There is a lack of quantitative assessments available on the effect of agricultural intensification on soil aggregate distribution and microbial properties. Here, we investigated how short-term nitrogen(N) intensification induced changes in aggregate size distribution and microbial properties in a soil of a hot moist semi-arid region(Bangalore, India). We hypothesised that N intensification would increase the accumulation of macroaggregates 2 mm and soil microbial biomass and activity, and that the specific crop plant sowed would influence the level of this increase. In November 2016, surface(0–10 cm) and subsurface(10–20 cm) soil samples were taken from three N fertilisation treatments, low N(50 kg N ha~(-1)), medium N(75 and 100 kg N ha~(-1) for finger millet and maize, respectively),and high N(100 and 150 kg N ha~(-1) for finger millet and maize, respectively). Distribution of water-stable aggregate concentrations,carbon(C) and N dynamics within aggregate size class, and soil microbial biomass and activity were evaluated. The high-N treatment significantly increased the concentration of large macroaggregates in the subsurface soil of the maize crop treatment, presumably due to an increased C input from root growth. Different N fertilisation levels did not significantly affect C and N concentrations in different aggregate size classes or the bulk soil. High-N applications significantly increased dehydrogenase activity in both the surface soil and the subsurface soil and urease activity in the surface soil, likely because of increased accumulation of enzymes stabilised by soil colloids in dry soils. Dehydrogenase activity was significantly affected by the type of crop, but urease activity not. Overall, our results showed that high N application rates alter large macroaggregates and enzyme activities in surface and subsurface soils through an increased aboveground and corresponding belowground biomass input in the maize crop. 相似文献

Two carboxylic acid haptens of glycitein were synthesized, with a spacer arm at the C2 position. They differed in the length of the spacer arm, with the length of the spacer arms being three or four carbon atoms, and were named Delta3-glycitein and Delta4-glycitein haptens, respectively. The different haptens were coupled to bovine serum albumin (BSA), and the coupling efficiency was assessed by MALDI mass spectrometry. Polyclonal antibodies were generated against the BSA conjugates. An additional conjugate of Delta4-glycitein hapten was generated with swine thyroglobulin (Thyr). Enzyme-linked immunosorbent assays (ELISAs) based on the competition between free glycitein and Delta4-glycitein-Thyr conjugates for specific antibodies were developed. The IC50 of the standard curves was 15.6 ng mL(-1) with anti-Delta3-glycitein and 62.5 ng mL(-1) with anti-Delta4-glycitein, that is, 10.9 and 44 pmol/well, respectively. With the Delta3-glycitein antibody, interassay and intra-assay variations were 12.2 and 11.5%, respectively. Specificity tests did not show any significant cross-reaction with any other soy isoflavone. This specificity is not influenced by the length of the spacer arm. Neospora caninum is a parasite responsible for paresis in dogs. The dog can harbour enkysted parasites in several organs. The detection of N. caninum was performed using 3 different real time PCR systems all amplifying the NC5 DNA region. One system was based on Sybrgreen, one on Plexor technology and the last on Taqman probe. Comparison of the three methods indicated that the detection limit was 1 equivalent genome on pure DNA but that this detection limit increased in the presence of foreign DNA using the Sybrgreen and Plexor systems. Therefore, the Taqman system was chosen to detect N. caninum in liver and spleen of naturally infected dogs. The overall prevalence was 32.2%. Comparison between PCR results and serological results using IFAT showed that among the 28 PCR positive dogs only 9 were seropositive and that 8 seropositive dogs were PCR negative. Therefore serology can underestimate the real carriage in dogs. However, PCR methods must be improved in terms of sensitivity and inhibition problems. 相似文献

1. Naked oats belong to the same species as 'common oats', Avena sativa, but have a non-lignified husk which readily becomes detached during harvesting. The absence of the indigestible husk can be predicted to give an increased metabolisable energy (ME) content for poultry. 2. Measurements of true metabolisable energy (TME(N)) were performed on 3-week-old broiler chicks (Cobb males), adult cockerels (ISA Brown, greater than one year old) and 7-week-old turkeys (BUT T8 males). The measurements were repeated in 2000, 2001 and 2002, with some measurements on a subset of varieties in 2004. 3. High-oil naked oat lines yielded 12% more energy (TME(N)) than wheat. Naked oats, excluding the experimental high-oil lines, yielded 8.5% more energy than simultaneously assayed wheat samples. 4. In samples from the 2004 harvest, conventional oats gave TME(N) values about 13% lower than those of wheat. 5. The addition of beta-glucanase produced an increase of about 4% in the apparent metabolisable energy (AME) of oats for broiler chickens. This effect was associated with a 70% decrease in the jejunal viscosity of broilers receiving a 500 g/kg naked-oat diet. 6. The oil content of naked oats was about 5 times greater than that of wheat, with the high-oil lines rising to more than 6 times greater. Naked oats had a lower starch content than wheat but not sufficiently lower to negate the energy benefits of the higher oil content. The crude protein (CP) contents of naked oats were similar to those of wheat, with the high-oil varieties tending to be higher in CP also. 相似文献
