The result of alterations in the levels of CCK, in the blood and in the cerebrospinal fluid, on the functioning of the growth hormone axis has been examined in sheep. Male Coopworth sheep of about 40 kg liveweight were given various doses of CCK either intracerebroventricularly (icv) or intravenously (iv). Other similar sheep were given various doses of a CCK antagonist (loxiglumide) by the same routes. Bolus iv administration of either 35 μg or 200 μg of CCK had no effect on plasma GH levels. When given icv, however, CCK resulted in a marked (P<0.01) prolonged depression in plasma GH levels. The decrease in GH secretion could be partially attenuated by concurrent administration of loxiglumide, but was completely unaffected by concurrent administration of anti-somatostatin serum icv. Loxiglumide alone had no effect on plasma GH levels when given at up to 200 μg icv, but intravenous administration of 8 mg of the CCK antagonist resulted in an increase in plasma GH concentrations (P<0.05). Plasma levels of somatostatin, glucose and cortisol were unaffected by both icv and iv administration of CCK. These results show that CCK can have a strong GH-inhibiting effect in the brain. Furthermore, this effect seems to be independent of hypothalamic somatostatin, suggesting another GH-inhibiting system exists.  相似文献

The vegetative compatibility of four hypovirulent isolates of Cryphonectria parasitica was checked with 35 virulent isolates obtained from chestnut samples collected in many regions of Italy. Artificial inoculation tests confirmed the effectiveness of a mixture prepared with hypovirulent isolates. This was packed in squeezing tubes, to simplify the biological control of chestnut blight in practice.  相似文献

A method for quantitative evaluation of surveillance for disease freedom has been presented in the accompanying paper (Martin et al., 2007). This paper presents an application of the methods, using as an example surveillance for classical swine fever (CSF) in Denmark in 2005. A scenario tree model is presented for the abattoir-based serology component of the Danish CSF surveillance system, in which blood samples are collected in an ad hoc abattoir sampling process, from adult pigs originating in breeding herds in Denmark. The model incorporates effects of targeting (differential risk of seropositivity) associated with age and location (county), and disease clustering within herds. A surveillance time period of one month was used in the analysis. Records for the year 2005 were analysed, representing 25,332 samples from 3528 herds; all were negative for CSF-specific antibodies. Design prevalences of 0.1-1% of herds and 5% of animals within an infected herd were used. The estimated mean surveillance system component (SSC) sensitivities (probability that the SSC would give a positive outcome given the animals processed and that the country is infected at the design prevalences) per month were 0.18, 0.63 and 0.86, for among-herd design prevalences of 0.001, 0.005 and 0.01. The probabilities that the population was free from CSF at each of these design prevalences, after a year of accumulated negative surveillance data, were 0.91, 1.00 and 1.00. Targeting adults and herds from South Jutland was estimated to give approximately 1.9, 1.6 and 1.4 times the surveillance sensitivity of a proportionally representative sampling program for these three among-herd design prevalences.  相似文献

The ovicidal effect of the nematophagous fungus Pochonia chlamydosporia on eggs of Ascaris suum was tested under laboratory conditions. A. suum eggs were plated on 2% water–agar with seven fungal isolates (Isol. 5, Isol. 31, Isol. 1, VC1, Isol. 12, Isol. 22 and VC4) and control without fungus. After 5, 7, 10, 14, 15 and 21 days of incubation, approximately 100 eggs were removed from the plates and classified according to the following parameters: type 1, biochemical and physiological effect without morphological damage to the eggshell, type 2, lytic effect with morphological alteration of the eggshell and embryo and type 3, lytic effect with morphological alteration of eggshell and embryo showing hyphal penetration and internal egg colonization. The isolates effectively destroyed A. suum eggs and all types of effects were observed during the experiment. There was no variation in ovicidal capacity (type 3 effect) among the isolates (p > 0.05) throughout the experiment. After 21 days, isolate 5 showed the highest percentages of type 3 effect (58.33%). The results indicated that P. chlamydosporia (Isol. 5, Isol. 31, Isol. 1, VC1, Isol. 12, Isol. 22 and VC4) can destroy A. suum eggs and is, therefore, a potential biological control agent of nematodes.  相似文献

