Brucellosis is a zoonosis caused by Brucella species. Brucellosis research in natural hosts is often precluded by practical, economical and ethical reasons and mice are widely used. However, mice are not natural Brucella hosts and the course of murine brucellosis depends on bacterial strain virulence, dose and inoculation route as well as breed, genetic background, age, sex and physiological statu of mice. Therefore, meaningful experiments require a definition of these variables. Brucella spleen replication profiles are highly reproducible and course in four phases: i), onset or spleen colonization (first 48 h); ii), acute phase, from the third day to the time when bacteria reach maximal numbers; iii), chronic steady phase, where bacterial numbers plateaus; and iv), chronic declining phase, during which brucellae are eliminated. This pattern displays clear physiopathological signs and is sensitive to small virulence variations, making possible to assess attenuation when fully virulent bacteria are used as controls. Similarly, immunity studies using mice with known defects are possible. Mutations affecting INF-γ, TLR9, Myd88, Tγδ and TNF-β favor Brucella replication; whereas IL-1β, IL-18, TLR4, TLR5, TLR2, NOD1, NOD2, GM-CSF, IL/17r, Rip2, TRIF, NK or Nramp1 deficiencies have no noticeable effects. Splenomegaly development is also useful: it correlates with IFN-γ and IL-12 levels and with Brucella strain virulence. The genetic background is also important: Brucella-resistant mice (C57BL) yield lower splenic bacterial replication and less splenomegaly than susceptible breeds. When inoculum is increased, a saturating dose above which bacterial numbers per organ do not augment, is reached. Unlike many gram-negative bacteria, lethal doses are large (≥ 108 bacteria/mouse) and normally higher than the saturating dose. Persistence is a useful virulence/attenuation index and is used in vaccine (Residual Virulence) quality control. Vaccine candidates are also often tested in mice by determining splenic Brucella numbers after challenging with appropriate virulent brucellae doses at precise post-vaccination times. Since most live or killed Brucella vaccines provide some protection in mice, controls immunized with reference vaccines (S19 or Rev1) are critical. Finally, mice have been successfully used to evaluate brucellosis therapies. It is concluded that, when used properly, the mouse is a valuable brucellosis model.  相似文献

Fruit and vegetables are normally presented to consumers in batches. The homogeneity and appearance of these have significant effect on consumer decision. For this reason, the presentation of agricultural produce is manipulated at various stages from the field to the final consumer and is generally oriented towards the cleaning of the product and sorting by homogeneous categories. The project ESPRIT 3, reference 9230 ‘Integrated system for handling, inspection and packing of fruit and vegetable (SHIVA)’ developed a robotic system for the automatic, non-destructive inspection and handling of fruit. The aim of this paper is to report on the machine vision techniques developed at the Instituto Valenciano de Investigaciones Agrarias for the on-line estimation of the quality of oranges, peaches and apples, and to evaluate the efficiency of these techniques regarding the following quality attributes: size, colour, stem location and detection of external blemishes. The segmentation procedure used, based on a Bayesian discriminant analysis, allowed fruits to be precisely distinguished from the background. Thus, determination of size was properly solved. The colours of the fruits estimated by the system were well correlated with the colorimetric index values that are currently used as standards. Good results were obtained in the location of the stem and the detection of blemishes. The classification system was tested on-line with apples obtaining a good performance when classifying the fruit in batches, and a repeatability in blemish detection and size estimation of 86 and 93% respectively. The precision and repeatability of the system, was found to be similar to those of manual grading.  相似文献

The purpose of this study was to compare the beta, Johnson SB, Weibull and truncated Weibull functions in describing the diameter distributions of forest stands in Catalonia. The data consisted of permanent sample plots from the Spanish National Forest Inventory in Catalonia. The empirical data represent left-truncated distributions, as the smallest diameter measured in the field was 7.5 cm. A total of 1,242 plots were used to fit the functions and analyze their performance. The distribution functions were fitted to the diameter distributions of the number of stems (DDN) and stand basal area (DDG). The performance of the candidate functions was compared by means of their bias and RMSE for different diameter sums measuring the difference between the empirical and fitted distributions. The leftmost part (from 0 to 7.5 cm) of the non-truncated functions was ignored in this analysis. Sensitivity analyses were conducted to check whether the results depended on the number of trees measured in the stand, or the main species of the stand. The truncated Weibull function for the diameter distribution of stand basal area appeared to be in all cases the most accurate and consistent function. Generally, functions describing the distribution of stand basal area performed better than functions that described the distribution of the number of trees. Of the basal area distributions, beta and Johnson’s SB were the second best and nearly equally good with each other. The order of precision of the tested functions was: truncated Weibull for DDG, truncated Weibull for DDN, Johnson’s SB for DDG, beta for DDG, beta for DDN and Weibull for DDG, Weibull for DDN, and Johnson’s SB for DDN.  相似文献

