of cardiopulmonary function.  相似文献

The accumulation of reactive oxygen species (ROS) during biotic stress is either part of a hypersensitive response of the plant or induced directly by the pathogen. Antioxidants such as ascorbate and glutathione counteract the accumulation of ROS and are part of the defense reaction. The aim of the present study was to investigate the compartment-specific importance of ascorbate and glutathione during a virulent and avirulent Pseudomonas syringae infection in Arabidopsis thaliana. Peroxisomes were found to be the hotspot for glutathione accumulation reaching 452% and 258% of control levels 24 h postinoculation during the virulent and avirulent infection, respectively. An accumulation of ascorbate could also be observed in vacuoles during Pseudomonas syringae infection, whereas glutathione remained absent in this cell compartment. Neither glutathione nor ascorbate accumulated in the apoplast during pathogen infection demonstrating an only negligible role of these antioxidants in the apoplast during pathogen infection. Compartment-specific changes followed a recently proposed stress model with an increase of ascorbate and glutathione in most cell compartments at the early stages of infection and a strong drop at the later stage of infection when a strong accumulation of ROS and symptoms occurred in the leaves. This study highlights the importance of certain cell compartments and antioxidants in general for the protection of pathogen-induced ROS accumulation.  相似文献

The reliability of Arctic climate predictions is currently hampered by insufficient knowledge of natural climate variability in the past. A sediment core from Lake El'gygytgyn in northeastern (NE) Russia provides a continuous, high-resolution record from the Arctic, spanning the past 2.8 million years. This core reveals numerous "super interglacials" during the Quaternary; for marine benthic isotope stages (MIS) 11c and 31, maximum summer temperatures and annual precipitation values are ~4° to 5°C and ~300 millimeters higher than those of MIS 1 and 5e. Climate simulations show that these extreme warm conditions are difficult to explain with greenhouse gas and astronomical forcing alone, implying the importance of amplifying feedbacks and far field influences. The timing of Arctic warming relative to West Antarctic Ice Sheet retreats implies strong interhemispheric climate connectivity.  相似文献

Summary A Giemsa-C-banded karyotype of a partial amphiploid, Triticum turgidum L. var. durum cv. Nodak × Agropyron intermedium (Host) P.B., called MT-2, was analyzed. MT-2 is a winterhardy grasslike octoploid germplasm which survived 5 winters in Montana, and its seed weight is 3 times that of A. intermedium seed. The MT-2 C-banding karyotype shows 6 chromosome pairs each of the A and B wheat genomes with 3A and 4B missing. Chromosomes 1B and 2B are involved in a reciprocal homozygous translocation (T1BS·2BS, T1BL·2BL) which was also confirmed by a nucleolus-associated quadrivalent in an MT-2 × durum wheat backcross. In addition to the wheat chromosomes, MT-2 consistently shows 16 A. intermedium chromosome pairs which are designated from A to P. These chromosomes show C-banding patterns similar to those reported earlier in the literature. A large amount of C-banding polymorphism and structural rearrangements in A. intermedium itself presently make a definite chromosome assignment to the homeologous groups of the Triticeae difficult.