Objective To examine the effect of dose and route of administration on the sedative‐hypnotic effects of midazolam. Design Prospective randomized controlled study Animals Six indigenous, African bred goats. Methods Pilot studies indicated that the optimum dose of midazolam for producing sedation was 0.6 mg kg?1 for intramuscular (IM) injection, while the optimum intravenous (IV) doses causing hypnosis without, and with loss of palpebral reflexes were 0.6 mg kg?1 and 1.2 mg kg?1, respectively. These doses and routes of administration were compared with a saline placebo in a randomized block design in the main experiment, and the sedative‐hypnotic effects evaluated according to pre‐determined scales. Results Intramuscular midazolam produced sedation with or without sternal recumbency in all animals with the peak effect occurring 20 minutes after administration. The scores for IM sedation with midazolam were significantly different (p < 0.05) from placebo. Intravenous midazolam at 0.6 mg kg?1 resulted in hypnosis, and at 1.2 mg kg?1 increased reflex suppression was observed. The maximum scores for hypnosis at both doses were obtained 5 minutes after IV injection. The mean (± SD) duration of lateral recumbency was 10.8 (± 3.8) minutes after IV midazolam (0.6 mg kg?1) compared to 20 (± 5.2) minutes after midazolam at 1.2 mg kg?1. Compared to baseline, the heart rate increased significantly (p < 0.05) after high dose IV midazolam. Conclusion Intramuscular midazolam (0.6 mg kg?1) produced maximum sedation 20 minutes after injection. Intravenous injection produced maximum hypnosis within 5 minutes. Increasing the IV dose from 0.6 to 1.2 mg kg?1 resulted in increased reflex suppression and duration of hypnosis. Clinical relevance For a profound effect with rapid onset midazolam should be given IV in doses between 0.6 and 1.2 mg kg?1.  相似文献

The sweetpotato whitefly, Bemisia tabaci (Gennadius) (Hemiptera: Aleyrodidae) is a widely distributed and destructive agricultural pest on various host plants. The biology of two biotypes of B. tabaci: the invasive B and a new defined Cv biotype, on a range of host plants (hibiscus, laurel, poinsettia, collard, cucumber and tomato) were studied in the laboratory. Results revealed that the developmental periods of the B biotype immatures were not significantly different on the tested host plants except those between laurel and collard. The Cv biotype immatures developed significantly slower on cucumber and tomato than on the other plants. B. tabaci B biotype had the highest survivorship on collard (68.55%), and the lowest on laurel (33.24%), while the Cv biotype had the highest and lowest survivorships on laurel (61.63%) and tomato (36.74%). Host plants did not significantly affect the pre-ovipostion period regardless of biotype. The longest averaged longevity and highest fecundity of B biotype were both recorded on collard: 25.15 days and 143.0 eggs. The highest fecundity of Cv biotype was 196.49 eggs on laurel and its longest longevity was on hibiscus (19.62 days). The intrinsic rate of natural increase (r m) of B biotype on the three vegetables were all higher than those on the three ornamentals whereas the r m of Cv biotype on the three ornamentals were all higher that those on the three vegetables. Our research indicates that B. tabaci B and Cv biotypes have different host plant suitabilities. The three tested vegetables were more suitable for B biotype while the three tested ornamental plants were more suitable for Cv biotype. The potential mechanism for the different suitability of B and Cv biotypes on various host plants is also discussed.  相似文献

Reach segmentation influences predicted water quality concentrations in water quality modeling. Many lumped or semi-distributed watershed models, including Soil and Water Assessment Tool (SWAT) and Hydrologic Simulation Program-Fortran (HSPF), simulate stream/river water quality with a single segmented reach per subwatershed (SSRS) modeling approach. A multiple-segmented reaches per subwatershed (MSRS) modeling approach was developed, and simulated BOD5 concentrations for this approach were compared with results from the SSRS modeling approach using the HSPF-Paddy model. The SSRS modeling approach has potential systemic errors for predicting BOD5 concentration even when the model is well calibrated. When the point source was loaded at the most upstream location with the same decay rate, the predicted BOD5 concentration using the SSRS modeling approach was higher compared with results for the MSRS modeling approach, and the difference between two methods increases with increasing load and decay rate. When BOD5 was loaded more downstream, BOD5 concentration using the SSRS modeling approach was lower compared with results for MSRS modeling. For a case study, simulated streamflow and BOD5 concentration for the SSRS and MSRS modeling approaches demonstrated good agreement with observed data. However, the estimated decay rate for the SSRS modeling approach was smaller than that for the MSRS modeling approach because BOD decays through total volume in the SSRS modeling approach, although BOD may be loaded anywhere in the reach. The MSRS modeling approach can minimize systematic errors and provide more detailed variation of water quality concentrations along a river length. The MSRS modeling approach does not always need to be applied to all subwatersheds but is recommended for reaches significantly polluted by point source pollution.  相似文献

Somatic mutations in a subset of growth hormone (GH)-secreting pituitary tumors convert the gene for the alpha polypeptide chain (alpha s) of Gs into a putative oncogene, termed gsp. These mutations, which activate alpha s by inhibiting its guanosine triphosphatase (GTPase) activity, are found in codons for either of two amino acids, each of which is completely conserved in all known G protein alpha chains. The likelihood that similar mutations would activate other G proteins prompted a survey of human tumors for mutations that replace either of these two amino acids in other G protein alpha chain genes. The first gene so far tested, which encodes the alpha chain of Gi2, showed mutations that replaced arginine-179 with either cysteine or histidine in 3 of 11 tumors of the adrenal cortex and 3 of 10 endocrine tumors of the ovary. The mutant alpha i2 gene is a putative oncogene, referred to as gip2. In addition, gsp mutations were found in 18 of 42 GH-secreting pituitary tumors and in an autonomously functioning thyroid adenoma. These findings suggest that human tumors may harbor oncogenic mutations in various G protein alpha chain genes.  相似文献