The objectives of this research were to evaluate gender differences and selection on body weight as they affect growth curves of chickens. Marginal posterior densities of growth curve parameters were studied by Gibbs sampling on 10,671 male and female chickens originating from five lines. Line X-+ was selected on low body weight at 8 wk (BW8) and high body weight at 36 wk (BW36), line X+- on high BW8 and low BW36, X++ on high BW8 and BW36, X-- on low BW8, and BW36, and X00 was an unselected control line. Growth was modeled by a Gompertz function. Heritabilities and genetic correlations among parameters of the Gompertz curve were estimated. Marginal posterior densities were drawn for parameters of the growth curve and for sexual dimorphism at ages ranging from hatching to 1 yr. Lines selected for a higher BW8 had higher initial specific growth rates (L), higher maturation rates (K), and lower ages at inflection (T(I)). Lines selected for a higher BW36 had higher asymptotic body weights (A). Estimates of A, L, and T(I) were higher in males and K was higher in females. Difference between sexes for A was greater in the line selected for a lower BW8 and a higher BW36. Dimorphism for L and K was the lowest in lines X++ and X--, respectively. The greatest difference in T(I) was observed in the line selected for lower BW8 and BW36. Sexual dimorphism of body weight was lower at most ages in the lightest line. Before 15 wk, sexual dimorphism in X++ line was lower than in the line selected for higher BW8 and lower BW36. The increase in sexual dimorphism with body weight could be reduced by selecting animals on body weight at two ages instead of one, as is usually done in commercial lines.  相似文献

Large discrepancies are usually found when different ELISAs for the diagnosis of pig salmonellosis are compared. Thus, our main goal was to estimate the diagnostic accuracy through Bayesian approaches of two commercial assays (Svanovir™ “test A” and HerdCheck™ “test B”) for the detection of antibodies to Salmonella spp. in slaughter pigs. Previously, we estimated the agreement between both tests and their relative sensitivity (Se) and specificity (Sp) with respect to bacteriology on caecal content and ileocaecal lymph nodes. Test A, at a cut-off OD% ≥20%, indicated higher prevalence than test B (OD% ≥10%) (14.6% vs. 8.6%). Relative Se with respect to overall bacteriology was low (≈30%) and similar for both tests, but the relative Sp was significantly lower for test A compared to B (88% vs. 95%). Both tests failed to detect some pigs infected with Salmonella serogroups B and C1, which they were supposed to identify. In general, tests showed only fair-to-moderate agreement when they were compared (kappa: 0.41). In the Bayesian models, Se of test A varied between 63% and 77%, while Se of test B was 73%. Sp of A was always lower than that of test B (89% vs. 95%). The implications derived from the use of these imperfect serological tests will have to be accounted for in large-scale Salmonella-control programs.  相似文献

Model-based accuracy, defined as the theoretical correlation between true and estimated breeding value, can be obtained for each individual as a function of its prediction error variance (PEV) and inbreeding coefficient F, in BLUP, GBLUP and SSGBLUP genetic evaluations. However, for computational convenience, inbreeding is often ignored in two places. First, in the computation of reliability = 1-PEV/(1 + F). Second, in the set-up, using Henderson's rules, of the inverse of the pedigree-based relationship matrix A . Both approximations have an effect in the computation of model-based accuracy and result in wrong values. In this work, first we present a reminder of the theory and extend it to SSGBLUP. Second, we quantify the error of ignoring inbreeding with real data in three scenarios: BLUP evaluation and SSGBLUP in Uruguayan dairy cattle, and BLUP evaluations in a line of rabbit closed for >40 generations with steady increase of inbreeding up to an average of 0.30. We show that ignoring inbreeding in the set-up of the A- inverse is equivalent to assume that non-inbred animals are actually inbred. This results in an increase of apparent PEV that is negligible for dairy cattle but considerable for rabbit. Ignoring inbreeding in reliability = 1-PEV/(1 + F) leads to underestimation of reliability for BLUP evaluations, and this underestimation is very large for rabbit. For SSGBLUP in dairy cattle, it leads to both underestimation and overestimation of reliability, both for genotyped and non-genotyped animals. We strongly recommend to include inbreeding both in the set-up of A- inverse and in the computation of reliability from PEVs.  相似文献

The serological response and protection conferred against Brucella ovis by the Rev 1 vaccine was evaluated in both adult (experiment 1) and young rams (experiment 2) vaccinated either subcutaneously or conjunctivally. In experiment 1 the Rev 1 vaccine protected 55.5 per cent and 100 per cent, respectively, of subcutaneously and conjunctivally vaccinated rams against three consecutive challenges that infected 100 per cent of unvaccinated controls. In experiment 2, Rev 1 protected 100 per cent of rams vaccinated subcutaneously and 70 per cent of those vaccinated conjunctivally against a challenge dose able to infect all the unvaccinated controls. The serological response after vaccination was significantly lower in rams vaccinated conjunctivally than in those vaccinated subcutaneously.  相似文献

The effect of selection for growth rate on carcass and meat quality was assessed by comparing selected and control populations of rabbits measured at the same stage of maturity and slaughtered at 9 and 13 wk of age. Embryos belonging to Generation 7 were frozen, thawed, and implanted in does to produce the control group. The control group was formed from the offspring of the embryos belonging to the Generation 7. Selected animals belonging to Generation 18 (S) were compared with contemporary animals of the control group (C). Carcasses were dissected and measured according to World Rabbit Science Association recommended practices. When animals were compared at similar degrees of maturity, selection for growth rate did not produce a negative effect on carcass and meat quality. There was no increase in fat content of the carcass, and there was an improvement of the meat:bone ratio with selection, with a difference between C and S groups of -0.42. However, the carcasses of S animals have 1.45% lower water-holding capacity. Carcass quality changed markedly with animal age. Heavy rabbit carcasses had lower organ percents and a higher loin percent. Dissectible and i.m. fat content were higher in older rabbits, with older animals having 0.97 and 0.79% more dissectible and i.m. fat content, respectively. Meat quality traits improved with age of slaughter, although there was an increase in glycolytic metabolism. Results from this study indicate that selection for growth rate has little effect in carcass and meat quality when rabbits are measured at the same stage of maturity.  相似文献
