The influence of charcoal on biotic processes in soils remains poorly understood. Charcoal is a natural product of wildfires that burned on a historic return interval of ∼100 years in Scots pine (Pinus sylvestris L.) forests of northern Sweden. Fire suppression and changes in forest stand management have resulted in a lack of charcoal production in these ecosystems. It is thought that charcoal may alter N mineralization and nitrification rates, however, previous studies have not been conclusive. Replicated field studies were conducted at three late-succession field sites in northern Sweden and supporting laboratory incubations were conducted using soil humus collected from these sites. We used activated carbon (AC), as a surrogate for natural-occurring fire-produced charcoal. Two rates of AC (0 and 2000 kg ha−1), and glycine (0 and 100 kg N as glycine ha−1) were applied in factorial combination to field microplots in a randomized complete block pattern. Net nitrification, N mineralization, and free phenol concentrations were measured using ionic and non-ionic resin capsules, respectively. These same treatments and also two rates of birch leaf litter (0 and 1000 kg ha−1) were applied in a laboratory incubation and soils from this incubation were extracted with KCl and analyzed for NH4+ and NO3. Nitrification rates increased with AC amendments in laboratory incubations, but this was not supported by field studies. Ammonification rates, as measured by NH4+ accumulation on ionic resins, were increased considerably by glycine applications, but some NH4+ was apparently lost to surface sorption to the AC. Phenolic accumulation on non-ionic resin capsules was significantly reduced by AC amendments. We conclude that charcoal exhibits important characteristics that affect regulating steps in the transformation and cycling of N.  相似文献

Thiamin deficiency was diagnosed in cats and dogs being fed fresh minced meat, which contained sulphur dioxide as a preservative and less than 0.5 mg/kg thiamin. Trophoblast from Day-14 bovine embryos was cultured in medium containing mitogens to determine if the mitotic index could be altered. Trophoblast from each of 15 embryos was cultured in minimum essential medium (Eagles) with 20 % fetal calf serum (control) or in this medium supplemented with pokeweed mitogen (1 %, v/v), phytohemagglutinin (1 %, v/v), concanavalin A (1 %, v/v) or thymidine (2 mg/ml). No mitogenic effect was observed due to any of the treatments. However, mitotic indexes were significantly lower when pokeweed (P < 0.05) or thymidine (P < 0.01) was added to the medium. A highly significant (P < 0.001) variation in mitotic index between embryos was observed.  相似文献

The main objective of this study was to estimate genetic correlations between fertility and production traits in first, second and third lactations as well as between fertility traits measured in the same way at different ages. The fertility traits studied were: number of inseminations per service period, number of treatments for reproductive disturbances, interval between first and last inseminations, interval between calving and first insemination, and interval between calving and last insemination. Early milk production was measured as the average of the energy-corrected milk yield at the second and third monthly testdays in a lactation. The number of records was approximately 450 000, 350 000, 180 000 and 75 000 in the heifer period, first, second, and third lactations, respectively. A linear, trivariate model that included the effects of herd-year, year, month, age and sire of the cow was applied. To reduce the effect of ongoing selection, 305-days kg protein production in first lactation was included as a variate in all of the analyses. Correlations between the herd-year effects indicated that factors of herd-year level conducive to increased production had a tendency to increase the number of inseminations as well as the number of reproductive treatments, although there was an earlier start and termination of the insemination period. Genetic correlations between fertility traits and production were in the range of 0.2-0.4, all of them unfavourable and higher at later parities. The genetic correlations between fertility traits in the heifer period and the same traits in first lactation were 0.7. Genetic correlations between the first and second lactation varied between 0.7 and 0.9, and between the second and third lactation they were all 0.9 or higher. In conclusion, fertility and production traits need to be selected for simultaneously if fertility is going to be maintained along further genetic improvement on production, and such selection should include fertility results from lactating cows.  相似文献

Fragmentation of old-growth forests and greatly reduced amounts of coarse dead wood in managed forests threat the persistence of many saproxylic species in boreal Fennoscandia. Individual old-growth forest remnants may lose species over time as they pay off their extinction debt. We tested this by comparing the observed site occupancy of individual wood-inhabiting fungal species in isolated old-growth stands (i.e. woodland key habitats; WKHs) with statistical predictions of their occupancy assuming potential extinction debt had already been paid off. The occupancy of species was analysed in two sets of WKHs differing in time since isolation (i.e. recent and old isolates).Few species occurred more frequently than expected in WKHs. However, patterns across species and across all WKHs masked important differences among species in their risk of facing future extinction. The site occupancy decreased significantly between recent and old isolates for a group of annual, red-listed specialist fungal species, suggesting that an extinction debt in WKHs may exist among specific species confined to coarse dead wood and old-growth forest habitat. Generalist species that also occur in the surrounding matrix showed no negative trends, or actually increased in site occupancy, making future extinctions less likely. Thus, continuing loss of threatened species are likely if not preservation of WKHs are combined with other conservation efforts in managed forest landscapes. Natural forest landscapes may serve as important references when aiming to identify species in risk of future extinction but more detailed knowledge about the biology of the most vulnerable species is also required.  相似文献

Fruit bodies and mycelia of shiitake mushroom ( Lentinus edodes) have been shown to contain the cholesterol-reducing compound eritadenine, 2( R),3( R)-dihydroxy-4-(9-adenyl)butyric acid. In the search for a production method for eritadenine, shiitake mycelia were investigated in the present study. The mycelia were cultivated both in shake flasks and in bioreactors, to investigate the effects of pH, stirring rate, and reactor type on the production and distribution of eritadenine. Both the biomass and the culture broth were examined for their eritadenine content. In the shake flasks, the final concentration of eritadenine was 1.76 mg/L and eritadenine was equally distributed between the mycelia and the growth media. In the bioreactors, the shiitake mycelia were found to contain eritadenine in relatively low levels, whereas the majority, 90.6-98.9%, was detected in the growth media. Applying a stirring rate of 250 rpm during bioreactor cultivation resulted in the highest eritadenine concentrations: 10.23 mg/L when the pH was uncontrolled and 9.59 mg/L when the pH was controlled at 5.7. Reducing the stirring rate to 50 rpm resulted in a decreased eritadenine concentration, both at pH 5.7 (5.25 mg/L) and when pH was not controlled (5.50 mg/L). The mycelia in the shake flask cultures appeared as macroscopic aggregates, whereas mycelia cultivated in bioreactors grew more as freely dispersed filaments. This study demonstrates for the first time the extra- and intracellular distribution of eritadenine produced by shiitake mycelial culture and the influence of reactor conditions on the mycelial morphology and eritadenine concentrations.  相似文献

This study investigated the prevalence of MRSA in samples taken in households, with and without backyard pigs in villages in a rural area of Shandong Province, China. Community‐associated MRSA and livestock‐associated MRSA, belonging to ST59 and ST9, respectively, were identified in both humans and pigs. The genotypic and phenotypic comparison of isolates indicates that bidirectional transmission of MRSA has occurred between humans and pigs in the villages.  相似文献
